Well-Tempered Metadynamics Simulations Combined with Free Energy Landscape Analysis Uncover the Conformational Transition Pathway of the DYG Motif in LRRK2.

Leucine-rich repeat kinase 2 (LRRK2) is one of the most promising targets for the treatment of Parkinson's Disease. The activation of LRRK2 is regulated by the conserved Asp-Tyr-Gly (DYG) motif of the activation loop. Understanding the flipping process of the DYG motif from active to inactive is crucial for the regulation of LRRK2 kinase catalytic activity. Typically, type I inhibitors stabilize LRRK2 in the active conformation, while type II inhibitors favor the inactive state. To investigate the DYG-flipping mechanism, we performed well-tempered metadynamics simulations on LRRK2 in three dis

Non-targeted analysis and suspect screening of organic contaminants in temperate snowfall using liquid chromatography high-resolution mass spectrometry.

Contaminants released into the atmosphere that undergo regional and long-range transport can deposit back to Earth through snowfall. When snow melts, these contaminants re-enter the environment, sometimes far from their original emission sources. Here we present the first comprehensive characterization of organic contaminants in snow from North America. Fresh snowfall samples were collected in the central United States over a three-year period and measured by liquid chromatography high-resolution mass spectrometry for suspect screening and non-targeted analysis. The resulting data set was scre

Formation of mineral-associated organic matter in temperate soils is primarily controlled by mineral type and modified by land use and management intensity.

Formation of mineral-associated organic matter (MAOM) supports the accumulation and stabilization of carbon (C) in soil, and thus, is a key factor in the global C cycle. Little is known about the interplay of mineral type, land use and management intensity in MAOM formation, especially on subdecadal time scales. We exposed mineral containers with goethite or illite, the most abundant iron oxide and phyllosilicate clay in temperate soils, for 5 years in topsoils of 150 forest and 150 grassland sites in three regions across Germany. Results show that irrespective of land use and management inten

Distribution of Medically Relevant Antibiotic Resistance Genes and Mobile Genetic Elements in Soils of Temperate Forests and Grasslands Varying in Land Use.

Antibiotic-resistant pathogens claim the lives of thousands of people each year and are currently considered as one of the most serious threats to public health. Apart from clinical environments, soil ecosystems also represent a major source of antibiotic resistance determinants, which can potentially disseminate across distinct microbial habitats and be acquired by human pathogens via horizontal gene transfer. Therefore, it is of global importance to retrieve comprehensive information on environmental factors, contributing to an accumulation of antibiotic resistance genes and mobile genetic e

West Nile Virus Lineage 2 Vector Competence of Indigenous Culex and Aedes Mosquitoes from Germany at Temperate Climate Conditions.

West Nile virus (WNV) is a widespread zoonotic arbovirus and a threat to public health in Germany since its first emergence in 2018. It has become of particular relevance in Germany in 2019 due to its rapid geographical spread and the detection of the first human clinical cases. The susceptibility of indigenous Culex pipiens (biotypes pipiens and molestus) for a German WNV lineage 2 strain was experimentally compared to that of Serbian Cx. pipiens biotype molestus and invasive German Aedes albopictus. All tested populations proved to be competent laboratory vectors of WNV. Culex pipiens biotyp

Comparison of UVB effects on growth and induction of UVB screening compounds in isolates of metaphytic algae from temperate zone streams and ponds.

Filaments in the surface layers of metaphytic mats are exposed to high photon flux densities of PAR and UVBR. We investigated the effect of UVBR exposure on growth of eight isolates of common metaphytic algae (Cladophora, Mougeotia, Oedogonium, Pithophora, Spirogyra, and Zygnema) acclimated to either high or low PAR levels prior to UVBR exposure. All isolates acclimated to low PAR exhibited significant reductions in growth rate caused by the UVBR exposure (P < 0.05). Acclimation to high PAR resulted in seven of the isolates being more tolerant of the UVB exposure. The two Zygnema isolates exhi

Insights on the host associations and geographic distribution of Hymenolepis folkertsi (Cestoda: Hymenolepididae) among rodents across temperate latitudes of North America.

