Nanoengineering of therapeutics for retinal vascular disease.

Retinal vascular diseases, including diabetic retinopathy, neovascular age related macular degeneration, and retinal vein occlusion, are leading causes of blindness in the Western world. These diseases share several common disease mechanisms, including vascular endothelial growth factor (VEGF) signaling, hypoxia, and inflammation, which provide opportunities for common therapeutic strategies. Treatment of these diseases using laser therapy, anti-VEGF injections, and/or steroids has significantly improved clinical outcomes. However, these strategies do not address the underlying root causes of

The role of glia in retinal vascular disease.

Retinal vascular diseases collectively represent a leading cause of blindness. Unsurprisingly, pathological characterisation and treatment of retinal 'vascular' diseases have primarily focused on the aetiology and consequences of vascular dysfunction. Far less research has addressed the contribution of neuronal and glial dysfunction to the disease process of retinal vascular disorders. Ample evidence now suggests that retinal vasculopathy only uncommonly occurs in isolation, usually existing in concert with neuropathy and gliopathy. Retinal glia (Müller cells, astrocytes and microglia) have be

Minor retinal degeneration in Parkinson's disease.

Parkinson's disease is a neurodegenerative disorder with selective and progressive loss of dopaminergic neurons in substantia nigra. Studies on Parkinson's disease patients and dopamine-depleted animals indicate that dopaminergic neurons in the retina degenerate due to the genetic and environmental factors that cause dopaminergic neuron loss in the substantia nigra. Besides motor and non-motor symptoms, visual symptoms are common in Parkinson's disease patients, ranging from complaints of reading and driving difficulties, to complex visual hallucinations. The delicate network of various neuron

Functional retinal changes in Gaucher disease.

In Gaucher disease, sphingolipid glucosylceramide is accumulated in cells of the reticulo-endothelial system. This leads to the formation of "Gaucher cells," which are enlarged macrophages. In the eye, circumscript preretinal deposits are a pathognomonic feature. We describe the case of a 23-year-old woman with no subjective complaints but a reduced ERG response, leading to the suspicion that in addition to circumscript deposition diffuse accumulation of metabolic endproducts might be present in the retina in Gaucher patients.

Retinal degenerative and hypoxic ischemic disease.

A broad spectrum of retinal diseases affects both the retinal vasculature and the neural retina, including photoreceptor and postreceptor layers. The accepted clinical hallmarks of acute retinopathy of prematurity (ROP) are dilation and tortuosity of the retinal vasculature. Additionally, significant early and persistent effects on photoreceptor and postreceptor neural structures and function are demonstrated in ROP. In this paper, we focus on the results of longitudinal studies of electroretinographic (ERG) and vascular features in rats with induced retinopathies that model the gamut of human…

[Angioproliferative retinal disease caused by ischemia].

Ischemia is a major stimulus for angiogenesis, a biological response mechanism that describes the formation of new blood vessels from existing vessels. An ischemic cell communicates with endothelial cells by soluble factors such as VEGF (vascular endothelial growth factor) and its receptors. A major transcriptional factor for VEGF is HIF-1 (hypoxia inducible factor). Proliferation of endothelial cells alone does not result in stable vascular tubes, this is only achieved by recruiting additional cells such as pericytes. The stabilisation and destabilisation of vessels, which are important prere

Retinal masqueraders of optic nerve disease.

Electrophysiologic testing of the visual system and ancillary imaging investigations may be crucial aids for correctly diagnosing patients with unexplained visual loss or isolated disc pallor. These patients may harbor a retinal disease process and not an optic neuropathy. A diverse range of retinal conditions may present in this manner, and misdiagnosis may lead to unnecessary neurologic investigation and subsequent delay in therapeutic intervention.

Visualization of retinal vasculitis in Eales' disease.

Between 1970 and 1991 the authors examined 466 patients with Eales' disease. The mean age at diagnosis was 30, ranging between 14 and 55 years. The mean follow-up period was 43.5 months. At the initial examination, 356 cases were bilateral and 110 cases were unilateral (822 eyes). Vitreous hemorrhage was present in 257 of the 822 eyes. In the remaining 565 eyes, the major retinal lesions were retinal neovascularization (40.7%), vascular sheathing (20.7%), vascular sheathing and retinal hemorrhages (10.6%), retinitis proliferans (9.4%), disc neovascularization (9.0%), branch vein occlusion (3.2

Chromatic and luminosity processing in retinal disease.

