Effect of exposure to ionizing radiation on competitive proliferation and differentiation of hESC.

PURPOSE: We studied the effects of computed tomography (CT) scan irradiation on proliferation and differentiation of human embryonic stem cells (hESCs). It was reported that hESC is extremely radiosensitive; exposure of hESC in cultures to 1 Gy of ionizing radiation (IR) results in massive apoptosis of the damaged cells and, thus, they are eliminated from the cultures. However, after recovery the surviving cells proliferate and differentiate normally. We hypothesized that IR-exposed hESC may still have growth rate disadvantage when they proliferate or differentiate in the presence of non-irrad

DNA damage and repair in plants under ultraviolet and ionizing radiations.

Being sessile, plants are continuously exposed to DNA-damaging agents present in the environment such as ultraviolet (UV) and ionizing radiations (IR). Sunlight acts as an energy source for photosynthetic plants; hence, avoidance of UV radiations (namely, UV-A, 315-400 nm; UV-B, 280-315 nm; and UV-C, <280 nm) is unpreventable. DNA in particular strongly absorbs UV-B; therefore, it is the most important target for UV-B induced damage. On the other hand, IR causes water radiolysis, which generates highly reactive hydroxyl radicals (OH(•)) and causes radiogenic damage to important cellular compon

p21CIP1 is not required for the early G2 checkpoint response to ionizing radiation.

We have previously reported that the immediate G2 checkpoint delay of normal human fibroblasts in response to ionizing radiation is correlated with inhibition of p34CDC2/cyclin B kinase activity. Here, we observed increased amounts of the cyclin-dependent protein kinase inhibitor p21CIP1 associated with p34CDC2/cyclin B protein complexes from irradiated normal human fibroblasts. Since wild-type p53 function is not required for the early G2 checkpoint response to ionizing radiation, we investigated whether a p53-independent induction of p21CIP1 was required for the G2 checkpoint. Early passage

Analysis of deletions induced in the genome of mammalian cells by ionizing radiation.

A theory is presented for the distribution in size of deletions induced by ionizing radiation, based on three assumptions: (1) deletions that are observed delete part or all of a gene to make a mutation, but not adjacent DNA sequences essential for survival of the mutant; (2) deletions are distributed at random along the DNA; (3) the probability of formation is proportional to the rate at which the two endpoints, which must meet to form the deletion, collide with each other. Experimental data for radiation-induced deletions in human and hamster hprt genes are in good agreement with calculation

Ferroptosis-immune crosstalk in ionizing radiation-induced testicular injury: mechanisms and therapeutic opportunities.

Accumulating evidence suggests that ferroptosis-an iron-dependent, lipid peroxidation-driven form of regulated cell death-may contribute substantially to ionizing radiation (IR)-induced testicular injury. In this review, we develop a hypothesis-generating framework for ferroptosis-immune crosstalk in the testis. Within this framework, ferroptotic injury to Sertoli cells is proposed to compromise the blood-testis barrier (BTB), potentially facilitating autoantigen exposure and adaptive immune activation; reciprocal inflammatory signaling may, in turn, increase the susceptibility of neighboring

MicroRNA-Mediated Regulation of Ionizing Radiation Responses: Mechanisms and Advances in Clinical Translation.

Ionizing radiation can induce DNA damage through direct or indirect mechanisms and activate the DNA damage response network, thereby influencing cellular repair, cell cycle regulation and cell fate determination. MicroRNAs (miRNAs), as key post-transcriptional regulatory factors, play a vital role in modulating the radiation response and cell fate. This review summarizes radiation-induced changes in miRNA expression and the molecular regulatory mechanisms they mediate, and further discusses recent advances in their application in the assessment of radiation damage, the prediction of radiothera

Adipose tissue: an overlooked target of medical ionizing radiation and its metabolic consequences.

Despite extensive research on radiation effects on various tissues, the impact on adipose tissue remains largely unexplored. This represents a critical knowledge gap given adipose tissue's role as an active endocrine organ and the extensive use of ionizing radiation in medical procedures. This review synthesizes the current evidence regarding the effects of ionizing radiation on adipose tissue pathophysiology, examining molecular mechanisms and clinical implications for metabolic health. We comprehensively reviewed studies investigating radiation effects on adipocytes and adipose-derived stem

Micronuclei and sister chromatid exchange in lymphocytes from physicians exposed to ionizing radiation.