Synoptic data and an understanding of helminth parasite diversity among diverse rodent assemblages across temperate latitudes of North America remain remarkably incomplete. Renewed attention to comprehensive survey and inventory to establish the structure of biodiverse faunas is essential in providing indicators and proxies for identifying the outcomes of accelerating change linked to climate warming and anthropogenic forcing. Subsequent to the description of Hymenolepis folkertsi in the oldfield mouse, Peromyscus polionotus, additional specimens of hymenolepidids were collected or discovered

Comparative exploration of hydrogen sulfide and water transmembrane free energy surfaces via orthogonal space tempering free energy sampling.

Hydrogen sulfide (H2 S), a commonly known toxic gas compound, possesses unique chemical features that allow this small solute molecule to quickly diffuse through cell membranes. Taking advantage of the recent orthogonal space tempering (OST) method, we comparatively mapped the transmembrane free energy landscapes of H2 S and its structural analogue, water (H2 O), seeking to decipher the molecular determinants that govern their drastically different permeabilities. As revealed by our OST sampling results, in contrast to the highly polar water solute, hydrogen sulfide is evidently amphipathic, a

Onchocerca gutturosa and Onchocerca lienalis in cattle: effect of age, sex, and origin on prevalence of onchocerciasis in subtropical and temperate regions of Florida and Georgia.

Evaluation of 845 cattle from Florida and Georgia for onchocerciasis indicated a significantly higher prevalence of infection with Onchocerca gutturosa and a significantly lower prevalence of infection with Onchocerca lienalis in the southern portion of Florida (subtropical region) than in the northern portion of Florida or in Georgia (temperature region). In all regions, prevalence of infection with O gutturosa was significantly higher than infection with O lienalis. In the temperate region, bulls were infected more often with O lienalis than were cows. Other sex-related differences in infect

Pasteurella haemolytica bacteriophage: identification, partial characterization, and relationship of temperate bacteriophages from isolates of Pasteurella haemolytica (biotype A, serotype 1).

Pasteurella haemolytica (biotype A, serotype 1) isolates (n = 15) from the upper respiratory tract of clinically normal cattle, as well as from lung lesions from cases of fatal bovine pasteurellosis, were examined for the presence of bacteriophage after irradiation with UV light. Treatment of all P haemolytica isolates with UV irradiation resulted in lysis of bacteria due to the induction of vegetative development of bacteriophages. The extent of growth inhibition and bacterial lysis in irradiated cultures was UV dose-dependent. Bacterial cultures exposed to UV light for 20 s reached peak cult

Dissolved organic matter molecular composition links water chemistry, microbial functional potential, and nitrous oxide variability in a temperate estuary.

Estuaries receive substantial carbon and nitrogen inputs and are sources of nitrous oxide (N₂O), but salinity-driven conservative mixing can obscure biogeochemical links between DOM molecular composition and estuarine N₂O accumulation. We integrated ultrahigh-resolution DOM molecular characterization, high-throughput sequencing, and functional gene quantification to examine how water chemistry, DOM molecular composition, and microbial functional potential were associated with N₂O dynamics after accounting for conservative mixing in the Luanhe River Estuary, China. Estuarine waters were consist

Tailoring Low-Temperature Tempering to Dramatically Enhance Compressive Ductility and Fatigue Contact Wear Resistance in High-Carbon Bearing Steel.

High-carbon martensitic steels for bearing components are conventionally low-temperature tempered for stress relief, yet the influence of tempering temperature on compressive and fatigue wear resistance remains unclear, directly affecting the service life of bearing races and rollers. In this study, a high-carbon martensitic steel was tempered at 170 °C, 200 °C, and 230 °C. The microstructural evolution, compressive properties, and contact fatigue wear resistance were systematically investigated, along with the corresponding strengthening and wear mechanisms. After spheroidizing annealing and

Continuous chlorophyll fluorescence measurements trace sudden cold spell effects on the photochemical efficiency of photosystem II in a temperate mixed forest.

The increasing frequency of air temperature extremes and short-term fluctuations poses a significant threat to the physiological functioning of temperate forests across Europe. However, the mechanisms by which these fluctuations induce stress and impair the photochemical efficiency of photosystem II (PSII) in tall tree canopies remain poorly understood. Moreover, the impact of air temperature extremes and fluctuations on ecosystem carbon fluxes remains poorly quantified. Here, we integrated real-time chlorophyll fluorescence (ChlF) and eddy covariance carbon flux measurements in a temperate mi

Temperate bacteriophage SapYZUs7 alters Staphylococcus aureus fitness balance by regulating expression of phage resistance, virulence and antimicrobial resistance gene.

