Characterization of expression patterns and dynamic relocation of Notch proteins during acrosome reaction of bull spermatozoa.

Notch is a conserved cell-signaling pathway involved in spermatogenesis regulation. This study firstly evaluated the presence, localization patterns, acquisition origin and relation to acrosome reaction of Notch proteins in bull sperm. Western Blot analysis detected all Notch proteins in ejaculated bull sperm, and immunostaining described their specific sperm localization. Recovery of sperm from different segments showed that Notch proteins have testicular origin (NOTCH1, NOTCH2, DLL4), are sequentially acquired during sperm maturation along epididymal transit (NOTCH3, DLL3, JAGGED1-2), or pos

Osteohematology: To be or Notch to be.

Osteohematology is an emerging research field that studies the crosstalk between hematopoietic and bone stromal cells, to elucidate the mechanisms of hematological and skeletal malignancies and diseases. The Notch is an evolutionary conserved developmental signaling pathway, with critical roles in embryonic development by controlling cell proliferation and differentiation. However, the Notch pathway is also critically involved in cancer initiation and progression, such as osteosarcoma, leukemia, and multiple myeloma. The Notch-mediated malignant cells dysregulate bone and bone marrow cells in

The Relevance of Notch Signaling in Cancer Progression.

The Notch signaling pathway controls normal embryonic development and tissue homeostasis of many cell types. It regulates cell proliferation, fate, differentiation, and cell death by short-range signaling between nearby cells that come in contact. The Notch pathway has also been critically involved in the pathobiology of a variety of malignancies, regulating cancer initiation and development, as well as early stages of cancer progression, by adjusting conserved cellular programs. Fibroblasts, an essential for tumor growth component of stroma, have also been affected by Notch regulation. Sequen

Distinct and shared effects of individual γ-secretase inhibitors on human keratinocytes.

Gamma secretase (γ-secretase) is an enzymatic transmembrane complex responsible for the cleavage of over 100 substrates, including notch, E-cadherin, amyloid precursor protein, specific cytokine receptors, and other type I transmembrane proteins. Genetic variants in the γ-secretase complex are associated with familial hidradenitis suppurativa (HS), where they are predicted to inactivate enzymatic function. Further, γ-secretase inhibitors cause HS-like pathologies in patients with desmoid tumors. However, it is unclear how modulation of γ-secretase activity in keratinocytes impacts cellular pro

Liver Sinusoidal Endothelial Cells Promote the Expansion of Human Cord Blood Hematopoietic Stem and Progenitor Cells.

Cord blood (CB) is an attractive source of hematopoietic stem cells (HSCs) for hematopoietic cell transplantation. However, its application remains limited due to the low number of HSCs/progenitors in a single CB unit and its notoriously difficulty in expanding ex vivo. Here, we demonstrated that the human fetal liver sinusoidal endothelial cells engineered to constitutively express the adenoviral E4orf1 gene (hFLSECs-E4orf1) is capable of efficient expansion ex vivo for human CB hematopoietic stem and progenitor cells (HSPCs). Coculture of CD34+ hCB cells with hFLSECs-E4orf1 resulted in gener

The tailless ortholog nhr-67 functions in the development of the C. elegans ventral uterus.

The development of the C. elegans uterus provides a model for understanding the regulatory pathways that control organogenesis. In C. elegans, the ventral uterus develops through coordinated signaling between the uterine anchor cell (AC) and a ventral uterine (VU) cell. The nhr-67 gene encodes the nematode ortholog of the tailless nuclear receptor gene. Fly and vertebrate tailless genes function in neuronal and ectodermal developmental pathways. We show that nhr-67 functions in multiple steps in the development of the C. elegans uterus. First, it functions in the differentiation of the AC. Sec

Mechanisms of Notch signaling in cancer drug resistance.

One of the most common treatments used for cancer is chemotherapy. Different drugs are used in chemotherapy depending on the type of cancer. In this regard, one of the most basic problems in chemotherapy is drug resistance, in which people undergoing treatment develop resistance to chemotherapy drugs over time. Therefore, the drug dose must be increased over time or the interval between treatments must be reduced, which leads to side effects and damage to normal cells and tissues. Various factors play a role in drug resistance, including ATP-binding cassette (ABC) transporters, which lead to d

Role of Notch signaling in corticalization.

Bone is a complex structure formed by cancellous and cortical bone compartments. Whereas cancellous bone remodeling has been studied extensively, less is known about mechanisms that lead to cortical bone formation and homeostasis. Corticalization is a poorly understood process, when newly formed trabeculae coalesce to consolidate as cortical bone in early life, but with an impact on skeletal integrity that is sustained throughout life. Failure to form cortical bone results in pronounced skeletal fragility, although the process is poorly understood. Mouse models of Sp7, Itgb3, Socs3 and Clcn7 i

Adenovirus-mediated effects of Wnt and Notch signalling pathways on hair cell regeneration in mice.

Some genes are delivered to cochleae by adenoviruses to restore partial hearing function. This provides promising prospects for gene therapies for hearing loss from hair cell damage. To study the adenovirus (AD)-mediated effect of the Wnt and Notch signalling pathways on hair cell regeneration in the mouse cochlea, we constructed a β-catenin-adenovirus (β-catenin-AD) to increase the activity of the Wnt signalling pathway and a NICD (intracellular domain of Notch1)-RNAi-adenovirus to decrease the activity of the Notch signalling pathway (NICD-RNAi-AD). Our study indicated that approximately 40%

Periodic formation of epithelial somites from human pluripotent stem cells.

