Osmotic stress inhibits osteoblast differentiation and mineralization by suppressing TRPV4-mediated calcium influx.

INTRODUCTION: Hyperglycemia increases the risk of bone fragility by promoting reactive oxygen species and advanced glycation end products, which disrupt osteoblast activity. Mechanical stress, including osmotic stress from elevated glucose levels, affects bone homeostasis; however, the specific impact of osmotic stress on osteoblast function is not fully understood. The transient receptor potential vanilloid 4 (TRPV4) channel, known to mediate calcium influx in response to mechanical stress, plays a key role in osteoblast differentiation. This study investigated the effects of osmotic stress o

Development of a thermal stabilizer formulation optimized by response surface methodology for Senecavirus A antigen.

Numerous members of the family Picornaviridae, such as the Senecavirus A (SVA) and foot-and-mouth disease virus (FMDV), exhibit thermal instability, resulting in the dissociation of viral particles, which affects the insufficient potency of the vaccine. Based on this characteristic, this study aimed to maintain the thermal stability of SVA by supplementing it with a stabilizer. Excipients, such as sucrose, mannitol, sorbitol, polyethylene glycol (PEG), L-arginine (L-Arg), glutamic acid (Glu), polyvinyl pyrrolidone (PVP), bovine serum albumin (BSA), and potassium chloride (KCl) dissolved in Tri

Paired metabolomics and volatilomics provides insight into transient high light stress response mechanisms of the coral Montipora mollis.

The coral holobiont is underpinned by complex metabolic exchanges between different symbiotic partners, which are impacted by environmental stressors. The chemical diversity of the compounds produced by the holobiont is high and includes primary and secondary metabolites, as well as volatiles. However, metabolites and volatiles have only been characterised in isolation so far. Here, we applied a paired metabolomic-volatilomic approach to characterise holistically the chemical response of the holobiont under stress. Montipora mollis fragments were subjected to high-light stress (8-fold higher t

Evolution of vancomycin-resistant Enterococcus faecium during colonization and infection in immunocompromised pediatric patients.

Patients with hematological malignancies or undergoing hematopoietic stem cell transplantation are vulnerable to colonization and infection with multidrug-resistant organisms, including vancomycin-resistant Enterococcus faecium (VREfm). Over a 10-y period, we collected and sequenced the genomes of 110 VREfm isolates from gastrointestinal and blood cultures of 24 pediatric patients undergoing chemotherapy or hematopoietic stem cell transplantation for hematological malignancy at St. Jude Children's Research Hospital. We used patient-specific reference genomes to identify variants that arose ove

Elevated Fructose and Uric Acid Through Aldose Reductase Contribute to Experimental and Human Alcoholic Liver Disease.

BACKGROUND AND AIMS: Alcohol-associated liver disease (ALD) is a common chronic liver disease worldwide with high morbidity and mortality, and no Food and Drug Administration-approved therapies. Fructose (dietary or endogenous), its metabolite uric acid, and aldose reductase (AR, the only endogenous enzyme that produces fructose) are strongly associated with the development of nonalcoholic fatty liver disease. However, the role of AR or its metabolites in ALD remains understudied and was examined using human specimens, cultured cells, and mouse model systems. APPROACH AND RESULTS: We demonstr

Polyphenols of marine red macroalga Symphyocladia latiuscula ameliorate diabetic peripheral neuropathy in experimental animals.

AIMS: Chronic hyperglycaemia activates the polyol pathway of glucose metabolism thereby stimulating the activation aldose reductase enzyme that in turn initiates a cascade of deleterious events, eventually, leading to nerve damage or neuropathy. Marine macroalgae and their isolated chemical constituents have been found to possess potential antidiabetic activity and have proved beneficial in the treatment of diabetes. In this study the neuroprotective effect of polyphenols isolated from the red macroalga Symphyocladia latiuscula was evaluated in experimental diabetic peripheral neuropathy. MAI

s-Allyl cysteine, s-ethyl cysteine, and s-propyl cysteine alleviate β-amyloid, glycative, and oxidative injury in brain of mice treated by D-galactose.

The neuroprotective effects of s-allyl cysteine, s-ethyl cysteine, and s-propyl cysteine in D-galactose (DG)-treated mice were examined. DG treatment increased the formation of Aβ(1-40) and Aβ(1-42), enhanced mRNA expression of β-amyloid precursor protein (APP) and β-site APP cleavage enzyme 1 (BACE1), and reduced neprilysin expression in brain (P < 0.05); however, the intake of three test compounds significantly decreased the production of Aβ(1-40) and Aβ(1-42) and suppressed the expression of APP and BACE1 (P < 0.05). DG treatments declined brain protein kinase C (PKC) activity and mRNA expr

Isolation of transcriptomal changes attributable to LHON mutations and the cybridization process.

Leber's hereditary optic neuropathy (LHON) is thought to be the most common disease resulting from mitochondrial DNA (mtDNA) point mutations, and transmitochondrial cytoplasmic hybrid (cybrid) cell lines are the most frequently used model for understanding the pathogenesis of mitochondrial disorders. We have used oligonucleotide microarrays and a novel study design based on shared transcripts to allocate transcriptomal changes into rho-zero-dependent, cybridization-dependent and LHON-dependent categories in these cells. The analysis indicates that the rho-zero process has the largest transcrip

Study of local anaesthetics. Part 139. Preformulation study of N-[2-(2-heptyloxyphenylcarbamoyloxy)-ethyl] dimethylammonium chloride.

