SabR enhances nikkomycin production via regulating the transcriptional level of sanG, a pathway-specific regulatory gene in Streptomyces ansochromogenes.

BACKGROUND: sabR is a pleiotropic regulatory gene which has been shown to positively regulate the nikkomycin biosynthesis and negatively affect the sporulation of Streptomyces ansochromogenes. In this study, we investigate the mechanism of SabR on modulating nikkomycin production in Streptomyces ansochromogenes. RESULTS: The transcription start point of sabR was determined by high-resolution S1 nuclease mapping and localized at the nucleotide T at position 37 bp upstream of the potential sabR translation start codon (GTG). Disruption of sabR enhanced its own transcription, but retarded the ni

Indicative oligodendrocyte dysfunction in spinal cords of human fetuses suffering from a lethal motoneuron disease.

Human spinal cord development is still poorly understood and detailed molecular analyses of human motoneuron diseases could improve our understanding of the normal developmental processes of the spinal cord. Lethal Congenital Contracture Syndrome (LCCS, MIM 253310) provides a human model to study the early motoneuronal development. A typical phenotype of LCCS fetuses consists of multiple joint contractures, distinct facial features, and hydrops. Tissue pathology is characterized by severe muscle atrophy, lung hypoplasia, and degeneration of the anterior horn of the spinal cord as the hallmark

Different consequences of EGR2 mutants on the transactivation of human Cx32 promoter.

The early growth response 2 (EGR2) transcription factor plays a crucial role in peripheral nerve myelination. Mutations of this gene are associated with a wide variety of demyelinating neuropathies differing from each other in the severity of nerve injury. Although the expression of EGR2 mutants inhibits the transactivation of myelin gene promoters, the exact molecular mechanism by which these mutations cause the alteration of the myelination process is still unknown. Recently, it was reported that EGR2 is directly involved in the transcriptional regulation of Connexin 32, a myelin gene freque

SRE1 and SRE2 are two specific steroid-responsive modules of Candida drug resistance gene 1 (CDR1) promoter.

CDR1 gene encoding an ATP-driven drug extrusion pump has been implicated in the development of azole-resistance in Candida albicans. Although the upregulation of CDR1 expression by various environmental factors has been documented, the molecular mechanism underlying such process is poorly understood. We have demonstrated earlier that the CDR1 promoter encompasses a large number of cis-regulatory elements, presumably mediating its response to various drugs. In this study we have identified a novel steroid responsive region (SRR) conferring beta-oestradiol and progesterone inducibility on the CD

Herkinorin Exerted Neuroprotective Effects After Spinal Cord Injury by Suppression of NF-κB Pathway Activation.

The disruption of axonal continuity and neuronal loss at the injury site leads to structural disconnections as well as functional consequences in spinal cord injury. Herkinorin, a structurally unique non-nitrogenous opioid receptor agonist, demonstrates significant neuroprotective efficacy in models of ischemic brain injury and epilepsy. In this report, we aimed to explore the role and underlying mechanism of Herkinorin in axon regeneration and functional recovery in SCI. Oxygen-glucose deprivation and restoration (OGD/R) condition was used in cell line rat pheochromocytoma cells (PC12) and pr

Synergistic Effect of Indole-3-Acetic Acid and Homogeneously Dispersed Ulva prolifera-Derived Carbon Quantum Dots in Enhancing CO2 Fixation, Biomass Production, and Macromolecules in Chlorella vulgaris.

To address light attenuation caused by self-shading in dense microalgal cultures within photobioreactors, this study proposes a synergistic strategy employing Ulva prolifera-derived carbon quantum dots (U-CQDs) combined with indole-3-acetic acid (IAA). Hydrothermally synthesized U‑CQDs exhibiting fluorescence emission at 580 nm were homogeneously suspended in the culture, converting UV‑blue light into orange‑red wavelengths readily absorbed by microalgae, thereby improving light distribution uniformity. The combination of 1 mg·L-1 U-CQDs and 5 mg·L-1 IAA increased biomass production to 2.17 g·

Greener Decellularization of Porcine Auricular Cartilage Using Supercritical Technology and Different Pretreatments for Application in Tissue Engineering.

