Clonidine as a sensitizing agent in the forced swimming test for revealing antidepressant activity.

The forced swimming test (FST) in mice has failed to predict antidepressant activity for drugs having beta adrenoreceptor agonist activity a … specific because it did not appear with drugs other than antidepressants (diazepam, chlorpromazine, sulpiride, atropine), except for amphetamine and apomorphine which have a strong effect on the dopaminergic system. The use of behaviorally subactive doses of clonidine may thus provide an important means of increasing the sensitivity of the forced swimming t …

Spontaneous eye-blink rates and dopaminergic systems.

A series of studies demonstrated a possible relationship between eye-blink rate and central dopamine activity. First, apomorphine and other dopamine agonists acutely increased blink rate in monkeys, an effect blocked by sulpiride. Secondly, parkinsonian patients with levodopa-induced dyskinesia exhibited twice the mean blink rate (21 blinks/min) of other parkinsonians (11 blinks/min, P less than 0.002) whereas the more symptomatic of the nondyskinetic patients had a very slow rate (3 blinks/min, P less than 0.01). Thirdly, schizophrenic patients had an elevated mean blink (31 vs 23 blinks/min …

Consensus on the use of substituted benzamides in psychiatric patients.

The class of substituted benzamides includes compounds able to modulate dopaminergic neurons selectively and specifically. The first synthetic substituted benzamide was sulpiride, which has been replaced in the clinic by the more modern amisulpride. The compound is very selective for mesolimbic D2 and D3 receptors and, therefore, has a dual mechanism of action, which is associated with two different indications. At low doses (50 mg), amisulpride preferentially blocks presynaptic autoreceptors, producing an increase in dopamine release, and therefore acting as a dopaminergic compound able to re…

[Parkinson syndrome induced by veralipride].

Veralipride, an antidopaminergic drug commonly prescribed to counteract postmenopausal symptoms, may cause reversible parkinsonism. A 49-year-old healthy woman reported a 5 month history of progressive slowing of voluntary movements, postural changes, e.g. sit-to-stand manouevres, and gait. Clinical examination revealed moderate hypokinesia, hypomimia and plastic rigidity of the four limbs. Locomotor oscillations of the upper limbs were decreased. She had been taking 100 mg of veralipride daily to counteract postmenopausal symptoms (vasomotor flushes and irritability) for 17 months uninterrupt

[Blood prolactin in 845 cases of clinically idiopathic sexual dysfunction].

Serum prolactin levels were measured in 845 subjects presenting with clinically idiopathic sexual dysfunction, including impotence, premature or defective ejaculation and frigidity in women. Stimulation tests using thyrotrophin-releasing hormone (TRH) and sulpiride were performed in 80 men. One p. cent of impotent subjects had high serum prolactin levels associated with pituitary adenoma in 4/5 cases; 2.2% of impotent men, 10% of men with premature ejaculation and 5.6% of frigid women had moderate hyperprolactinaemia (50 ng/ml), probably not responsible for the sexual disorder. Mean serum prol…

[The treatment of hot flashes due to menopause. Comparative study of two drugs (author's transl)].

The efficacy of veralipride, a new non-hormonal treatment for the treatment of menopausal symptoms, was evaluated following a randomised comparative study with a commonly used non-hormonal drug, bêta-alanine. Two groups of patients between 53 and 59 years received, one veralipride (22) at a dose of 100 mg/day for 20 days, the other beta-alanine (21) during the same period. 14 patients noted an improvement in their hot flushes while receiving veralipride as against 4 receiving beta-alanine. The superiority of veralipride was quite significant (P less than 0.01). Both products were well tolerate

Clinical manifestations of tardive truncal dystonia--abdominal movements: report of two cases.

A variety of involuntary abdominal movements may result from peripheral insults or have a central origin. However, the spectrum of differential diagnoses can be broadened by this report of two patients whose involuntary abdominal movements were related to chronic use of haloperidol or sulpiride. Electromyographic studies revealed two patterns of muscle activity in these two patients. The first patient showed long-duration bursts of the thoracic and lumbar paraspinal muscles, causing repetitive backward tilting of the pelvis and downward shifting of the umbilicus. The second patient showed pers…

The D2 dopamine receptor agonist LY171555 induces catalepsy in the mouse.

