Draft genome assembly of hyper citric acid-producing Aspergillus niger mutant strain developed through gamma radiation bombardment.

OBJECTIVES: Aspergillus niger is a renowned filamentous fungus with extensive industrial and biotechnological applications. A. niger is widely used to produce diverse organic acids, enzymes, and other value-added metabolites. Although strain-specific metabolic capabilities have been widely applied in industry, the genomic foundations of these capabilities have not yet been fully characterized. Here, we have described the isolation and provided the draft genome sequence of A. niger strain AN-L103_M1 to further study the genetic determinants of primary and secondary metabolism, given its metabol

Comparison of cytogenetic tests for monitoring overexposures from ionizing radiation.

The frequencies of chromosome aberrations induced by ionizing radiations were studied for use in assessment of doses in radiation accidents. The dose-effect relationships of formations of dicentric and acentric aberrations are compared with that of micronuclei detected in cultured human lymphocytes. The data suggest the applicability of micronucleus test as "biological dosimeter" with a similar sensitivity like the widely used dicentric analysis.

Complementation analysis of ataxia-telangiectasia.

In a number of laboratories genetic analysis of ataxia-telangiectasia (AT) has been performed by studying the expression of the AT phenotype in fused somatic cells or mixtures of cell-free extracts from different patients. Complementation of the defective response to ionizing radiation was observed frequently, considering four different parameters for radiosensitivity in AT. The combined results from studies on cultured fibroblasts or lymphoblastoid cells from 17 unrelated families revealed the presence of at least four and possibly nine complementation groups. These findings suggest that ther

Role of DNA damage in cardiovascular disease.

Patients with some progeroid syndromes, such as Werner syndrome, exhibit atherosclerotic cardiovascular disease (CVD) at a young age as a manifestation of premature aging. Recent studies have revealed that most progeroid syndromes are caused by genetic defects in specific molecules involved in the DNA damage response, a cornerstone of genome stability. Ionizing radiation is one of the most potent genotoxic stimuli and causes various kinds of DNA damage. Further, there is increasing evidence that therapeutic radiation treatments can cause cardiovascular complications. Here, we describe the DNA

Monitoring spontaneous and induced human mutation by RAPD-PCR: a response to Weinberg et al. (2001).

Weinberg et al. (2001, Proc. R. Soc. Lond. B 268, 1001-1005) have recently reported a major increase in mutation rate in the children of Chernobyl liquidators as a result of their fathers' exposure to ionizing radiation. If correct, this would provide dramatic evidence for radiation-induced mutation in humans, and would raise major concerns over the genetic effects of radiation. However, mutants were mainly detected using random amplified polymorphic DNA-PCR, an unreliable technology. These mutants were not validated and had no obvious molecular basis. They may, instead, have arisen as PCR art

Investigation of the Relationship Between Radiation Dose and Gene Mutations and Fusions in Post-Chernobyl Thyroid Cancer.

BACKGROUND: Exposure to ionizing radiation during childhood is a well-established risk factor for thyroid cancer. However, the genetic mechanisms of radiation-associated carcinogenesis remain not fully understood. METHODS: In this study, we used targeted next-generation sequencing and RNA-Seq to study 65 papillary thyroid cancers (PTCs) from patients in the Ukrainian-American cohort with measurement-based iodine-131 (I-131) thyroid doses received as a result of the Chernobyl accident. We fitted linear regression models to evaluate differences in distribution of risk factors for PTC according

Sensitivity, specificity, and persistence of chromosome translocations for radiation biodosimetry.

The analysis of chromosome translocations by fluorescence in situ hybridization ("whole chromosome painting") is the gold standard endpoint for radiation biological dosimetry. Translocation formation requires mis-repair of two or more DNA double-strand breaks, a critical class of damage that is efficiently induced by ionizing radiation in a dose-responsive manner. Translocations are widely used for radiation exposure assessment because they provide a sensitive detection system for low doses, show good specificity for radiation exposure, persist for decades, and are highly relevant to tumorigen

Ataxia-telangiectasia: an interdisciplinary approach to pathogenesis.

Ataxia-telangiectasia is a syndrome with many facets, involving a progressive cerebellar ataxia, immunodeficiency, cancer susceptibility, radiosensitivity, defects in DNA repair/processing, chromosomal breakage and rearrangements, elevated serum alphafetoprotein, and premature aging. Ataxia-telangiectasia is an autosomal recessive disorder, rare in outbred populations; carriers of the ataxia-telangiectasia gene may be as common as 1 in 60 and have subclinical radiosensitivity and cancer susceptibility. One estimate suggests that 8.8% of patients with breast cancer could be carriers of ataxia-t

Therapeutic Strategies for Gliomas Associated With Cancer Predisposition Syndromes.

PURPOSE: The purpose of this article was to provide an overview of syndromic gliomas. DESIGN: The authors conducted a nonsystematic literature review. RESULTS: Cancer predisposition syndromes (CPSs) are genetic conditions that increase one's risk for certain types of cancer compared with the general population. Syndromes that can predispose one to developing gliomas include neurofibromatosis, Li-Fraumeni syndrome, Lynch syndrome, and tuberous sclerosis complex. The standard treatment for sporadic glioma may involve resection, radiation therapy, and/or alkylating chemotherapy. However, DNA-da

Connexin 47 mutations increase risk for secondary lymphedema following breast cancer treatment.

