Comparative analysis of lignin peroxidase and laccase responses to graphene-related nanomaterials.

Graphene-related materials (GRMs) are revolutionizing sectors such as electronics, energy storage, agriculture, and biomedicine due to their exceptional properties. However, concerns are emerging about their environmental impact, particularly regarding their persistence, potential toxicity to aquatic ecosystems, and challenges in safe disposal. These issues highlight the need for more robust sustainable-by-design and risk-assessment strategies. In this context, this research investigated the influence of GRMs on lignin peroxidase (LiP) and laccase (Lac), key enzymes involved in lignin breakdow

Systemic induction of peroxidase and phenolic responses to UV radiation in Nicotiana tabacum.

Systemic changes in isoperoxidase profile and phenolic composition mirror those in leaves exposed to ultraviolet radiation, underscoring the essential role of systemic responses in plant health and development. Plants respond to direct exposure to broadband ultraviolet (UV) light (280-400 nm) through localized protective and regulatory mechanisms. In our recent study using Nicotiana tabacum as a model plant, we showed that protective responses triggered by moderate, non-harmful UV doses could spread from directly treated leaves to UV-unexposed leaves situated above (systemic leaves). This stud

Mechanistic study of the novel peroxidase mimetic DhHP-6 in metabolic cataracts.

Metabolic disorders such as diabetes and hypergalactosemia often induce oxidative damage and mitochondrial dysfunction in the lens, leading to metabolic cataracts. Although surgical intervention remains the primary treatment, its associated risks and complications underscore the need for pharmacological alternatives with multifaceted protective effects. Deuterohemin-βAla-His-Thr-Val-Glu-Lys (DhHP-6), a novel enzyme mimic of microperoxidase-11, has demonstrated potent reactive oxygen species (ROS)-scavenging activity in vivo. Our previous studies have confirmed its efficacy in mitigating lens o

Zirconium-doped iron oxide nanoparticles for enhanced peroxidase-like activity.

Fe3O4 nanoparticles (NPs) have emerged as pioneering nanozymes with applications in clinical diagnosis, environmental protection and biosensing. However, it is currently limited by insufficient catalytic activity due to poor electron transfer. In this study, we synthesized electron-rich-Zr-doped defect-rich Fe3O4 NPs (Zr3Fe3O4) using a one-pot solvothermal method. Compared with intrinsic Fe3O4 NPs, the resultant Zr3Fe3O4 NPs exhibit enhanced peroxidase (POD)-like activity attributed to the presence of active centers of Zr-O-Fe bridges and adsorption sites of asymmetric oxygen vacancies (OVs) Z

[Role of thyroid peroxidase autoantibody in the comorbidities of pemphigus vulgaris and Hashimoto thyroiditis].

Pemphigus vulgaris (PV) is a group of autoimmune bullous diseases characterized by life-threatening intradermal blisters. Hashimoto thyroiditis (HT) is a kind of autoimmune disease with abnormal increase of thyroid peroxidase autoantibody (TPOAb), which is the thyroid specific antibody, leading to hypothyroidism. In recent years, the probability of HT in patients with PV is increasing, and the co-disease may be related to the effect of TPOAb autoantibody on oral keratinocytes. This article reviews the epidemiological relationship between PV and HT and the mechanism of TPOAb in their co-disease

APX2 Is an Ascorbate Peroxidase-Related Protein that Regulates the Levels of Plastocyanin in Chlamydomonas.

The function of ascorbate peroxidase-related (APX-R) proteins, present in all green photosynthetic eukaryotes, remains unclear. This study focuses on APX-R from Chlamydomonas reinhardtii, namely, ascorbate peroxidase 2 (APX2). We showed that apx2 mutants exhibited a faster oxidation of the photosystem I primary electron donor, P700, upon sudden light increase and a slower re-reduction rate compared to the wild type, pointing to a limitation of plastocyanin. Spectroscopic, proteomic and immunoblot analyses confirmed that the phenotype was a result of lower levels of plastocyanin in the apx2 mut

A dye-decolorizing peroxidase from Vibrio cholerae can demetallate heme.