In the course of the drug form development of substance XIX B (N-[2-(2-heptyloxyphenylcarbamoyloxy)-ethyl] dimethylammonium chloride [1]) the influence of auxiliary substance, namely glycerol (GL), propylene glycol (PG) and sorbitol (SO), used in the form of 5, 10, 15 and 20% solution, on the surface tension, partition coefficient, viscosity and liberation from aqueous solutions was studied. These parameters can influence the bioavailability. The obtained results show that as an optimal additive 5% SO may be used since this ensured maximal amount of liberated substance XIX B.

Reorganization of a synthetic microbial consortium for one-step vitamin C fermentation.

BACKGROUND: In the industry, the conventional two-step fermentation method was used to produce 2-keto-L-gulonic acid (2-KGA), the precursor of vitamin C, by three strains, namely, Gluconobacter oxydans, Bacillus spp. and Ketogulonicigenium vulgare. Despite its high production efficiency, the long incubation period and an additional second sterilization process inhibit the further development. Therefore, we aimed to reorganize a synthetic consortium of G. oxydans and K. vulgare for one-step fermentation of 2-KGA and enhance the symbiotic interaction between microorganisms to perform better. Carbohydrate malabsorption in clinical routine: a prospective observational study.

BACKGROUND/AIMS: Nonspecific abdominal symptoms are a serious problem throughout the world. Among the multitude of differential diagnoses in carbohydrate malabsorption, only incomplete absorption of lactose is mentioned, while malabsorption of fructose and sorbitol--which occurs much more often, at least in the Western world--is usually not included. METHODOLOGY: During a 6-month period, all patients (n=90; 33 males, median age 45 years, range 10-81; 57 females, median age 47 years, range 15-71) who consecutively presented for H2 exhalation tests were evaluated. Mixing of the intestinal content and variations of fermentation capacity do not affect the results of hydrogen breath test.

OBJECTIVE: Although the hydrogen (H(2)) breath test has been in use for many years for diagnosis of sugar malabsorption, research is still underway to improve its diagnostic accuracy. In this study, we investigated whether possible confusing factors caused by the ingestion of the test solution itself (such as the delivery to the colon of other fermentable substrates pre-existing in the small bowel lumen, the release of preformed H(2) trapped in the feces, or differences in the fermenting capacity of the colonic bacteria) may interfere with the increase of breath H(2) concentration, an expressi

Choline chloride deep eutectic solvents as plasticizers for thermoplastic starch adhesives for wood bonding: thermal stability, melt viscoelasticity, and lap-shear performance.

Regulatory and environmental pressure on petroleum-derived polymers, including those used as thermoplastic and hot-melt adhesives, is driving demand for renewable alternatives across multiple applications, such as non-structural wood adhesives. Thermoplastic starch (TPS) is a promising candidate, derived from an abundant, biodegradable polysaccharide, but how plasticizer chemistry, particularly chloride-based deep eutectic solvents (DESs), governs its adhesive performance on wood remains poorly understood. Eight TPS adhesive films were prepared from native corn starch by twin-screw extrusion a

Radial branch hydraulics and cellular injury thresholds in poplar: insights from controlled dehydration and high-resolution dendrometry.

Branch water release and radial exchange influence drought responses. Although mechanistic models describe radial hydraulic and storage dynamics, experimental quantification of hydraulic capacitance (C) and radial resistance (Rr) across broad ranges of xylem water-potential (Ψx), particularly under severe dehydration, remains limited. We quantified how C and Rr vary with Ψx in Populus albaL., assessed the value of coupling controlled osmotic dehydration with high-resolution dendrometry, and evaluated cellular damage and bud viability across increasing dehydration levels. Branch segments were s

Regulatory roles of Smt3, an ortholog of small ubiquitin-like modifier, in fungal development and the production of amylases and organic acids in Aspergillus niger.

The small ubiquitin-like modifier (SUMO) is pivotal to SUMOylation and exerts diverse roles in filamentous fungi, yet its roles in the production of carbohydrate-active enzymes (CAZymes) and organic acids remain unclear and few studies have addressed its functions in Aspergillus niger, a filamentous fungus with dual roles in food fermentation and food spoilage. This study identified a single SUMO homologue (Smt3) in Pu-erh tea-isolated A. niger RAF106 and characterized its functions via multi-phenotypic and transcriptional analyses. smt3 deletion caused defects in fungal growth, correlating wi

Rhodanine-3-acetamide derivatives as aldose and aldehyde reductase inhibitors to treat diabetic complications: synthesis, biological evaluation, molecular docking and simulation studies.

In diabetes, increased accumulation of sorbitol has been associated with diabetic complications through polyol pathway. Aldose reductase (AR) is one of the key factors involved in reduction of glucose to sorbitol, thereby its inhibition is important for the management of diabetic complications. In the present study, a series of seven 4-oxo-2-thioxo-1,3-thiazolidin-3-yl acetamide derivatives 3(a-g) were synthesized by the reaction of 5-(4-hydroxy-3-methoxybenzylidene)-4-oxo-2-thioxo-1,3-thiazolidin-3-yl acetic acid (2a) and 5-(4-methoxybenzylidene)-4-oxo-2-thioxo-1,3-thiazolidin-3-yl acetic aci

A unifying model of glucotoxicity in human renal proximal tubular epithelial cells and the effect of the SGLT2 inhibitor dapagliflozin.

BACKGROUND: Glucotoxicity in renal tubular epithelial cells (RPTECs) contributes to the pathogenesis of diabetic nephropathy. Sodium-glucose cotransporter 2 (SGLT2) inhibitors may exert their renoprotective effect by preventing glucotoxicity. We tested whether the confirmed in capillary endothelial cells unifying model of glucotoxicity can be applied in RPTECs and the impact of dapagliflozin. METHODS: In primary human RPTECs cultured in normal or high glucose medium in the presence or not of dapagliflozin, we assessed glucose consumption, SCLT2 expression, reactive oxygen species (ROS) produc