PURPOSE: Secondary lymphedema is a frequent complication of breast cancer associated with surgery, chemotherapy, or radiation following breast cancer treatment. The potential contribution of genetic susceptibility to risk of developing secondary lymphedema following surgical trauma, radiation, and other tissue insults has not been studied. EXPERIMENTAL DESIGN: To determine whether women with breast cancer and secondary lymphedema had mutations in candidate lymphedema genes, we undertook a case-control study of 188 women diagnosed with breast cancer recruited from the University of Pittsburgh

European perspectives on albinism: Epidemiology, clinical features, and management.

Albinism is a genetically heterogeneous group of disorders characterized by reduced or absent melanin synthesis, leading to variable hypopigmentation of the skin, hair, and eyes, and associated ocular abnormalities such as foveal hypoplasia, nystagmus, and reduced visual acuity. Although considered rare, the prevalence of albinism varies widely across regions and is likely underestimated in Europe, particularly because mild phenotypes in fair-skinned populations may be overlooked. In people with albinism in Europe (PWA), reduced cutaneous pigmentation increases susceptibility to ultraviolet ra

Gene Mutations and Targeted Therapies of Myeloid Sarcoma.

Myeloid sarcoma, a rare malignant tumor characterized by the invasion of extramedullary tissue by immature myeloid cells, commonly occurs concomitantly with acute myeloid leukemia, myelodysplastic syndromes, or myeloproliferative neoplasms. The rarity of myeloid sarcoma poses challenges for diagnosis and treatment. Currently, treatments for myeloid sarcoma remain controversial and primarily follow protocols for acute myeloid leukemia, such as chemotherapy utilizing multi-agent regimens, in addition to radiation therapy and/or surgery. The advancements in next-generation sequencing technology h

Germline BAP1-positive patients: the dilemmas of cancer surveillance and a proposed interdisciplinary consensus monitoring strategy.

The germline BAP1 (BRCA1-associated protein-1) mutation and associated cancer pre-disposition syndrome was first described in 2011. Since then, physicians have considered this diagnosis for patients with a characteristic personal or family history of BAP1-associated tumours (mainly uveal and cutaneous melanoma, pleural/peritoneal mesothelioma, renal cell carcinoma and BAP1-deficient melanocytic lesions). However, a positive germline BAP1 mutation detection creates significant uncertainty in terms of appropriate cancer surveillance. A number of groups have proposed surveillance plans but import

Dietary Isoflavones and Breast Cancer Risk.

Breast cancer is the deadliest neoplasm in women globally, resulting in a significant health burden. In many cases, breast cancer becomes resistant to chemotherapy, radiation, and hormonal therapies. It is believed that genetics is not the major cause of breast cancer. Other contributing risk factors include age at first childbirth, age at menarche, age at menopause, use of oral contraceptives, race and ethnicity, and diet. Diet has been shown to influence breast cancer incidence, recurrence, and prognosis. Soy isoflavones have long been a staple in Asian diets, and there appears to be an incr

Molecular perspectives in differentiated thyroid cancer.

Progress in understanding the molecular genetics of thyroid cancer in the last 20 years has accelerated recently with the advent of high-throughput sequencing technologies known as Next-Generation Sequencing. Besides classical molecular abnormalities involving the MAPK (Mitogen Activated Protein Kinase) and PI3K (PhosphoInositide 3-Kinase) pathways that play a key role in follicular-derived thyroid tumorigenesis, new molecular abnormalities have been discovered. The major advances in recent years have been the discovery of new somatic driver gene point mutations (such as RASAL1 [RAS protein ac

The genetic architecture of human skin pigmentation: evolution and adaptation across global populations.

Human skin pigmentation is a dynamic and highly adaptive trait influenced by genetic, environmental, and cultural factors. It is driven by variation in ultraviolet radiation, governed by a complex polygenic structure, and further modulated through gene-culture interactions. This review synthesizes advances from evolutionary biology, anthropology, molecular genetics, and gene-culture coevolution to provide an integrated framework for understanding global diversity of pigmentation. We take a distinct approach by focusing on the major evolutionary hypotheses and the polygenic architecture underly

Breast Cancer Screening in Women at Higher-Than-Average Risk: Recommendations From the ACR.

Early detection decreases breast cancer mortality. The ACR recommends annual mammographic screening beginning at age 40 for women of average risk. Higher-risk women should start mammographic screening earlier and may benefit from supplemental screening modalities. For women with genetics-based increased risk (and their untested first-degree relatives), with a calculated lifetime risk of 20% or more or a history of chest or mantle radiation therapy at a young age, supplemental screening with contrast-enhanced breast MRI is recommended. Breast MRI is also recommended for women with personal hist

Clonal evolution of a case of treatment refractory maxillary sinus carcinoma.

BACKGROUND: Maxillary sinus carcinoma (MSC) is a rare cancer of the head and neck region. Patients are treated with surgery, radiation therapy, and chemotherapy and the treatment regimen is based on patient's age, general health condition, disease stage, and its extent of spread. There is very little information available on the genetics of this disease. DNA content based flow sorting of tumor cells followed by array comparative genomic hybridization allows for high definition global assessment of distinct clonal changes within tumor populations. METHODS: We applied this technique to primary