During embryonic development, epithelial cell blocks called somites are periodically formed according to the segmentation clock, becoming the foundation for the segmental pattern of the vertebral column. The process of somitogenesis has recently been recapitulated with murine and human pluripotent stem cells. However, an in vitro model for human somitogenesis coupled with the segmentation clock and epithelialization is still missing. Here, we report the generation of human somitoids, organoids that periodically form pairs of epithelial somite-like structures. Somitoids display clear oscillatio

Notch signaling at the crossroads of innate and adaptive immunity.

Notch signaling is an evolutionarily conserved cell-to-cell signaling pathway that regulates cellular differentiation and function across multiple tissue types and developmental stages. In this review, we discuss our current understanding of Notch signaling in mammalian innate and adaptive immunity. The importance of Notch signaling is pervasive throughout the immune system, as it elicits lineage and context-dependent effects in a wide repertoire of cells. Although regulation of binary cell fate decisions encompasses many of the functions first ascribed to Notch in the immune system, recent ad

NRF2 mitigates radiation-induced hematopoietic death.

Fractionated, high-dose total body irradiation (TBI) is used therapeutically to myeloablate and immune suppress patients undergoing hematopoietic stem cell (HSC) transplantation. Acute exposure to ionizing radiation can have fatal effects on the hematopoietic and immune systems. Currently, therapies aimed at ameliorating ionizing radiation-associated toxicities are limited. In the February 2014 issue of the JCI, Kim and colleagues demonstrated that induction of nuclear factor erythroid 2-related factor 2 (NRF2) enhances HSC regeneration and increases survival following ionizing radiation expos

[Pathomechanisms and treatment of CADASIL].

The pathomechanisms of cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) are still under debate. Granular osmiophilic material (GOM), which accumulates around the basement membrane, and the extracellular domain of NOTCH3 (NECD) in the vessel are key molecules that contributes to the destruction of smooth muscle cells in CADASIL. In addition, GOM and NECD may be related to the dysfunction of cerebral small vessels in patients with CADASIL. In this review, the role of the accumulation of these abnormal proteins in the cerebral small vessels, and

The Notch signaling pathway in retinal dysplasia and retina vascular homeostasis.

The retina is one of the most essential elements of vision pathway in vertebrate. The dysplasia of retina cause congenital blindness or vision disability in individuals, and the misbalance in adult retinal vascular homeostasis leads to neovascularization-associated diseases in adults, such as diabetic retinopathy or age-related macular degeneration. Many developmental signaling pathways are involved in the process of retinal development and vascular homeostasis. Among them, Notch signaling pathway has long been studied, and Notch signaling-interfered mouse models show both neural retina dyspla

Identifying NOTCH signaling-specialized hematopoietic supportive subpopulation from mesenchymal stem cells.

BACKGROUND: Human umbilical cord mesenchymal stem cells (UC-MSCs) are widely studied as a promising cellular therapy for a variety of diseases due to their accessible sources, less of ethical concerns, and capacities for immunomodulation and tissue repair. However, cellular heterogeneity arising during in vitro expansion imposes challenges in quality control for clinical applications and transform, requiring further investigation. METHODS: Two fetal umbilical cords were obtained from Sinogene C&G Stem Cells Co., Ltd. Primary UC-MSCs were isolated and subjected to continuous in vitro passaging

Adgrg6/Gpr126 is required for compact wall integrity and establishing trabecular identity during cardiac trabeculation.

How adhesion G protein-coupled receptors (aGPCRs) control development remains unclear. aGPCR Adgrg6/Gpr126 has been associated with heart trabeculation. Defects in this process cause cardiomyopathies and cardiac dysfunction. How cardiomyocytes attain trabecular identity is poorly understood. Here, we show that different domains of Gpr126 distinctly regulate compact wall integrity and trabecular identity. Maternal zygotic (MZ) gpr126stl47 early truncation mutants exhibit hypotrabeculation, whereby N-cadherin distributes randomly along apical/basal/lateral membranes of compact layer cardiomyocyt

Abrogation of stemness in osteosarcoma by the mithramycin analog EC-8042 is mediated by its ability to inhibit NOTCH-1 signaling.

Osteosarcomas are frequently associated to a poor prognosis and a modest response to current treatments. EC-8042 is a well-tolerated mithramycin analog that has demonstrated an efficient ability to eliminate tumor cells, including cancer stem cell subpopulations (CSC), in sarcomas. In transcriptomic and protein expression analyses, we identified NOTCH1 signaling as one of the main pro-stemness pathways repressed by EC-8042 in osteosarcomas. Overexpression of NOTCH-1 resulted in a reduced anti-tumor effect of EC-8042 in CSC-enriched 3D tumorspheres cultures. On the other hand, the depletion of

Notch4 Signaling Induces a Mesenchymal-Epithelial-like Transition in Melanoma Cells to Suppress Malignant Behaviors.

The effects of Notch signaling are context-dependent and both oncogenic and tumor-suppressive functions have been described. Notch signaling in melanoma is considered oncogenic, but clinical trials testing Notch inhibition in this malignancy have not proved successful. Here, we report that expression of the constitutively active intracellular domain of Notch4 (N4ICD) in melanoma cells triggered a switch from a mesenchymal-like parental phenotype to an epithelial-like phenotype. The epithelial-like morphology was accompanied by strongly reduced invasive, migratory, and proliferative properties
