Development of vascularized adipose organoids from PBMSCs and their application in evaluating Celastrol's effects.

Obesity lacks physiologically relevant human models, which constrains in-depth investigation into its underlying mechanisms and hampers the development of effective therapeutic strategies for obesity and its associated comorbidities. Here, we developed vascularized adipose organoids (VAOs) from peripheral blood mesenchymal stem cells (PBMSCs) using a standardized three-dimensional dynamic culture system that sequentially induces angiogenesis and adipogenesis. These VAOs mimic native adipose tissue with coexisting adipocytes and endothelial cells. Transcriptomic analysis shows that vascularizat

Local and Systemic Effects of Topical Betulinic Acid in a Psoriasis-like Inflammation Model in Mice.

Psoriasis patients often discontinue oral and injectable treatments due to concerns about both safety and efficacy. Issues such as adverse side effects, limited long-term effectiveness, and fear of potential complications contribute to non-adherence, impacting treatment outcomes and patients' quality of life. Betulinic acid (BA) forms supramolecular aggregates through self-assembly in a hydroalcoholic vehicle, which was hypothesized to have antipsoriatic activity when applied topically. To test this, imiquimod was applied to induce psoriasis-like skin inflammation in mice (except in the untrea

Microbial transformation of cycloastragenol by Penicillium spinulosum AS3.149.

Two new metabolites, 29-demethyl-3-ene-cycloastragenol (2) and 1α,12α-dihydroxy-cycloastragenone (3), along with six known compounds (4-9) were obtained from the microbial transformation of cycloastragenol (1) by Penicillium spinulosum AS3.149. The structures of these metabolites were determined by extensive spectroscopic (IR, UV, HRESIMS, 1D and 2D NMR) data analyses. Compound 2 features a 29-nor-triterpenic skeleton, possibly formed through sequential oxidation, decarboxylation, and dehydration reactions. Biological assays revealed that compound 6 exhibited significant cytotoxic activity aga

Defense of winter-dormant Alaska paper birch against snowshoe hares.

Mature growth-phase internodes of Alaska paper birch (Betula resinifera) are preferred by the snowshoe hare (Lepus americanus) over juvenile growth-phase internodes due to the low food value of the latter. While the mature over juvenile preferencec cannot be explained by the levels of inorganic nutrients or gross chemical fractions (resins or phenols), it can be explained by the striking differences in secondary metabolites of the two growth phases. The principle compound which renders the juvenile phase internodes unpalatable is papyriferic acid, a triterpene which is a demonstrated feeding d

Phytosterols: Potential Therapeutic Effects and Challenges in Food Industry.

Increases in serum total and low-density lipoprotein (LDL) cholesterol are known as hypercholesterolemia, and it is a significant risk factor for the emergence of cardiovascular illnesses. Any action strategy for lowering serum cholesterol is supported by lifestyle changes. Phytosterols are organic substances from the triterpene family. Phytosterols can lower serum LDL cholesterol levels because of their structural resemblance to cholesterol. Phytosterols are used to enrich or fortify a broad spectrum of food products. Phytosterols are quickly oxidized, just like cholesterol and unsaturated fa

Anti-ulcerogenic effects of Spartium junceum flowers on in vivo test models in rats.

Flowers of Spartium junceum L. (Fabaceae) are used for the treatment of gastric ulcers in Turkish folk medicine. Through bioassay-guided fractionation using chemical and chromatographical means and water immersion and restraint-induced stress ulcer model in rats, a saponin fraction was determined as the potent anti-ulcerogenic ingredient. The active fraction was also highly effective in preventing ethanol- and pyloric ligation-induced gastric lesions as well as inhibiting gastric secretion volume, gastric pH and titratable acidity, but did not affect the hexosamine content of the gastric mucos

Current ideas on the pathophysiology and treatment of lantana poisoning of ruminants.

Lantana poisoning in ruminants results from the ingestion of toxic varieties of the plant Lantana camara, which contain the triterpene acids lantadene A and lantadene B. Poisoning results in intrahepatic cholestasis and the consequences of the liver injury include jaundice, photosensitisation and ruminal stasis. The decreased ruminal motility causes toxic material to be retained in the rumen and continuous absorption of toxins from the rumen maintains the disease. Lantana poisoning can best be treated by preventing further absorption of toxins. This can be achieved by giving a large quantity o

Acute Toxicological Evaluation in Alternative Method Galleria mellonella and Murine Models of the Betulinic Acid Derivatives.

Betulinic acid (BA) has been described with several therapeutic properties, including anticancer, anti-inflammatory, antibacterial, antiviral, antidiabetic, antimalarial, and anti-HIV activities. Unfortunately, the long-term use of BA can cause mild toxicity in users, which may be solved by modifying its chemical structure to obtain BA derivatives. As proposed in the present study, the influence on the toxicity profile of the modification at carbon-3 of BA was evaluated by the replacement of the hydroxyl group with oxime and amine, resulting in derivatives with good yields. These derivatives w

Boswellic Acid and Betulinic Acid Pre-treatments Can Prevent the Nephrotoxicity Caused by Cyclophosphamide Induction.

Cyclophosphamide (CYP) is a chemotherapeutic drug used to treat various cancers. However, its clinical use is limited due to severe organ damage, particularly to the kidneys. While several phytochemicals have been identified as potential therapeutic targets for CYP nephrotoxicity, the nephroprotective effects of boswellic acid (BOSW) and betulinic acid (BET) have not yet been investigated. Our study used 42 rats divided into six equal groups. The study included six groups: control, CYP (200 mg/kg), CYP+BOSW20 (20 mg/kg), CYP+BOSW40 (40 mg/kg), CYP+BET20 (20 mg/kg), and CYP+BET40 (40 mg/kg). Th
