Co-administration of rIpaB domain of Shigella with rGroEL of S. Typhi enhances the immune responses and protective efficacy against Shigella infection.

Shigella species cause severe bacillary dysentery in humans and are associated with high morbidity and mortality. The Invasion plasmid antigen (IpaB) protein, which is conserved across all Shigella spp., induces macrophage cell death and is required to invade host cells. The present study evaluates the immunogenicity and protective efficacy of the recombinant (r) domain region of IpaB (rIpaB) of S. flexneri. rIpaB was administered either alone or was co-administered with the rGroEL (heat shock protein 60) protein from S. Typhi as an adjuvant in a mouse model of intranasal immunization. The Ipa…

Shigella flexneri undergoes niche-specific adaptation during infection.

Shigella flexneri is a leading cause of diarrheal disease worldwide, yet bacterial factors required within distinct host niches remain poorly understood. Here, we used genome-wide transposon sequencing in an infant rabbit model of shigellosis to identify genes promoting bacterial fitness in vivo. The screen identified virulence factors on the large virulence plasmid and novel chromosomal fitness factors. Characterization of zitB, encoding a cation diffusion facilitator family Zn transporter, revealed a niche-specific fitness contribution. The ΔzitB mutant showed no growth defect in vitro in ri…

Detection of OXA-181 Carbapenemase in Shigella flexneri.

We report the detection of OXA-181 carbapenemase in an azithromycin-resistant Shigella spp. bacteria in an immunocompromised patient. The emergence of OXA-181 in Shigella spp. bacteria raises concerns about the global dissemination of carbapenem resistance in Enterobacterales and its implications for the treatment of infections caused by Shigella bacteria.

Molecular and Cellular Mechanisms of Shigella flexneri Dissemination.

The intracellular pathogen Shigella flexneri is the causative agent of bacillary dysentery in humans. The disease is characterized by bacterial invasion of intestinal cells, dissemination within the colonic epithelium through direct spread from cell to cell, and massive inflammation of the intestinal mucosa. Here, we review the mechanisms supporting S. flexneri dissemination. The dissemination process primarily relies on actin assembly at the bacterial pole, which propels the pathogen throughout the cytosol of primary infected cells. Polar actin assembly is supported by polar expression of the…

Macrophage Apoptosis Triggered by IpaD from Shigella flexneri.

Shigellosis, a potentially severe bacillary dysentery, is an infectious gastrointestinal disease caused by Shigella spp. Shigella invades the human colonic epithelium and avoids clearance by promoting apoptosis of resident immune cells in the gut. This process is dependent on the Shigella type III secretion system (T3SS), which injects effector proteins into target cells to alter their normal cellular functions. Invasion plasmid antigen D (IpaD) is a structural component that forms a complex at the tip of the T3SS apparatus needle. Recently, IpaD has also been shown to indirectly induce apopto…

Symptomatic Shigella sonnei urinary tract infection in pregnancy.

PURPOSE OF INVESTIGATION: This report describes a case of urinary tract infection (UTI) due to Shigella sonnei during pregnancy. METHODS: A 31-year-old pregnant woman was admitted complaining of left-flank tenderness, dysuria, and fever. RESULTS: Following examination, significant laboratory data were collected including increased leukocyte count (10,800/ul with 86% neutrophils) and C-reactive protein (9.6 mg/dl). Urinalysis revealed 30 to 50 leukocytes per high power field while from the quantitative urine culture Shigella sonnei was recovered after 24 h incubation at 37 degrees C. After a …

Multiplexed immunoassay to assess Shigella-specific antibody responses.

Infection with Shigella spp. results in bacillary dysentery and a systemic and mucosal antibody response. The immune response is directed at multiple antigens, including LPS and the invasin plasmid antigen (Ipa) proteins, and is capable of conferring short-term, serotype-specific protection. Both live-attenuated and several subunit vaccine approaches have focused on inducing a pronounced mucosal immune response directed to the same antigens recognized after natural infection. Traditionally, Shigella-specific antibody responses are measured using ELISA. Although ELISAs are robust immunological …

Reduced azithromycin susceptibility in Shigella sonnei, United States.