Scaffolds for tissue engineering can be obtained from synthetic or natural materials, with decellularized tissues being particularly attractive. Among these, porcine auricular cartilage is of special interest because of its availability, similarity to the human extracellular matrix (ECM), and cost-effectiveness. Decellularization of animal tissues yields extracellular matrices (ECM) rich in collagen, elastin, and glycosaminoglycans (GAGs), which are essential for providing mechanical support and creating a favorable environment for cell adhesion and tissue development. Traditional decellulariz

Green-to-red spectral labeling: A novel polysynaptic retrograde tracing strategy in the marker footprint mouse model.

BACKGROUND: Rabies virus (RABV)-derived neuronal tracing tools are extensively applied in retrograde tracing due to their strict retrograde transsynaptic transfer property and low neurotoxicity. However, the RABV infection and expression of fluorescence products would be gradually cleared while the infected neurons still survive, a phenomenon known as non-cytolytic immune clearance (NCLIC). This phenomenon introduced the risk of fluorescence loss and led to the omission of a subset of neurons that should be labeled, thereby interfering in the analysis of tracing results. METHODS: To compensat

NeoI represents a group of transcriptional repressors regulating the biosynthesis of multiple aminoglycosides.

In general, the initiation or closure of antibiotic biosynthesis is determined by regulatory proteins, but most of their mechanisms of action remain unknown. The 2-deoxystreptamine-containing aminoglycosides (2-DOS AGs) form a unique category among antibiotics. Genomic analysis revealed that a group of hypothetical regulatory genes represented by neoI are widely distributed in the biosynthetic gene clusters (BGCs) of natural products from Streptomyces species, including several 2-DOS AGs. Only limited knowledge is available for the roles of NeoI-type regulators although neomycin and some of th

A novel transcriptional complex on the VE-cadherin promoter regulated the downregulation of VE-cadherin in the Down Syndrome Candidate Region 1 isoform 1L-mediated angiogenesis.

Angiogenesis is critical for many diseases. Previously, we reported that Down Syndrome Candidate Region 1 isoform 1L (DSCR1-1L) was one of the most up-regulated genes in endothelial cells induced by VEGF and histamine, and regulated endothelial cell proliferation and Matrigel angiogenesis in mice. However, it was not known whether DSCR1-1L regulated angiogenesis in vivo and what was the molecular mechanism underlying it. In this study, gene knockdown and overexpression models were established to study the role of DSCR1-1L in angiogenesis in vivo. Further, the downstream regulatory target of DS

Dietary phytonutrients and animal health: regulation of immune function during gastrointestinal infections.

The composition of dietary macronutrients (proteins, carbohydrates, and fibers) and micronutrients (vitamins, phytochemicals) can markedly influence the development of immune responses to enteric infection. This has important implications for livestock production, where a significant challenge exists to ensure healthy and productive animals in an era of increasing drug resistance and concerns about the sector's environmental footprint. Nutritional intervention may ultimately be a sustainable method to prevent disease and improve efficiency of livestock enterprises, and it is now well establish

Sex differences in Alzheimer's disease and common neuropathologies of aging.

Alzheimer's dementia is significantly more common in women than in men. However, few pathological studies have addressed sex difference in Alzheimer's disease (AD) and other brain pathologies. We leveraged postmortem data from 1453 persons who participated in one of two longitudinal community-based studies of older adults, the Religious Orders Study and the Rush Memory and Aging Project. Postmortem examination identified AD pathologies, neocortical Lewy bodies, DNA-binding protein 43 (TDP-43), hippocampal sclerosis, gross and micro infarcts, atherosclerosis, arteriolosclerosis, and cerebral am

A novel DNA element mediates transcription of Nkx2.1 by Sp1 and Sp3 in pulmonary epithelial cells.

NKX2.1 is a member of the NK2 family of homeodomain-containing transcription factors whose targeted disruption in mouse results in the absence of thyroid tissue and a severely abnormal lung phenotype. Little is known regarding the mechanisms that control tissue and temporal specificity of Nkx2.1 gene expression. The Nkx2.1 gene has been cloned from a number of species and it is composed of three exons and two introns. Two distinct DNA domains located 5' of exon I and within intron I have been found to exhibit promoter activity in lung and thyroid cells. In the current study we used deletional

Poliumoside Alleviates Neuroinflammation and Oxidative Stress After Spinal Cord Injury by Activating the PI3K/AKT/mTOR Signaling Pathway.

