Structural and functional changes in the retina in Parkinson's disease.

Parkinson's disease is caused by degeneration of dopaminergic neurons, originating in the substantia nigra pars compacta and characterised by bradykinesia, rest tremor and rigidity. In addition, visual disorders and retinal abnormalities are often present and can be identified by decreased visual acuity, abnormal spatial contrast sensitivity or even difficulty in complex visual task completion. Because of their early onset in patients with de novo Parkinson's disease, the anatomical retinal changes and electrophysiological modification could be valuable markers even at early stages of the dise…

[In vivo cellular imaging of the retina: present and future].

Capn5 Expression in the Healthy and Regenerating Zebrafish Retina.

PURPOSE: Autosomal dominant neovascular inflammatory vitreoretinopathy (ADNIV) is a devastating inherited autoimmune disease of the eye that displays features commonly seen in other eye diseases, such as retinitis pigmentosa and diabetic retinopathy. ADNIV is caused by a gain-of-function mutation in Calpain-5 (CAPN5), a calcium-dependent cysteine protease. Very little is known about the normal function of CAPN5 in the adult retina, and there are conflicting results regarding its role during mammalian embryonic development. The zebrafish (Danio rerio) is an excellent animal model for studying v…

A biochemical basis for induction of retina regeneration by antioxidants.

The use of antioxidants in tissue regeneration has been studied, but their mechanism of action is not well understood. Here, we analyze the role of the antioxidant N-acetylcysteine (NAC) in retina regeneration. Embryonic chicks are able to regenerate their retina after its complete removal from retinal stem/progenitor cells present in the ciliary margin (CM) of the eye only if a source of exogenous factors, such as FGF2, is present. This study shows that NAC modifies the redox status of the CM, initiates self-renewal of the stem/progenitor cells, and induces regeneration in the absence of FGF2…

Inhibitory Interneurons in the Retina: Types, Circuitry, and Function.

Visual signals in the vertebrate retina are shaped by feedback and feedforward inhibition in two synaptic layers. In one, horizontal cells establish fundamental center-surround receptive-field properties via morphologically and physiologically complex synapses with photoreceptors and bipolar cells. In the other, a panoply of amacrine cells imbue ganglion cell responses with spatiotemporally complex information about the visual world. Here, I review current ideas about horizontal cell signaling, considering the evidence for and against the leading, competing theories. I also discuss recent work…

Role of melatonin and its receptors in the vertebrate retina.

Melatonin is a chemical signal of darkness that is produced by retinal photoreceptors and pinealocytes. In the retina, melatonin diffuses from the photoreceptors to bind to specific receptors on a variety of inner retinal neurons to modify their activity. Potential target cells for melatonin in the inner retina are amacrine cells, bipolar cells, horizontal cells, and ganglion cells. Melatonin inhibits the release of dopamine from amacrine cells and increases the light sensitivity of horizontal cells. Melatonin receptor subtypes show differential, cell-specific patterns of expression that are l…

Apelin is required for non-neovascular remodeling in the retina.

Retinal pathologies are frequently accompanied by retinal vascular responses, including the formation of new vessels by angiogenesis (neovascularization). Pathological vascular changes may also include less well characterized traits of vascular remodeling that are non-neovascular, such as vessel pruning and the emergence of dilated and tortuous vessel phenotypes (telangiectasis). The molecular mechanisms underlying neovascular growth versus non-neovascular remodeling are poorly understood. We therefore undertook to identify novel regulators of non-neovascular remodeling in the retina by using …

Blood flow in the peripapillary retina in exfoliation glaucoma.

PURPOSE: The aim of this article is to study peripapillary retinal blood flow in patients with progressive and stable exfoliation glaucoma (ExG). METHODS: Fifty-eight eyes with ExG were included; 25 of them had progressive and 33 stable glaucoma. Retinal blood flow in the peripapillary retina was measured with scanning laser Doppler flowmetry. Acquired flow maps were analysed with the automatic full-field perfusion image analyser. Multiple logistic regression was used to model progression of glaucoma. RESULTS: Mean retinal flow (MF; correlation coefficient, P-value; R = 0.36, P = 0.006) and …

Neural responses elicited by electrical stimulation of the retina.