To investigate azithromycin susceptibility in Shigella sonnei in the United States, we examined the azithromycin minimum inhibitory concentrations (MICs) of outbreak and routine human S. sonnei isolates. Isolate susceptibility clustered at 8 mg/L, but three isolates displayed higher MICs (>64  mg/L) to azithromycin. All three isolates contained a plasmid-encoded mphA gene, known to encode a macrolide-2'-phosphotransferase enzyme. Transformation of the mphA gene into Escherichia coli DH10B allowed the transfer of decreased susceptibility to azithromycin. Although these isolates might traditiona…

Outbreak of Shigella sonnei gastroenteritis in Northeastern Taiwan.

A gastroenteritis outbreak of Shigella sonnei involving 122 children occurred between April 2001 and January 2002 in I-lan, Taiwan. The resistant rates of the isolates to nalidixic acid, trimethoprim-sulfamethoxazole, ampicillin and ceftriaxone were 98, 97.5, 32 and 27%, respectively. None of the isolates was resistant to ciprofloxacin. Prolonged bacterial shedding in the feces can occur when an inappropriate antibiotic is used.

Expression of His-tagged Shigella IpaC in Arabidopsis.

Although the expression of histidine (His)-tagged proteins in bacteria is routine, few His-tagged proteins have been expressed in plants, and no His-tagged proteins from bacterial pathogens have been expressed in plants, to our knowledge. Here, we demonstrate expression of the Shigella flexneri invasion plasmid antigen, IpaC, in Arabidopsis thaliana. S. flexneri is the causitive trigger for bacillary dysentery, and IpaC is essential for bacterial entry into epithelial cells. IpaC, attached to a 5' leader containing six tandem His codons, was cloned into a pBI121 vector. This clone was introduc…

Histone-like proteins and the Shigella invasivity regulon.

The contribution of histone-like proteins to the transcriptional regulation of virulence gene networks is a common feature among pathogenic bacteria. In this article we review current knowledge about the regulative role of major histone-like proteins in the silencing/activation of the invasivity phenotype of Shigella, the etiological agent of bacillary dissentery.

Subtyping of Shigella sonnei for tracing nosocomial transmission.

Nosocomial transfer of Shigella spp. occurred between two patients sharing the patient room and toilet in a ward for internal medicine. Both patients had travelled abroad shortly before the onset of illness. Based on the epidemiology of Shigella sonnei strains imported into Finland, nosocomial infection was strongly suspected. The isolates of S. sonnei were subtyped using ribotyping, plasmid analysis and antibiotic resistance patterns. The greatest discrimination between the strains was achieved by ribotyping. The results of the typing scheme confirmed the nosocomial transmission of S. sonnei.…

Rectal histopathology in endemic Shigella and Salmonella diarrhea.

Rectal histopathology was evaluated in 34 cases (2 months-12 yrs old) of endemic "invasive diarrhea" [> 20 WBCs per high-power field on stool microscopy with (RBC positive) or without (RBC negative) associated RBCs] where S. dysenteriae (n = 9), S. flexneri (n = 11), and nontyphoidal Salmonella were isolated as the sole identifiable enteropathogens. Persistent diarrhea (> 14 days duration) was more common with Salmonella infection whereas RBC-positive "invasive diarrhea" was more frequent with Shigella, particularly S. dysenteriae (all cases) infection. The histopathological profile was compar…

[Molecular and cellular bases of Shigella flexneri virulence].

Shigella flexneri, a Gram negative bacillus, causes bacillary dysentery, an ulcerative disease of the human colon, by invading intestinal epithelial cells. Entry into epithelial cells occurs via an induced phagocytic process which involves the actino-myosin complex. The host-cell receptor and the transmembrane signal which initiate reorganization of the cytoskeleton are under study. Binding to integrins has recently been demonstrated in related models such as the entry of Yersinia pseudotuberculosis and Bordetella pertussis into cells. Bacterial genes necessary to achieve entry are located on …

Detection of Shigella in feces using DNA amplification.

A rapid diagnostic method employing a polymerase chain reaction procedure (PCR) was used to identify Shigella and enteroinvasive Escherichia coli. This procedure amplified a region of the invasive-associated locus (ial) from a crude DNA extract of feces. A synthetic 21-base oligonucleotide corresponding to the ial gene sequence was shown to specifically hybridize only with enteroinvasive E. coli (EIEC) strains and Shigella species. Upon PCR amplification, a 320-base pair fragment was generated in DNA extracted from feces reconstituted with EIEC or Shigella flexneri but not in DNA from 70 norma…
