The fate and stability of messenger RNA (mRNA), from transcription to degradation is regulated by a dynamic shuttle of epigenetic modifications and RNA binding proteins in maintaining healthy cellular homeostasis and disease development. While Transforming Growth Factor Beta 1 (TGFβ1) has been implicated as a key regulator for diabetic retinopathy, a microvascular complication of diabetes, the RNA binding proteins post-transcriptionally regulating its expression remain unreported in the ocular context. Further, dysfunction of TGFβ1 signalling is also strongly associated with angiogenesis, infl

BACKGROUND: Hepatitis C is an inflammatory condition of the liver caused by the hepatitis C virus, exhibiting acute and chronic manifestations with severity ranging from mild to severe and lifelong illnesses leading to liver cirrhosis and cancer. According to the World Health Organization's global estimates, a population of about 58 million have chronic hepatitis C virus infection, with around 1.5 million new infections occurring every year.
OBJECTIVE: The present study aimed to identify novel molecules targeting the Hepatitis C viral RNA Dependent RNA polymerases, which play a crucial role i

R-loops, RNA-DNA hybrid structures, are integral to key cellular processes such as transcriptional regulation, DNA replication, and repair. However, aberrant accumulation of R-loops can compromise genomic integrity, leading to the development of various diseases. Emerging evidence underscores the pivotal role of RNA methylation modifications, particularly N6-methyladenosine (m6A) and 5-methylcytosine (m5C), in orchestrating the formation, resolution, and stabilization of R-loops. These modifications dynamically regulate R-loop metabolism, exerting bidirectional control by either facilitating o

Normal c-myc RNAs are very unstable with a half-life of less than 30 min whereas those rearranged in 5', as found in Burkitt's lymphomas and mouse plasmacytomas, are significantly more stable. To learn about the sequence determinants controlling their turnover, we have studied naturally occurring and artificially constructed c-myc RNAs rearranged in 5' or 3'. The first conclusion is that sequences necessary for rapid c-myc RNAs turnover are localized in their 3' untranslated region. The second conclusion is that stabilization of truncated c-myc RNAs in tumors does not result from deletion of t

RNA-binding proteins (RBPs), as posttranscriptional regulators, can modulate the activity and stability of target RNAs and participate in the whole life cycle of RNA processing, localization, modification and translation. RNA-binding motif single-stranded interacting proteins (RBMSs) comprise a critical subgroup within the RNA-binding protein (RBP) family, sharing the same domain characteristics as other RBPs. Several studies have shown that RBMSs can participate in tumorigenesis and tumor progression through mechanisms such as regulating the expression of oncogenes, growth factors and cell cy

We describe the effect of guanidinylation of the aminoglycoside moiety on acridine-neamine-containing ligands for the stem-loop structure located at the exon 10-5'-intron junction of Tau pre-mRNA, an important regulatory element of tau gene alternative splicing. On the basis of dynamic combinatorial chemistry experiments, ligands that combine guanidinoneamine and two different acridines were synthesized and their RNA-binding properties were compared with those of their amino precursors. Fluorescence titration experiments and UV-monitored melting curves revealed that guanidinylation has a posit

RNA therapeutics hold transformative potential for precision oncology; however, their systemic application remains constrained by a fundamental tradeoff between prolonged circulation stability and efficient intracellular activation. Conventional polyethylene glycol (PEG)-based carriers provide antifouling protection but lack tumor responsiveness, resulting in limited tumor accumulation and suboptimal gene expression. In this study, we developed a nanosystem, the dual-state tumor-activated RNA platform (D-STAR), which enables a dynamic transition from a stealth circulatory state to an activated

Autosomal dominant dyskeratosis congenita (DKC), as well as aplastic anemia, has been linked to mutations in the RNA component of telomerase, the ribonucleoprotein responsible for telomere maintenance. Here we examine the effect of the DKC mutations on the structure and stability of human telomerase RNA pseudoknot and CR7 domains by using NMR and thermal melting. The CR7 domain point mutation decreases stability and alters a conserved secondary structure thought to be involved in human telomerase RNA accumulation in vivo. We find that pseudoknot constructs containing the conserved elements of

Tasso T20 (T20) is a finger stick-free, self-sampling device capable of at-home blood sample collection. We optimized HIV-1 RNA elution from T20, evaluated its analytical performance, compared its performance to Whatman dried blood spots (DBS), and assessed HIV-1 RNA stability. We used mock viral samples to optimize HIV-1 RNA elution using viral concentrations 10,000 and 3000 copies/mL. Limit of detection (LOD) and limit of quantification (LOQ) were determined, and linearity and reproducibility were verified. Analytical agreement between T20 and DBS was assessed using eight concentrations, fro…

The Alba superfamily proteins share a common RNA-binding domain. These proteins participate in a variety of regulatory pathways by controlling developmental gene expression. They also interact with ribosomal subunits, translation factors, and other RNA-binding proteins. The Leishmania infantum genome encodes two Alba-domain proteins, LiAlba1 and LiAlba3. In this work, we used homology modeling, protein-protein docking, and molecular dynamics (MD) simulations to explore the details of the Alba1-Alba3-RNA complex from Leishmania infantum at the molecular level. In addition, we compared the struc

Small non-coding RNA controls the expression of target genes and is related with mRNA degradation, chromatin modification and genome stability. Recent studies showed that small non-coding RNA is not only associated with the incidence of schistosomiasis, but also acts as a potential biomarker. In this article, we will review the applications of small non-coding RNA in schistosomiasis diagnosis and its potential role in schistosome development and pathogenesis, in the aim to provide hints for developing detection methods and vaccines for schistosomiasis and for drug development.

RNA methylation was detected using MeRIP assay. Protein expression was detected using western blot. Cell proliferation was detected using EdU assay. Macrophage functions was detected using flow cytometry. METTL17 was upregulated after exposure to LPS. However, METTL17 knockdown promoted inflammatory response. Moreover, METTL17 knockdown promoted M1 macrophage polarization. Mechanically, METTL17 regulate RNA

A major challenge in neurobiology in the 21st century is to understand how the brain adapts with experience. Activity-dependent gene express … hyladenosine (m6A) has recently been shown to regulate RNA processing through alternative splicing, RNA stability, and translation. Here, we discuss the emerging idea that m6A could also coordinate the transport, localization, and local translation of key mRNAs in learning and memory and expand on the notion of dynamic functional RNA states in the brain.

Dysregulation of RNA m6A modification has been implicated in Alzheimer's disease (AD), but the molecular mechanisms remain largely unclear. Here, we identified the presence of m6A on mitochondria-encoded messenger RNAs (mt-mRNAs) in the brain, with elevated levels correlated with amyloid-β (Aβ) deposition. Under physiological conditions, cytosolic m6A-modified mt-Nd4 is recognized and degraded by the m6A reader protein YTHDF2, thereby preventing aberrant activation of the RIG-I-MAVS innate immune pathway in neurons. Under Aβ-associated pathological conditions, YTHDF2 expression is markedly dow
