Ribosomal protein eL22 contributes to the assembly of 60S ribosomal subunits in Saccharomyces cerevisiae.

Ribosome biogenesis is a highly coordinated pathway that involves the assembly of ribosomal RNAs (rRNAs) with ribosomal proteins (r-proteins) to generate functional ribosomal subunits (r-subunits). The Saccharomyces cerevisiae (yeast) large 60S r-subunit consists of three rRNA molecules and 46 r-proteins. The contributions of nearly all r-proteins of the yeast large r-subunit have been characterized; however, a few non-essential proteins remain poorly understood. Although non-essential, human eL22 has been identified as a key player in p53 regulation during ribosomal stress and as a highly mut…

Saccharomyces boulardii as a Probiotic Supplement in Helicobacter pylori Eradication Therapy: A Meta-analysis.

BACKGROUND AND AIMS: Helicobacter pylori (H. pylori) is an obligate pathogen affecting approximately 50% of the world population. With rising antibiotic resistance rates in H. pylori and increasing dropout rates from eradication therapies due to adverse effects, H. pylori treatment is increasingly challenged. Saccharomyces boulardii (S. boulardii), a probiotic yeast, was shown to reduce infection- and therapy-associated adverse events and increase H. pylori eradication rates. Our meta-analysis summarizes the beneficial effects of S. boulardii supplementation on H. pylori eradication and therap…

Genetic Diversity analysis of industrial Saccharomyces cerevisiae strains and their correlation with fermentation performance.

Sugarcane molasses serves as a vital non-food feedstock for ethanol fermentation, and its efficient utilisation depends on breeding of excellent microbial strains. Therefore, identification of industrial strains for ethanol fermentation is central to improving fermentation efficiency. To identify high-efficiency ethanol-fermenting strains, we developed a method for preparing high-throughput polymerase chain reaction (PCR) templates within 3 min using seven industrial Saccharomyces cerevisiae strains, integrating single-cell morphology and phenotypic variations in ethanol fermentation. Method s…

Characteristics of Two Saccharomyces cerevisiae Strains and Their Extracellular Vesicles as New Candidates for Probiotics.

There is a huge disparity between the number of bacterial and yeast probiotics in favor of the former. The latest reports indicate that extracellular vehicles (EVs) play a significant role in probiotic mechanisms. In the present work, we compared the probiotic properties of Saccharomyces cerevisiae strains (WUT3 and WUT151), which have never been previously characterized in this context, with commercial probiotic yeast-Saccharomyces cerevisiae var. boulardii CNCM-745. Notably, WUT3 and WUT151 reacted more mildly to the unfavorable simulated environment of saliva, stomach, small, and large inte…

Cell surface display of VP1 of foot-and-mouth disease virus on Saccharomyces cerevisiae.

BACKGROUND: Foot-and-mouth disease (FMD) is a highly infectious disease that mainly affects cloven-hoofed animals. The rapid spread of this disease hinders the success of control measures. This necessitates the urgent development of a vaccine to ensure the safe rearing of affected animals. However, traditional vaccines have some drawbacks including partial protection against persistent infection and a lack of cross-protection against different strains. RESULTS: We developed an oral vaccine that targets VP1, the major immunogenic protein of the causative FMD virus (FMDV). We expressed this pro

Possible regulatory network and associated pathways governing the expression of ADH2 in Saccharomyces cerevisiae.

With the day to day increase in energy consumption due to increase in urbanization production of bioethanol is highly in demand. At this point where the traditional methods are not able to suffice the demands due to its high cost and low productivity, new methods need to be developed. This review aims to understand the importance and the regulation of ADH2 in Saccharomyces cerevisiae because Adh2p is the only enzyme that initiates the reaction for the conversion of ethanol, the end product of fermentation to acetaldehyde. The effect of glucose on regulatory mechanisms of Alcohol dehydrogenase …

Development and Characterization of Probiotic Beers with Saccharomyces boulardii as an Alternative to Conventional Brewer's Yeast.

The development of new non-dairy probiotic foods is interesting, given lactose intolerance, milk allergies, and the growing trend of vegetarianism. In this paper, beer has been used as a probiotic delivery matrix, using Saccharomyces boulardii as an alternative to conventional brewer's yeast. The strain was able to grow in worts prepared with hops containing different alpha-acid concentrations, attaining in all cases a final cell concentration above 1·108 cells mL-1. Some differences were found in the physicochemical parameters of beers brewed with S. boulardii compared to those brewed with a …

In-depth understanding of molecular mechanisms of aldehyde toxicity to engineer robust Saccharomyces cerevisiae.

Aldehydes are ubiquitous electrophilic compounds that ferment microorganisms including Saccharomyces cerevisiae encounter during the fermentation processes to produce food, fuels, chemicals, and pharmaceuticals. Aldehydes pose severe toxicity to the growth and metabolism of the S. cerevisiae through a variety of toxic molecular mechanisms, predominantly via damaging macromolecules and hampering the production of targeted compounds. Compounds with aldehyde functional groups are far more toxic to S. cerevisiae than all other functional classes, and toxic potency depends on physicochemical charac…

Persistence of Resident and Transplanted Genotypes of the Undomesticated Yeast Saccharomyces paradoxus in Forest Soil.

One might expect yeasts in soil to be highly dispersed via water or insects, forming ephemeral, genetically heterogeneous populations subject to competition and environmental stochasticity. Here, we report persistence of genotypes of the yeast Saccharomyces paradoxus in space and time. Within 1 km2 in a mixed hardwood forest on scales from centimeters to tens of meters, we detected persistence over 3 years of native genotypes, identified by single nucleotide polymorphisms (SNPs) genome-wide, of the wild yeast Saccharomyces paradoxus growing around Quercus rubra and Quercus alba Yeasts were rec…

Inhibition of autophagy contributes to the toxicity of cadmium telluride quantum dots in Saccharomyces cerevisiae.

Cadmium telluride quantum dots (CdTe QDs) are used as near-infrared probes in biologic and medical applications, but their cytological effects and mechanism of potential toxicity are still unclear. In this study, we evaluated the toxicity of CdTe QDs of different sizes and investigated their mechanism of toxicity in the yeast Saccharomyces cerevisiae. A growth inhibition assay revealed that orange-emitting CdTe (O-CdTe) QDs (half inhibitory concentration [IC50] =59.44±12.02 nmol/L) were more toxic than green-emitting CdTe QDs (IC50 =186.61±19.74 nmol/L) to S. cerevisiae. Further studies on tox…

Impact of photocatalysis on fungal cells: depiction of cellular and molecular effects on Saccharomyces cerevisiae.

We have investigated the antimicrobial effects of photocatalysis on the yeast model Saccharomyces cerevisiae. To accurately study the antimicrobial mechanisms of the photocatalytic process, we focused our investigations on two questions: the entry of the nanoparticles in treated cells and the fate of the intracellular environment. Transmission electronic microscopy did not reveal any entry of nanoparticles within the cells, even for long exposure times, despite degradation of the cell wall space and deconstruction of cellular compartments. In contrast to proteins located at the periphery of th…