BACKGROUND: Spinal cord injury (SCI) constitutes a profoundly debilitating neurological disorder precipitating motor and sensory function impairment. Curtailing microglia-driven neuroinflammation alongside oxidative stress proves indispensable for efficacious SCI patient management. Poliumoside (POL), a phenylethanoid glycoside molecule, manifests anti-inflammatory, antioxidant, and neuroprotective capacities. Nevertheless, documentation concerning its SCI therapeutic efficacy remains sparse. METHODS: Systemic drug toxicity for two POL dosages (15 mg/kg, 30 mg/kg) was evaluated across multipl

Transcriptomic analysis by RNA sequencing reveals that hepatic interferon-induced genes may be associated with feed efficiency in beef heifers.

In beef cattle, production feedstuffs are the largest variable input cost. Beef cattle also have a large carbon footprint, raising concern about their environmental impact. Unfortunately, only a small proportion of dietary energy is directed toward protein deposition and muscle growth whereas the majority supports body maintenance. Improving feed efficiency would, therefore, have important consequences on productivity, profitability, and sustainability of the beef industry. Various measures of feed efficiency have been proposed to improve feed utilization, and currently, residual feed intake (

Fidaxomicin is an inhibitor of the initiation of bacterial RNA synthesis.

Fidaxomicin was recently approved for the treatment of Clostridium difficile infection. It inhibits transcription by bacterial RNA polymerase. Because transcription is a multistep process, experiments were conducted in which fidaxomicin was added at different stages of transcriptional initiation to identify the blocked step. DNA footprinting experiments were also conducted to further elucidate the stage inhibited. Fidaxomicin blocks initiation only if added before the formation of the "open promoter complex," in which the template DNA strands have separated but RNA synthesis has not yet begun.

The FxRxHrS motif: a conserved region essential for DNA binding of the VirR response regulator from Clostridium perfringens.

The VirSR two-component signal transduction pathway regulates virulence and toxin production in Clostridium perfringens, the causative agent of gas gangrene. The response regulator, VirR, binds to repeat sequences located upstream of the promoter and is directly responsible for the transcriptional activation of pfoA, the structural gene for the cholesterol-dependent cytolysin, perfringolysin O. Comparative sequence analysis of the 236 amino acid residue VirR protein revealed a two-domain structure: a typical N-terminal response regulator domain and an uncharacterised C-terminal domain. Thiolutin attenuates neuroinflammation and neuronal apoptosis by suppressing microglial pyroptosis through NLRP3 inflammasome inhibition after spinal cord injury.

Traumatic spinal cord injury (SCI) initiates a neuroinflammatory response dominated by tissue-resident microglia. Thiolutin (THL) can weaken several inflammatory diseases by blocking the NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome activation. At present, whether THL improves functional recovery after SCI is unclear. Crosstalk between neurons and microglia was simulated by a co-culture system in which microglia were activated by LPS + ATP and treated with THL. Contused SCI mouse models were constructed for studying the efficacy of THL and related mechanisms. Motor fun

Transaldolase 1 impacts Parkinson's disease pathogenesis via metabolic reprogramming and autophagy-lysosomal pathway.

Parkinson's disease (PD) progression involves dopaminergic neurodegeneration and pathological α-synuclein aggregation, processes linked to metabolic dysregulation and autophagy-lysosomal pathway (ALP) impairment. Transaldolase1 (TAL1) is a key enzyme of the pentose phosphate pathway. While elevated TAL1 protein levels have been observed in postmortem substantia nigra of PD patients, the enzyme's functional role in disease pathogenesis remains undefined. In this study, we explored the role of TAL1 in PD-related pathologies using MPTP-induced and AAV-A53T mouse models. We demonstrate that TAL1 u

Can we associate environmental footprints with production and consumption using Monte Carlo simulation? Case study with pork meat.

BACKGROUND: Growing population demands more animal protein products. Pork remains one of the traditional and relatively sustainable types of meats for human consumption. In this paper, life-cycle assessment was performed using data from 12 pig farms. In parallel, a survey on the consumption of pork meat products was conducted analyzing responses from 806 pork meat consumers. The study aims to provide a quantitative calculation of six environmental footprints associated with the consumption of pork meat products in Serbia by analyzing data from pig farms and a pork meat consumption survey.
