Enhancing the chemical transformation of Candida parapsilosis.

Candida parapsilosis is a leading cause of invasive mycoses and the major cause of nosocomial fungaemia amongst low and very low birth weight neonates. However, the molecular and physiological characteristics of this fungus remain understudied. To advance our knowledge about the pathobiology of this pathogen, we sought to develop and validate an effective method for chemical transformation of C. parapsilosis. Chemical transformation is the primary procedure for introducing foreign DNA into Candida yeast as it requires no special equipment, although its performance efficacy drops rapidly when t

Conjugation of ϕBT1-derived integrative plasmid pDZL802 in Amycolatopsis mediterranei U32.

The genus Amycolatopsis is well known for its ability to produce antibiotics, and an increasing number of valuable biotechnological applications, such as bioremediation, biodegradation, bioconversion, and potentially biofuel, that use this genus have been developed. Amycolatopsis mediterranei is an industrial-scale producer of the important antibiotic rifamycin, which plays a vital role in antimycobacterial therapy. Genetic studies of Amycolatopsis species have progressed slowly due to the lack of efficient transformation methods and stable plasmid vectors. In A. mediterranei U32, electroporat

The transforming Streptococcus pneumoniae in the 21st century.

Streptococcus pneumoniae, an important pathogen causing sepsis, sinusitis, otitis media, bacterial meningitis and bacterial pneumonia, results in global morbidity and mortality each year. The burden of pneumococcal disease is highest in children and the elderly. Treatment of pneumococcal infection has been hampered by the complexity of the host immune response. In recent decades, the increase of S. pneumoniae strains' resistance to beta-lactam antibiotics and other classes of antimicrobials has made treatment even more complicated. Fortunately, the advent of heptavalent conjugate vaccine confe

Biologically-enhanced removal of PCE from NAPL source zones.

The influence of dechlorinating microorganisms on PCE and its reduction products in a residual nonaqueous phase liquid (NAPL) source zone was investigated. Experiments were conducted in upflow columns containing glass beads (diameters 500-750 microns) contaminated with a residual NAPL consisting of tridecane and labeled 14C-PCE. Three columns were inoculated with a mixed PCE-dechlorinating culture, that was fed electron donor (pyruvate) at concentrations of 25, 100, and 250 mM. Pyruvate was fermented in all columns with essentially no methanogenic activity. Comparisons between actively dechlor

Plasmid-mediated resistance to antibiotic synergism in enterococci.

Mating experiments have shown that high-level resistance (minimal inhibitory concentration greater than 2,000 microgram/ml) to streptomycin and kanamycin, and resistance to penicillin-streptomycin and penicillin-kanamycin synergism are transferable by conjugation from resistant clinical isolates of enterococci to a sensitive recipient strain. Cesium chloride-ethidium bromide ultracentrifugation revealed a satellite (plasmid) band in resistant clinical isolates and the transconjugant strains but not in the sensitive recipient. Examination of these satellite bands by agarose gel electrophoresis

Critical insights into antibiotic resistance transferability in probiotic Lactobacillus.

Probiotics are live microorganisms that, when administered in adequate amounts, confer a health benefit on the host, with respect to metabolism, immune function, and nutrition. Any perturbation of these beneficial microbes leads to gut dysbiosis, which triggers the development of various disorders in the gastrointestinal system. Probiotics play a key role in resolving the dysbiosis posed by external factors such as antibiotics, other substances, or interventions. Supplementing probiotics with antibiotics is favorable in reducing the harmful effects of antibiotics on gut flora. These microbes a

Lysine is more effective than arginine in two tryptophan-rich antimicrobial peptides.

We compare biophysical and microbiological results to link structure and function in determining the mechanism of bacterial killing by two antimicrobial peptides (AMPs). Two 14-mer peptides were compared, each containing 6 tryptophans, in addition to 8 lysines (W6K8) or 8 arginines (W6R8). MIC values show W6K8 is more effective at killing bacteria, and less toxic to red blood cells than is W6R8. Small angle X-ray scattering (SAXS) shows W6K8 is more efficient at fusing unilamellar vesicles (ULVs) that mimic bacterial lipid model membranes (LMMs), and W6R8 is more efficient at fusing ULVs that

Isolation and Characterization a Novel Catabolic Gene Cluster Involved in Chlorobenzene Degradation in Haloalkaliphilic Alcanivorax sp. HA03.

Chlorobenzene (CB) poses a serious risk to human health and the environment, and because of its low degradation rate by microorganisms, it persists in the environment. Some bacterial strains can use CB as growth substrates and their degradative pathways have evolved; very little is known about these pathways and the enzymes for CB degradation in high pH and salinity environments. Alcanivorax sp. HA03 was isolated from the extremely saline and alkaline site. HA03 has the capability to degrade benzene, toluene and chlorobenzene (CB). CB catabolic genes were isolated from HA03, which have a compl

Alteration of insulin signaling to control insect pest by using transformed bacteria expressing dsRNA.

BACKGROUND: Insulin/insulin-like growth factor signaling (IIS) is known to mediate larval growth and adult reproduction in the legume pod borer, Maruca vitrata (Lepidoptera: Crambidae). Four IIS components (InR, FOXO, Akt, and TOR) play crucial roles in the IIS pathway. RESULTS: RNA interference (RNAi) against any of these four IIS component genes was effective in suppressing each target mRNA level by either hemocoelic injection or oral administration using gene-specific double-stranded RNAs (dsRNAs). These RNAi treatments interfered with larval growth, leading to small pupae or significant l

Phospholipid synthesis in Borrelia burgdorferi: BB0249 and BB0721 encode functional phosphatidylcholine synthase and phosphatidylglycerolphosphate synthase proteins.