The dopamine agonist LY171555 (quinpirole), a specific D2 receptor agonist, induces catalepsy in mice at doses ranging from 0.3 to 10 mg/kg. The effects of an intermediate dose of this compound (1 mg/kg SC) were antagonized by 25 mg/kg of the selective D2 antagonist (-)-sulpiride (IP) injected 20 min before LY171555. SCH 23390, a selective D1 antagonist, administered (0.3 mg/kg IP) 20 min before LY171555 (1 mg/kg) enhanced the cataleptic effects of this compound. Finally, when the D1 dopamine receptor agonist SKF 38393 (20 mg/kg SC) was administered immediately beforehand, the cataleptic effec…

Effect of repeated treatment with desipramine in the behavioral "despair" test in rats: antagonism by "atypical" but not "classical" neuroleptics or antiadrenergic drugs.

A 7-day treatment with 20 mg/kg/day desipramine reduced the immobility time in the behavioral "despair" test in rats. The effect of DMI was antagonized by sulpiride (100 mg/kg i.p.), metoclopramide (20 mg/kg i.p.) and clopazine (20 mg/kg i.p.) but not by haloperidol (0.5 mg/kg i.p.) or chlorpromazine (5 mg/kg i.p.). Alpha-adrenoreceptor blockers (prazosin 3 mg/kg s.c.; aceperone 10 mg/kg i.p.; azapetine 24 mg/kg s.c.; phentolamine 20 mg/kg i.p.), dl-propranolol (5 mg/kg i.p.) and clonidine (0.1 mg/kg i.p.) failed to modify the anti-immobility effect of DMI. The data suggest that a particular s…

Continuous stress promotes expression of VGF in melanotroph via suppression of dopamine.

Prolonged exposure to stress elicits profound effects on homeostasis that may lead to cryptogenic disorders such as chronic fatigue syndrome … altered in the IL of rats receivingthe dopamine D2 receptor agonist bromocriptine or the antagonist sulpiride. In vitro, dopamine dose-dependently decreased the mRNA levels in cultured melanotrophs. These findings suggest that VGF expression under CS is negatively regulated by dopaminergic neurons projecting from the hypothalamus.

Characterization of the action of antipsychotic subtypes on valproate-induced chromatin remodeling.

Recent advances in schizophrenia (SZ) research indicate that the telencephalic gamma-aminobutyric acid (GABA)ergic neurotransmission deficit … strongly indicate that VPA facilitates chromatin remodeling when it is associated with clozapine or sulpiride but not with haloperidol or olanzapine. This remodeling might contribute to reelin- and GAD(67)-promoter demethylation and might reverse the GABAergic-gene-expression downregulation associated with SZ morbidity.

The treatment of Tourette's syndrome: current opinions.

Correct education of the patient is one of the most important aspects in the treatment of Tourette's syndrome. Pharmacotherapy is often unsatisfactory and therefore should be limited to those patients who are significantly impaired. Therapy must be individualised and the most troublesome symptom should be targeted first. In the treatment of tics, dopamine receptor blocking agents are currently the most effective drugs. It is currently unknown whether classic neuroleptics, such as pimozide, selective dopamine receptor antagonists, such as sulpiride, or newer atypical antipsychotics, such as ris…

Effects of forskolin on pancreatic exocrine secretion and cyclic nucleotide concentrations of the dog pancreas.

The effects of forskolin on the secretion of pancreatic juice and on the intracellular levels of cyclic nucleotides were investigated in preparations of the isolated and blood-perfused dog pancreas. Forskolin (3-100 micrograms) injected intra-arterially caused a dose-dependent increase in the secretion of pancreatic juice. The concentration of bicarbonate in the pancreatic juice induced by forskolin was increased. However, protein concentration was decreased. Forskolin-induced secretion was not modified by pretreatment with atropine, phentolamine, propranolol, cimetidine, sulpiride and verapam…