Temperate bacteriophages are crucial for maintaining the pathogenicity and fitness of S. aureus, which also show promise as a biocontrol agent for S. aureus. However, the fitness benefit and cost of lysogeny by S. aureus temperate phages and their underlying mechanisms remain unexplored. In this study, phage resistance, virulence, antimicrobial resistance (AMR), transcriptome, and metabolome of phage SapYZUs7 lysogenic and non-lysogenic S. aureus strains were compared. Whole-genome analysis revealed that SapYZUs7 harbouring smaII associated with a single-protein MazF-like antiphage system coul

Carbon dioxide removal (CDR) potential in temperate macroalgal forests: A comparative study of chemical and biological net ecosystem production (NEP).

The carbon dioxide removal (CDR) capacity of macroalgae, a crucial component in climate regulation, has gained increasing attention. However, accurately estimating the CDR potential of macroalgae in natural conditions remains challenging, necessitating the use of multiple independent methods to reduce the uncertainties in these estimates. In this study, we compared two methods for estimating net ecosystem production (NEP), a key parameter in determining CDR potential: 1) NEPChem., derived from seawater carbonate chemistry and 2) NEPBiol., based on photorespiratory measurements using benthic te

Sporadic diurnal fluctuations of cyanobacterial populations in oligotrophic temperate systems can prevent accurate characterization of change and risk in aquatic systems.

Cyanobacteria increasingly threaten recreational water use and drinking water resources globally. They require dynamic monitoring to account for variability in their distribution arising from diel cycles associated with oscillatory vertical migration. While this has been discussed in marine and eutrophic freshwater contexts, reports of diurnal vertical migration of cyanobacteria in oligotrophic freshwater lakes are scant. Typical monitoring protocols do not reflect these dynamics and frequently focus only on surface water sampling approaches, and either ignore sampling time or recommend large

Soil organic matter carbon chemistry signatures, hydrophobicity and humification index following land use change in temperate peat soils.

Peatlands play a critical role in the global carbon cycle, storing large amounts of carbon because of a net imbalance between primary production and the microbial decomposition of the organic matter. Nevertheless, peatlands have historically been drained for energy sources (e.g. peat briquettes), forestry, or agriculture - practices that could affect the quality of the soil organic matter (SOM) composition, hydrophobicity and humification index. This study compared the effect of land use change on the quality and composition of peatland organic material in Co-Offaly, Ireland. Specifically, dra

The white-footed deermouse, an infection-tolerant reservoir for several zoonotic agents, tempers interferon responses to endotoxin in comparison to the mouse and rat.

The white-footed deermouse Peromyscus leucopus, a long-lived rodent, is a key reservoir for agents of several zoonoses, including Lyme disease. While persistently infected, this deermouse is without apparent disability or diminished fitness. For a model for inflammation elicited by various pathogens, the endotoxin lipopolysaccharide (LPS) was used to compare genome-wide transcription in blood by P. leucopus, Mus musculus and Rattus norvegicus and adjusted for white cell concentrations. Deermice were distinguished from the mice and rats by LPS response profiles consistent with non-classical mon

Transmission of West Nile and five other temperate mosquito-borne viruses peaks at temperatures between 23°C and 26°C.

The temperature-dependence of many important mosquito-borne diseases has never been quantified. These relationships are critical for understanding current distributions and predicting future shifts from climate change. We used trait-based models to characterize temperature-dependent transmission of 10 vector-pathogen pairs of mosquitoes (Culex pipiens, Cx. quinquefascsiatus, Cx. tarsalis, and others) and viruses (West Nile, Eastern and Western Equine Encephalitis, St. Louis Encephalitis, Sindbis, and Rift Valley Fever viruses), most with substantial transmission in temperate regions. Plant community and litter composition in temperate deciduous woodlots along two field gradients of soil Ni, Cu and Co concentrations.

Perennial plant communities in the proximity of metal smelters and refineries may receive substantial inputs of base metal particulate as well as sulphate from the co-emission of sulphur dioxide. The Ni refinery at Port Colborne (Canada) operated by Inco (now Vale Canada Ltd.) emitted Ni, Co and Cu, along with sulphur dioxide, between 1918 and 1984. The objectives were to determine if vascular plant community composition, including standing litter, in twenty-one woodlots on clay or organic soil, were related to soil Ni concentration which decreased in concentration with distance from the Ni re