Phospholipids are an important component of bacterial membranes. Borrelia burgdorferi differs from many other bacteria in that it contains only two major membrane phospholipids: phosphatidylglycerol (PG) and phosphatidylcholine (PC). B. burgdorferi appears to lack enzymes required for synthesis of PC through the well-described methylation pathway. However, B. burgdorferi does contain a gene (BB0249) with significant identity to a recently described phosphatidylcholine synthase gene (pcs) of Sinorhizobium meliloti. B. burgdorferi also contains a gene (BB0721) with significant identity to the ge

Estrogenic, Genotoxic, and Antibacterial Effects of Chemicals from Cryogenically Milled Tire Tread.

Tire and road wear particles (TRWP) contain complex mixtures of chemicals and release them to the environment, and potential toxic effects of these chemicals still need to be characterized. We used a standardized surrogate for TRWP, cryogenically milled tire tread (CMTT), to isolate and evaluate effects of tire-associated chemicals. We examined organic chemical mixtures extracted and leached from CMTT for the toxicity endpoints genotoxicity, estrogenicity, and inhibition of bacterial luminescence. The bioassays were performed after chromatographic separation on high-performance thin-layer chro

Gaps in the wall: understanding cell wall biology to tackle amoxicillin resistance in Streptococcus pneumoniae.

Streptococcus pneumoniae is the most common cause of community-acquired pneumonia, and one of the main pathogens responsible for otitis media infections in children. Amoxicillin (AMX) is a broad-spectrum β-lactam antibiotic, used frequently for the treatment of bacterial respiratory tract infections. Here, we discuss the pneumococcal response to AMX, including the mode of action of AMX, the effects on autolysin regulation, and the evolution of resistance through natural transformation. We discuss current knowledge gaps in the synthesis and translocation of peptidoglycan and teichoic acids, maj

Triphenyl phosphate (TPHP) stress-induced bacterial community differentiation, succession and energy metabolism inhibition under distinct contamination histories.

Triphenyl phosphate (TPHP), a widely used organophosphate flame retardant, is increasingly detected in wastewater systems, landfill leachate-impacted environments, and receiving waters. However, the responses of microbial communities with different contamination histories to TPHP have remained poorly understood. Here, landfill soil and wastewater bacterial communities were subjected to a gradient of TPHP stress under controlled microcosm conditions to assess community, network, and functional responses. The results demonstrated that microbial responses to TPHP were strongly shaped by environme

Disruption of CFTR-dependent lipid rafts reduces bacterial levels and corneal disease in a murine model of Pseudomonas aeruginosa keratitis.

PURPOSE: Pseudomonas aeruginosa enters corneal epithelial cells in vitro via membrane microdomains or lipid rafts. Bacterial entry, mediated by the cystic fibrosis transmembrane conductance regulator (CFTR), promotes infection and disease. This study was conducted to determine whether P. aeruginosa and CFTR are colocalized to rafts in isogenic corneal cells expressing wild-type (WT) or mutant DeltaF508-CFTR and whether disruption of the rafts both in vitro and in vivo affects the bacterial levels and the course of the disease. METHODS: Transformed human corneal epithelial cells from a patient

A Pharmacodynamic Study of Aminoglycosides against Pathogenic E. coli through Monte Carlo Simulation.

This research focuses on combating the increasing problem of antimicrobial resistance, especially in Escherichia coli (E. coli), by assessing the efficacy of aminoglycosides. The study specifically addresses the challenge of developing new therapeutic approaches by integrating experimental data with mathematical modeling to better understand the action of aminoglycosides. It involves testing various antibiotics like streptomycin (SMN), kanamycin (KMN), gentamicin (GMN), tobramycin (TMN), and amikacin (AKN) against the O157:H7 strain of E. coli. The study employs a pharmacodynamics (PD) model t

Complementary Bacterial Functions Enhance Mineralization of Aromatic Aliphatic Copolyesters within a Marine Microbial Consortium.

Plastics are a major environmental concern due to their persistence in natural systems. Biodegradable plastics can mitigate this impact by reducing their residence time in the environment. To constrain the environmental lifetime of these materials, understanding the fundamental principles dictating their biodegradation is crucial. The work presented here probes this complexity by using a 30-member bacterial community enriched from the marine ecosystem to investigate how bacterial consortia mineralize polybutylene sebacate-co-terephthalate (PBSeT), a biodegradable aromatic aliphatic copolyester

Histopathology of the early phase during experimental Corynebacterium pseudotuberculosis infection in lambs.

Histological responses during experimental Corynebacterium pseudotuberculosis infection in lambs were investigated in parotid lymph nodes for ten days following inoculation. Lambs were infected by the subcutaneous route into the right eyelid with a virulent strain of C. pseudotuberculosis. Multiple microscopic acute abscesses, predominantly infiltrated with polymorphonuclear (PMN) leucocytes, were seen in the right parotid lymph node on the 1st day post-inoculation (PI). This massive PMN infiltration coincided with a peripheral blood granulocytosis. On day 3 PI, an influx of histiocytes was ob
