Evolution of radiation-induced temporal lobe injury after intensity-modulated radiation therapy in nasopharyngeal carcinoma: a large cohort retrospective study.

BACKGROUND: Previous studies have demonstrated conflicting findings regarding the initial MRI patterns of radiotherapy-induced temporal lobe injury (RTLI) and the evolution of different RTLI patterns. The aim of this study was to evaluate the initial MRI pattern and evolution of RTLI in patients with nasopharyngeal carcinoma (NPC) by means of a large cohort study. METHODS: Data of patients with RTLI were retrospectively collected from two hospitals between January 2011 and December 2021. The injured lobes were categorized into three patterns based on initial MRI patterns: isolated white matte

Medical countermeasures for radiation combined injury: radiation with burn, blast, trauma and/or sepsis. report of an NIAID Workshop, March 26-27, 2007.

Non-clinical human radiation exposure events such as the Hiroshima and Nagasaki bombings or the Chernobyl accident are often coupled with other forms of injury, such as wounds, burns, blunt trauma, and infection. Radiation combined injury would also be expected after a radiological or nuclear attack. Few animal models of radiation combined injury exist, and mechanisms underlying the high mortality associated with complex radiation injuries are poorly understood. Medical countermeasures are currently available for management of the non-radiation components of radiation combined injury, but it i…

Early HMGB1 Inhibition Reduces Hippocampal Injury During Adolescence in a Young Mouse Model of Radiation-Induced Brain Injury.

Radiotherapy is a primary treatment for childhood malignant brain tumors. However, it often leads to radiation-induced brain injury (RIBI), which significantly impairs neurodevelopment in pediatric patients. Effective treatment for this complication remains limited. Given the established link between RIBI-associated neuroinflammation and HMGB1, we investigated the neuroprotective potential of early HMGB1 inhibition in a young mouse model. Four-week-old male C57BL/6 mice received a single 10 Gy whole-brain irradiation and were divided into Sham + PBS, Rad + PBS, and Rad + glycyrrhizin (GL, 10 m

Recurrent Upper Gastrointestinal Bleeding due to Radiation-Induced Hemorrhagic Gastroduodenal Ectasia: A Review of Current Treatment Options for Radiation-Induced Gastric Injury.

INTRODUCTION: Acute upper gastrointestinal bleeding is one of the most common medical emergencies that present to the hospital, and delineating the underlying etiology is essential to provide adequate definitive treatment. The purpose of this case report was to review the diagnosis and treatment of a rare complication known as radiation-induced hemorrhagic gastritis (RIHG) that can occur in patients with prior radiation exposure. The motivation for this study arose from the identification of a case within our institution. CASE PRESENTATION: The study involved a review of the diagnosis and man

Acute activation of autophagy enables growth plate regeneration following radiation-induced injury.

PURPOSE: Radiation injury to growth plates commonly leads to skeletal late complications including short stature, limb length-discrepancy, and scoliosis/kyphosis in pediatric oncology patients. We aimed to understand the acute responses of direct growth plate irradiation that result in skeletal late complications. MATERIALS AND METHODS: We first established an in vivo murine model of focal growth plate irradiation that recapitulates the clinical development of skeletal late complications and used it to explore the responses of growth plate chondrocytes within the first 72 hours of radiation e

Ferroptosis-immune crosstalk in ionizing radiation-induced testicular injury: mechanisms and therapeutic opportunities.

Accumulating evidence suggests that ferroptosis-an iron-dependent, lipid peroxidation-driven form of regulated cell death-may contribute substantially to ionizing radiation (IR)-induced testicular injury. In this review, we develop a hypothesis-generating framework for ferroptosis-immune crosstalk in the testis. Within this framework, ferroptotic injury to Sertoli cells is proposed to compromise the blood-testis barrier (BTB), potentially facilitating autoantigen exposure and adaptive immune activation; reciprocal inflammatory signaling may, in turn, increase the susceptibility of neighboring

A γ-Ray-Responsive Dynamic Covalent Polymeric Radioprotectant for Efficient Radiation Injury Treatment.

