Green Microbiology for Planetary Resilience: A Research Agenda for Microbial Sustainability Science.

The planet is facing a crisis occasioned by climate change, biodiversity loss, physical, chemical, and biological soil degradation, water insecurity and pollution, infectious disease crises, and unsustainable production systems and vulnerable food systems. These problems are deeply interconnected and share a common solution: microbial science. Microbes are central to maintaining or restoring water safety and quality, the carbon and nitrogen cycles responsible for maintaining and improving soil quality and fertility, plant productivity and immunity from pests and diseases, animal and human heal

Identification of Haustorially Expressed Candidate Effector Proteins from Gymnosporangium yamadae.

Biotrophic rust fungi secrete effector proteins through haustoria to promote successful colonization. Consequently, the functional analysis of haustorial effector proteins has become central to understanding rust pathogenicity. In previous work, we generated the haustorial transcriptome of Gymnosporangium yamadae, the causal agent of apple rust disease, and predicted numerous candidate effector genes. Here, we systematically characterized the functions of 13 haustorially expressed candidate effectors of G. yamadae using a heterologous system to assess their modulation of plant immunity and sub…

A salivary protein NlSP2 engages the OsCBSX3-SNAC3 module to enhance rice immunity.

The molecular dialogue between rice and the brown planthopper (BPH; Nilaparvata lugens) is mediated by insect salivary effectors, yet the molecular mechanisms underlying plant perception and immune activation remain largely unclear. Here, we characterized the BPH salivary protein NlSP2, which is secreted into rice tissues during feeding. Silencing of NlSP2 in BPH impairs salivary sheath formation, feeding ability, and survival, whereas its ectopic expression in rice enhances resistance to BPH via MAPK activation, H2O2 accumulation, and induction of defense-related genes. We identify the rice c

Cucumber Mosaic Virus Promotes Sphingolipid Biosynthesis to Alleviate Host Unfolded Protein Response (UPR)-Mediated Immunity.

RNA viruses usually reprogramme host cellular lipid metabolism to create favourable replication microenvironments, yet the broader biological significance of these lipid changes is largely unknown. To explore the potential roles of virus-induced lipids, we employed an economically important plant pathogen cucumber mosaic virus (CMV), which has a single-stranded, positive-sense, tripartite RNA genome replicating in host tonoplasts. Lipidomic analysis of purified tonoplasts revealed that CMV infection increased the relative accumulation of sphingolipids in tonoplasts. This is consistent with the

Rhamnolipids elicit defense responses and induce disease resistance against biotrophic, hemibiotrophic, and necrotrophic pathogens that require different signaling pathways in Arabidopsis and highlight a central role for salicylic acid.

Plant resistance to phytopathogenic microorganisms mainly relies on the activation of an innate immune response usually launched after recognition by the plant cells of microbe-associated molecular patterns. The plant hormones, salicylic acid (SA), jasmonic acid, and ethylene have emerged as key players in the signaling networks involved in plant immunity. Rhamnolipids (RLs) are glycolipids produced by bacteria and are involved in surface motility and biofilm development. Here we report that RLs trigger an immune response in Arabidopsis (Arabidopsis thaliana) characterized by signaling molecul…

Regulatory mechanisms of synthetic microbial communities on rhizosphere microecology to alleviate continuous monoculture obstacles of medicinal plants.

Driven by industrial policies and market expansion, China has formed the world's largest medicinal plant cultivation industry. However, sustainable herbal production is seriously hindered by continuous monoculture obstacles (CMOs), which arise from prolonged single-species successive cultivation and manifest as comprehensive soil degradation featuring autotoxic allelochemical buildup, unbalanced rhizosphere microbiota, impaired soil physicochemical conditions, and frequent soil-borne pathogen outbreaks. Such adverse alterations lower soil enzyme activity, disrupt microbial community homeostasi

Autophagy meets plant virology: From molecular crosstalk to outstanding challenges.