OBJECTIVE: Occupational exposure to ionizing radiation is a known risk factor for genomic instability. This study evaluated whether physicians occupationally exposed to ionizing radiation exhibit higher frequencies of chromosomal alterations compared to unexposed hospital workers, utilizing micronuclei and sister chromatid exchanges as biomarkers. METHODS: Peripheral blood samples were analyzed from 50 participants: 25 physicians exposed to ionizing radiation (radiologists) and a control group of 25 sex- and age-matched hospital staff with no history of occupational exposure. The cytokinesis-

Time-resolved lipidomic atlas of the murine lung response to acute ionizing radiation.

The lung is among the most radiosensitive organs and a principal dose-limiting organ in thoracic radiotherapy, yet the acute metabolic consequences of irradiation at the lipidomic level remain poorly characterized. We constructed a time-resolved lipidomic atlas of the murine lung response to high-dose irradiation. Male C57BL/6 J mice received a single 10 Gy whole-body X-ray dose, and lungs were collected at 5 minutes, 1 hour, 4 hours, and 24 hours post-exposure (n = 5 per group) and analyzed by untargeted liquid chromatography-tandem mass spectrometry. Radiation triggered rapid and coordinated

Occupational exposure to ionizing radiation in a nuclear medicine department: myth and reality.

Nuclear medicine is a diagnostic and therapeutic medical specialty based on the administration of radiopharmaceuticals emitting ionizing radiation (IR). In this context, healthcare professionals are exposed to low doses of radiation during their activity. This review aims to provide an up-to-date overview of occupational exposures in nuclear medicine in France, based on data from the 2023 IRSN and 2024 ASN reports, and to put them into perspective with those of other medical or industrial sectors, as well as with natural background exposure. Occupational exposures in nuclear medicine remain ve

Mitochondrial Dysfunction Predisposes Radioresistant Insect Cells to Ionizing Radiation by Impairing DNA Repair.

Radioresistant insect cell lines, such as Spodoptera frugiperda Sf9 cells, are well-established eukaryotic model systems for investigating cellular responses to ionizing radiation (IR). Mitochondria are central to these responses due to their roles in reactive oxygen species (ROS) production and in regulating apoptotic signaling pathways. In this study, we compared the radiation responses of parental Sf9 cells with those of a mitochondrial dysfunction variant, Sf9N, to elucidate the contribution of mitochondria to the intrinsic radioresistance of lepidopteran insect cells. Both cell types were

Effect of Thrombopoietin Receptor Agonist Romiplostim on the Ionizing Radiation-Induced Premature Aging.

Environmental stressors, such as ionizing radiation, accelerate aging by causing DNA damage and triggering pathways that lead to cell cycle arrest, apoptosis, and subsequent inflammation. The thrombopoietin receptor agonist romiplostim (RP), which is used as a clinical treatment for chronic idiopathic thrombocytopenic purpura and aplastic anemia, is known to be promising in reducing radiation-induced tissue damage. In this study, we established a mouse model of radiation-induced premature aging to evaluate the potential of RP in ameliorating this process. Female C57BL/6JJcl mice were subjected

Sexual dimorphism during integrative endocrine and immune responses to ionizing radiation in mice.

Exposure to cosmic ionizing radiation is an innate risk of the spaceflight environment that can cause DNA damage and altered cellular function. In astronauts, longitudinal monitoring of physiological systems and interactions between these systems are important to consider for mitigation strategies. In addition, assessments of sex-specific biological responses in the unique environment of spaceflight are vital to support future exploration missions that include both females and males. Here we assessed sex-specific, multi-system immune and endocrine responses to simulated cosmic radiation. For t

Low-Dose Occupational Exposure to Ionizing Radiation and Cardiovascular Effects: A Narrative Review.

Historically, non-cancer diseases have not been considered a health risk following low-dose exposure to ionizing radiation. However, it is now well known that high-dose ionizing radiation causes cardiovascular disease, and emerging epidemiological evidence suggests an excess risk of non-cancer diseases even following exposure to lower doses of ionizing radiation than previously thought. In fact, the evidence is strongest for cardiovascular disease (CVD). The aim of this review was to report the most representative studies and data on the risk of CVD from low-dose radiation in people with occup

Impact of Ionizing Radiation Exposure on Placental Function and Implications for Fetal Programming.

Accidental exposure to high-dose radiation while pregnant has shown significant negative effects on the developing fetus. One fetal organ which has been studied is the placenta. The placenta performs all essential functions for fetal development, including nutrition, respiration, waste excretion, endocrine communication, and immunological functions. Improper placental development can lead to complications during pregnancy, as well as the occurrence of intrauterine growth-restricted (IUGR) offspring. IUGR is one of the leading indicators of fetal programming, classified as an improper uterine e