The increasing risk of radiation exposure has intensified the demand for advanced radioprotective strategies to counteract radiation injury. Here, a γ-ray-responsive dynamic covalent polymeric radioprotectant, engineered by integrating phenylboronic acid (PBA)-functionalized polymeric micelles with the potent antioxidants epigallocatechin-3-gallate (EGCG) and curcumin (Cur), is developed to provide efficient systemic protection. The architecture of this dynamic covalent polymeric radioprotectant is stabilized by reversible dynamic covalent linkages between the PBA moieties and the catechol gro

CSF2 drives radiation-induced lung injury by eliciting PADI4-high pathogenic neutrophils.

BACKGROUND: Radiation-induced lung injury (RILI) represents a severe, therapy-refractory complication of thoracic radiotherapy. Although neutrophils have been implicated in RILI pathogenesis, the specific pathogenic subsets and the molecular mechanisms governing their recruitment and functional specialization remain elusive. This study aims to decode the crosstalk between damaged epithelial cells and neutrophils in the RILI microenvironment. METHODS: We established a murine RILI model using 17 Gy thoracic irradiation and performed integrated single-cell RNA sequencing (scRNA-seq) and bulk tra

Keratin nanoparticle-loaded hydrogel microneedles for programmed repair of radiation-induced skin injury.

Radiation-induced skin injury (RISI) poses a significant clinical challenge, with its core pathology involving epidermal barrier disruption caused by excessive reactive oxygen species (ROS) accumulation, oxidative stress, and cell apoptosis. However, effective local treatment options for RISI remain highly limited. Therefore, we developed a phase-transition hydrogel microneedle system loaded with keratin nanoparticles (KNPs@M/G MNs) for the multi-target combinatorial therapy of RISI based on a physical phase-transition mechanism. Specifically, these microneedles penetrate the stratum corneum t

Mitophagy activation for jawbone radiation injury therapy via angiogenesis/osteogenesis-active nanozymes.

Radiotherapy-induced osteoradionecrosis of the jaw (ORNJ) affects quality of life by causing severe pain, persistent infections, and stomatognathic dysfunction. Current ORNJ treatments are limited by several factors, including surgical trauma and notable individual differences. Oxidative stress, which is a critical driver of radiation-induced injury, promotes inflammation and disrupts the balance between bone formation and resorption. Here, we developed ceria-doped silicate nanozymes (CeSNs) to eliminate reactive oxygen species (ROS) by inducing a superoxide dismutase-like (SOD-like) and catal

Intranasal administration of Aloe-derived nanovesicles attenuates early radiation-induced lung injury.

BACKGROUND: Radiation-induced lung injury (RILI) is a common complication of thoracic radiotherapy and typically progresses from acute radiation pneumonitis to late-stage pulmonary fibrosis. This study investigated whether intranasally delivered Aloe vera-derived nanovesicles (AVNVs) could attenuate early lung injury following high-dose thoracic irradiation. METHODS: A single-fraction 20 Gy whole-thorax irradiation model was established in C57BL/6 N mice. Mice in the treatment group received intranasal AVNVs at 5 mg kg-1 day-1 for three consecutive days. Histopathological changes were evaluat

The role of macrophages in radiation-induced lung injury: from pathological mechanisms to therapeutic targets.

Radiation-induced lung injury (RILI) is a major dose-limiting toxicity of chest radiotherapy (RT), and its occurrence and development are closely related to the complex role of macrophages. RT significantly modulates the recruitment and functional status of macrophages. In the early stage, macrophages predominantly exhibit a pro-inflammatory M1 phenotype, releasing a large amount of inflammatory factors and exacerbating lung tissue damage. In the late stage, they shift to a pro-fibrotic M2 phenotype, promoting fibroblast activation, extracellular matrix accumulation, and epithelial-mesenchymal