Iron is an essential element for bacterial survival. Bacterial pathogens have therefore developed methods to obtain iron. Vibrio cholerae, the intestinal pathogen that causes cholera, utilizes heme as an iron source. DyP from V. cholerae (VcDyP) is a dye-decolorizing peroxidase. When VcDyP was expressed in Escherichia coli and purified, it was found to contain protoporphyrin IX (PPIX) but not heme, indicating that the protein possesses deferrochelatase activity. Here, we examined the demetallation reaction of VcDyP using fluorescence spectroscopy. Treatment of heme-reconstituted VcDyP with sod

Impact of malaria on glutathione peroxidase levels: a systematic review and meta-analysis.

The primary antioxidant, glutathione peroxidase (GPx), is hypothesized to contribute to the pathophysiology of malaria. This current study conducted a meta-analysis to examine variations in GPx blood levels in malaria patients. Seven electronic databases-ProQuest, Scopus, Embase, MEDLINE, PubMed, Ovid, and Google Scholar-were searched for relevant studies with no limitations to publication language or publication date. The Joanna Briggs Institute critical appraisal tools were used to appraise the risk of bias among the included studies critically. The meta-analysis was conducted by pooling the

Peroxidase-like Nanozymes for Point-of-Care SERS Sensing and Wound Healing.

The emergence of nanozymes provides a potential method for combating multidrug-resistant bacteria resulted from the abuse of antibiotics. However, in nanozyme-catalyzed systems, few studies have addressed the actual hydrogen peroxide (H2O2) level involved in sterilization. Herein, we designed a high-efficiency peroxidase-mimicking nanozyme with surface-enhanced Raman scattering (SERS) property by assembling gold nanoparticles on single-layer Cu2+-C3N4 (AuNP-Cu2+-C3N4). The nanozyme effectively converts the low-active Raman reporter 3,3',5,5'-tetramethylbenzidine (TMB) into its oxidized form wi

Ag-MXene as peroxidase-mimicking nanozyme for enhanced bacteriocide and cholesterol sensing.

Reactive oxygen species (ROS) are ideal alternative antibacterial reagents for rapid and effective sterilization. Although a variety of ROS-based antimicrobial strategies have been developed, many are still limited by their inefficiency. Herein, we report the synthesis of the Ag-MXene nanozyme, which have superior peroxidase-like activity for antibacterial applications. As a result, Ag-MXene nanozyme can efficiently increase the level of intracellular ROS, converting H2O2 into hydroxyl radicals that effectively kill both Gram-negative and Gram-positive bacteria and disrupting the bacterial bio

CuCeTA nanoflowers as an efficient peroxidase candidate for direct colorimetric detection of glyphosate.

Conventional nanozyme-based pesticide detection often requires the assistance of acetylcholinesterase. In this work, a CuCeTA nanozyme was successfully designed for the direct colorimetric detection of glyphosate. Direct detection can effectively avoid the problems caused by cascading with natural enzymes such as acetylcholinesterase. By assembling tannic acid, copper sulfate pentahydrate and cerium(III) nitrate hexahydrate, CuCeTA nanoflowers were prepared. The obtained CuCeTA possessed excellent peroxidase-like activity that could catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB

Glutathione peroxidase 4-regulated neutrophil ferroptosis induces systemic autoimmunity.

The linkage between neutrophil death and the development of autoimmunity has not been thoroughly explored. Here, we show that neutrophils from either lupus-prone mice or patients with systemic lupus erythematosus (SLE) undergo ferroptosis. Mechanistically, autoantibodies and interferon-α present in the serum induce neutrophil ferroptosis through enhanced binding of the transcriptional repressor CREMα to the glutathione peroxidase 4 (Gpx4, the key ferroptosis regulator) promoter, which leads to suppressed expression of Gpx4 and subsequent elevation of lipid-reactive oxygen species. Moreover, th

"Green" Prussian Blue Analogues as Peroxidase Mimetics for Amperometric Sensing and Biosensing.

Prussian blue analogs (PBAs) are well-known artificial enzymes with peroxidase (PO)-like activity. PBAs have a high potential for applications in scientific investigations, industry, ecology and medicine. Being stable and both catalytically and electrochemically active, PBAs are promising in the construction of biosensors and biofuel cells. The "green" synthesis of PO-like PBAs using oxido-reductase flavocytochrome b2 is described in this study. When immobilized on graphite electrodes (GEs), the obtained green-synthesized PBAs or hexacyanoferrates (gHCFs) of transition and noble metals produce