Autophagy is an evolutionarily conserved degradation and recycling pathway that plays essential roles in plant development, stress adaptation, and immunity. Over the past decade, increasing evidence has revealed that autophagy also shapes the outcome of plant-virus interactions. This review summarizes current understanding of the plant autophagy machinery as well as the experimental approaches used to monitor autophagic flux during viral infection. We then explore the diverse roles of autophagy in the plant-virus interface, highlighting its function as an antiviral mechanism that targets viral

Induced resistance in cucumber by oleamide from Streptomyces lydicus JCK-6019 against cucumber Fusarium wilt and damping-off.

Oleamide (cis-9-octadecenamide), a primary fatty acid amide derived from oleic acid, is well known in mammalian systems for its roles in neural and immune regulation. However, its function in plant immunity has remained unexplored. In this study, oleamide was identified as the major bioactive metabolite of Streptomyces lydicus JCK-6019 and was characterized as a novel plant resistance inducer (PRI) in cucumber. Although oleamide exhibited no direct antifungal activity against Fusarium oxysporum f. sp. cucumerinum and Rhizoctonia solani AG-4 in vitro, in vivo assays demonstrated strong disease …

Disruption of sphingolipid biosynthesis in Nicotiana benthamiana activates salicylic acid-dependent responses and compromises resistance to Alternaria alternata f. sp. lycopersici.

Sphingolipids play an important role in signal transduction pathways that regulate physiological functions and stress responses in eukaryote … ulators of programmed cell death. Therefore, to gain more insight into the role of sphingolipids in plant immunity against SAM-producing necrotrophic fungi, we disrupted sphingolipid metabolism in Nicotiana benthamiana through virus-induced gene silencing (VIGS) of the serine palmitoyltransfersase (SPT). This enzyme catalyzes the first reaction in LCB synthesis. …

Molecular Basis of Citrus sunki Susceptibility and Poncirus trifoliata Resistance Upon Phytophthora parasitica Attack.

Coevolution has shaped the molecular basis of an extensive number of defense mechanisms in plant-pathogen interactions. Phytophthora parasitica, a hemibiothrophic oomycete pathogen and the causal agent of citrus root rot and gummosis, interacts differently with Citrus sunki and Poncirus trifoliata, two commonly favored citrus rootstocks that are recognized as susceptible and resistant, respectively, to P. parasitica. The molecular core of these interactions remains elusive. Here, we provide evidence on the defense strategies employed by both susceptible and resistant citrus rootstocks, in para

The potato StMKK5-StSIPK module enhances resistance to Phytophthora pathogens through activating the salicylic acid and ethylene signalling pathways.

Mitogen-activated protein kinase (MAPK) cascades play pivotal roles in plant responses to both biotic and abiotic stress. A screen of a Nicotiana benthamiana cDNA virus-induced gene silencing (VIGS) library for altered plant responses to inoculation with Phytophthora infestans previously identified an NbMKK gene, encoding a clade D MAPKK that we renamed as NbMKK5, which is involved in immunity to P. infestans. To study the role of the potato orthologous gene, referred to as StMKK5, in the response to P. infestans, we transiently overexpressed StMKK5 in N. benthamiana and observed that cell dea

Autoimmunity in plants.

Attenuation in the activity of the negative regulators or the hyperactivity of plant innate immune receptors often causes ectopic defense activation manifested in severe growth retardation and spontaneous lesion formations, referred to as autoimmunity. In this review, we have described the cellular and molecular basis of the development of autoimmune responses for their useful applications in plant defense. Plants are exposed to diverse disease-causing pathogens, which bring infections by taking over the control on host immune machineries. To counter the challenges of evolving pathogenic races

Comparative metabolomic analysis highlights the involvement of sugars and glycerol in melatonin-mediated innate immunity against bacterial pathogen in Arabidopsis.

Melatonin is an important secondary messenger in plant innate immunity against the bacterial pathogen Pseudomonas syringe pv. tomato (Pst) DC3000 in the salicylic acid (SA)- and nitric oxide (NO)-dependent pathway. However, the metabolic homeostasis in melatonin-mediated innate immunity is unknown. In this study, comparative metabolomic analysis found that the endogenous levels of both soluble sugars (fructose, glucose, melibose, sucrose, maltose, galatose, tagatofuranose and turanose) and glycerol were commonly increased after both melatonin treatment and Pst DC3000 infection in Arabidopsis.