PURPOSE: To study electrically elicited responses (EERs) that are produced by epiretinal stimulation of normal and degenerated retina. METHODS: Three biological models of retinal degeneration are compared: normal and rd1 mouse, normal and RCD1 dog, and human with retinitis pigmentosa. In mouse, epiretinal stimulation was accomplished by means of a wire inserted in the vitreous cavity, and single-unit activity was recorded in visual cortex. In dog and human, an implantable retinal stimulator was used to stimulate the retina, and evoked potentials were recorded from the cortical surface (dog) o…

[Effect of nitrites and nitrates on bovine retina in vitro].

BACKGROUND/AIM: Uveitis is an endocular inflammation that forms one of the most serious manifestations of Behçet disease. Nitric oxide (NO) is a molecule that expresses important immunoinflammatory properties and is produced by NO synthases (NOS). In previous studies, we showed an elevated production of endogenous NO during Behçet's and idiopathic uveitis. These results led us to investigate the effect of nitrites (NO2-) and nitrates (NO3-) (physiologically stable metabolites of NO) on fragments of bovine retina in order to determine the role of nitric oxide (NO) in the genesis of retinal lesi…

Melanopsin expression in the chick retina and pineal gland.

The chick retina and pineal gland exhibit circadian oscillations in biochemical and physiological processes in vivo and in vitro and entrain to 24 h LD cycles. However, the phototransductive mechanisms responsible for entrainment are largely unknown. Experiments utilizing mice lacking functional rods and cones reveal normal circadian responses to light. These studies also implicate a subset of photoresponsive cells in the ganglion cell layer (GCL) which express melanopsin (opn4). The current study investigates opn4 mRNA distribution and regulation in the chick. Opn4 mRNA is widespread in the c…

GABA content in the retina of pigmented and albino rats.

Reduction of the melanin precursor DOPA associated with albinism leads to spatiotemporal disturbances in retinal neurogenesis and thus seems to be responsible for numerous neuronal alterations found in albino retinae. To investigate whether these cellular alterations are reflected in retinal neurotransmitter concentrations we compared the levels of GABA and glutamate in the retina of adult pigmented Long Evans and albino Wistar rats using reversed phase-liquid chromatography (RP-HPLC). When normalized to retinal weight, GABA levels showed a statistically insignificant trend to be lower and glu…

Aprosencephaly: histopathological features of the rudimentary forebrain and retina.

Aprosencephaly/atelencephaly is an extremely rare central nervous system defect of the human fetus, which has been attributed to an encephaloclastic process. We report here the histopathological features of the rudimentary forebrain and retina of a case of aprosencephaly in a male fetus that was examined at 20 weeks of gestation, in which there was no evidence of destruction. The prosencephalon was excessively small and replaced by a solid, cylindrical mass without a hemispheric cleavage or a ventricle. Histologically, the rudimentary forebrain tissue consisted of primitive cells that had accu…

Astrocyte invasion and vasculogenesis in the developing ferret retina.

We studied the time course of astrocyte invasion and blood vessel formation in the developing ferret retina using glial fibrillary acidic protein (GFAP)-immunohistochemistry for astrocytes and isolectin B4 histochemistry for blood vessels. As in other mammals, strongly GFAP positive astrocytes invade the ferret retina from the optic nerve. At birth, strongly GFAP positive astrocytes have reached about 22% of the distance between optic disc and outer retinal edge whereas weakly GFAP positive processes already extend to the edge of the retina. At postnatal days P30-P37 about 82% of the distance …

Localization and characterization of immunocompetent cells in the human retina.

PURPOSE: Recent studies have shown that experimental uveitis can be induced by the appropriate administration of various retinal antigens. Little is known about the in-situ interactions between immune cells in the retina as a prerequisite for understanding the mechanisms involving the presentation of antigens by local antigen-presenting cells (APC) to invading T cells. The study described here was therefore designed to investigate the presence of immunocompetent cells with a focus on the characterization of retinal APCs. MATERIALS AND METHODS: Retinal wholemounts, cytospins, and ocular sectio…