Color vision loss can be an early sign of eye disease; in many retinal disorders the loss precedes any change in visual acuity. Noninvasive psychophysical methods allow factoring out of preretinal, receptoral, and postreceptoral (neural) components of the color vision change. A loss of chromatic but not achromatic sensitivity occurs for diabetics; the loss is selective for pathways subserving blue-sensitive photoreceptors. Both chromatic and achromatic pathways are altered in glaucoma and senile macular degeneration; the most marked change in central serous choroidopathy is loss of sensitivity

Ultrasonic patterns in vitreo-retinal disease.

The reliability of diagnostic ultrasound in the assessment of vitreo-retinal disease was studied using an all-purpose ultrasonic machine. Five vitreous echo patterns are described, two representing retinal detachment, two demonstrating the features of vitreous membranes (condensation of vitreous tissue) and one typical of simple vitreous haemorrhage. Retinal detachment was correctly diagnosed in over 95% of cases and simple vitreous haemorrhage in 100% of cases. The diagnostic accuracy rate for vitreous membranes was 80--90%. The most frequent source of error arose in the differentiation of lo

Subretinal suspensions of hiPSC-derived retinal pigment epithelium cells form functional monolayers in NOD-SCID mice facilitating treatment of advanced retinal diseases.

INTRODUCTION: Transplantation of human induced pluripotent stem cells derived retinal pigment epithelium (hiPSC-RPE) is regarded as one of the most promising strategies for advanced retinal degenerative diseases leading to blindness, such as age-related macular degeneration. Despite its therapeutic potential, this approach is encumbered by critical challenges, notably the survival of donor RPE cells post-transplantation and the successful reconstruction of a functional RPE layer. METHODS: With our previously reported strategy, abundant hiPSC-RPEs were generated from human induced pluripotent

Mitophagy, Ferritinophagy and Ferroptosis in Retinal Pigment Epithelial Cells Under High Glucose Conditions: Implications for Diabetic Retinopathy and Age-Related Retinal Diseases.

Diabetic retinopathy (DR) is a devastating disease leading to blindness among majority of working adults around the globe. Nonetheless, an effective treatment or cure for the disease is still to be achieved. This is because the cellular and molecular mechanisms of DR are complex and not fully understood yet. In this article, we describe how high glucose induced TXNIP upregulation and associated redox stress may cause mitochondrial dysfunction, mitophagy, ferritinophagy (iron release by autophagy) and lysosome destabilization. Labile irons react with hydrogen peroxide (H2O2) to generate hydroxy

Automated quantification of retinal non-perfusion areas in ultra-widefield SS-OCTA image of patient with retinal vascular diseases by NP-Unet model.

PURPOSE: Retinal non-perfusion areas (NPAs) are a crucial biomarker for assessing the degree of retinal ischaemia of patients with retinal vascular diseases. Accurate identification of NPA is useful for assessing therapeutic approaches, guiding laser treatments and predicting prognosis. The ultra-widefield swept-source optical coherence tomography angiography (UWF-SS-OCTA) has emerged as a superior non-invasive imaging modality with an outstanding ability to detect NPA. This study aimed to establish an efficient NP-Unet model to accurately segment and quantify NPA in the UWF-SS-OCTA image of b

Retinal vascular signatures predict risk of depression and subsequent chronic diseases: A longitudinal study.

OBJECTIVE: Depression poses major global health challenges and accelerates chronic disease development, yet remains undertreated due to reliance on subjective diagnostics. The retina, as an embryological extension of the central nervous system and the organ allowing direct visualization of neurovascular structures, has been demonstrated to serve as an accessible window for systemic health assessment. This study aimed to prospectively examine associations of retinal vascular features with incident depression in healthy individuals and subsequent chronic disease development in those with depress

The pathogenesis of müller cell glial-mesenchymal transition in retinal fibrosis-related eye diseases.

Retinal fibrosis represents an important pathological feature of several vision-threatening ocular diseases, including advanced diabetic retinopathy and neovascular age-related macular degeneration. Its progression may be associated with reactive changes in Müller glial cells and their acquisition of mesenchymal-like phenotypes. As the predominant glial population in the retina, Müller glial cells play crucial physiological roles in maintaining retinal homeostasis. However, under pathological conditions, they can become reactive and acquire mesenchymal- or myofibroblast-like features through a
