Identification of OTUD7a downregulation and global ubiquitination remodeling in a murine asthma model.

Airway epithelial dysfunction is a hallmark of asthma, contributing to chronic inflammation and structural remodeling. Recent research sugge … itinome profiling in ovalbumin (OVA)-induced asthmatic lungs revealed widespread changes in protein ubiquitination, with more than 3000 differentially modified ubiquitination sites. These changes were enriched in pathways associated with immune signaling, cytoskeletal dynamics, and epithelial barrier function. Collectively, our findings suggest that OTUD7a, as a …

PRMT1-mediated methylation controls OFIP ubiquitination and centrosome duplication to maintain epidermal homeostasis.

Epidermal homeostasis relies on the precise balance between the self-renewal and differentiation of basal keratinocytes, a process highly de … ting protein (OFIP) at arginine 50. This methylation is required for the subsequent recognition and ubiquitination of OFIP by the E3 ubiquitin ligase STUB1. Consequently, PRMT1 deficiency prevents OFIP methylation and degradation, resulting in its dramatic accumulation at the centrosome. This buildup drives aberrant centrosome overduplication and the formation o …

Protein tyrosine kinase-6 is post-translationally regulated by arginine methylation and ubiquitination.

Protein tyrosine kinase 6 (PTK6) has context-specific, kinase-dependent and -independent functions in normal cells and cancer. In prostate cells, nuclear PTK6 suppresses growth, while active membrane-associated PTK6 promotes oncogenic signaling. Unlike SRC-family kinases, PTK6 is not modified by lipidation to promote membrane association. Although arginine residues in the PTK6 SH2 domain were previously linked to lipid binding, we found they are instead critical for protein stability and arginine methylation. Mutation of R131/R136 or R132 impaired PTK6 ubiquitination and its association with a…

Liquid-liquid phase separation couples MKRN2-mediated ubiquitination of CSDE1 with neurodevelopmental disorders.

BACKGROUND: Makorin-2 (MKRN2) is an E3 ubiquitin ligase involved in multiple biological processes, yet its role in neurological disorders remains poorly understood. This study aims to elucidate how MKRN2 regulates the RNA-binding protein CSDE1-a molecule linked to autism-related genes-and to explore the functional implications of this interaction in neurodevelopment. METHODS: Using mass-spectrometry screening, we identified CSDE1 as a direct substrate of MKRN2. Ubiquitination sites were validated through mutagenesis of conserved lysine residues. Liquid-liquid phase separation (LLPS) assays we…

TRIM21 Neutralizes Hazara Virus Using a Dual Mechanism of Nucleoprotein Caging and Ubiquitination.

Crimean-Congo Hemorrhagic Fever Virus (CCHFV) is a tick-borne bunyavirus with widespread and growing geographic distribution that causes sev … echanism in which NP cross-linking by TRIM21 physically interferes with NP function whilst parallel ubiquitination labels the protein for degradation. This dual mechanism is similar to that used by TRIM5 against retroviruses and suggests that antiviral TRIMs may utilize their capacity for self-assembly both for catalytic activation and viral caging.

Targeting muscle wasting: Bioinformatics-derived ubiquitination genes as potential therapeutic targets for sarcopenia.

Sarcopenia is a major health concern characterized by progressive loss of muscle mass and function among the elderly. Its prevalence ranges from 10% to 27% in individuals over 60 and increases further in those above 80. The condition reduces mobility and strength, increasing risks of falls, fractures, and other age-related health issues. The ubiquitin-proteasome system (UPS) plays a central role in muscle protein degradation and contribute to muscle wasting. However, therapeutic strategies targeting this pathway remain poorly understood. This study aimed to identify ubiquitination-related gene…

Comprehensive Lysine Ubiquitome Profiling Reveals Diverse Functions of Lysine Ubiquitination in Eimeria tenella.

Eimeria tenella is a parasitic protozoan with a direct lifecycle in a single host, but with diverse intermediate stages while developing in the host. Each E. tenella developmental stage has distinct structural, pathogenic, genetic, and protein expression, emerging from gene modifications and post-translational modifications (PTMs) of proteins. Ubiquitination regulates diverse cellular functions and activities. Until now, however, there have been sparse data on E. tenella ubiquitination except on some ubiquitin-related enzymes. It is pertinent to unveil the mechanism through which E. tenella de…

UFMylation of SHBs increases protein stability by antagonizing ubiquitination and promotes virion production.

UNLABELLED: Post-translational modifications orchestrate virus-host interactions, yet their roles in the hepatitis B virus (HBV) life cycle remain incompletely defined. The small hepatitis B surface antigen (SHBs)-the most abundant HBV protein-drives immune evasion, pathogenesis, and virion/subviral particle (SVP) production. Here, we identify UFL1, the E3 ligase of UFMylation, as a host factor that modifies and stabilizes SHBs by antagonizing its ubiquitination to potentiate HBV output and SHBs-driven pro-oncogenic activities. Using affinity purification-mass spectrometry, we discovered UFL1 …

RNF138-Mediated Ubiquitination and Degradation of NS5 Restricts Tick-Borne Encephalitis Virus Infection.

Tick-borne encephalitis virus (TBEV) causes severe neurological disease. However, whether host restricts its replication and the underlying mechanisms remain incompletely delineated. Here, we identify the E3 ubiquitin ligase RNF138 as an intrinsic restriction factor targeting the viral RNA-dependent RNA polymerase (RdRp) NS5. Mechanistically, RNF138 directly interacts with the RdRp domain of NS5 through its ubiquitin-interacting motif, catalyzes K48-linked polyubiquitination and induces degradation of TBEV NS5. Ubiquitin-remnant profiling and mutational analyses identify K372, K462, and K470 w…

CLIC1 regulates cellular oxidative stress and gastric cancer progress by enhancing ACOX1 ubiquitination.

Oxidative stress is a two-edged sword for tumors. However, how oxidative stress influences tumor progression remains unclear. We determined that chloride intracellular channel 1 (CLIC1) accelerates gastric cancer progression by enhancing acyl-CoA oxidase 1 (ACOX1) ubiquitination and increasing cellular oxidative stress. CLIC1 overexpression increased cellular oxidative stress. Furthermore, our results demonstrate that ACOX1 is an oxidative stress-related gene that is closely related to CLIC1. Functionally, downregulating ACOX1 promoted gastric cancer cell proliferation, migration, and invasion…

KLHL8-mediated ubiquitination and TAX1BP1-dependent autophagic degradation of GPX4 drive neuronal ferroptosis.

Ferroptosis, characterized by iron-dependent lipid peroxidation, is considered a key cell death pathway activated during ischemic stroke. GPX4, a negative regulator of ferroptosis, exacerbates brain damage. However, the precise regulatory mechanisms governing this process remain poorly understood. Here, we determined that oxygen-glucose deprivation/reoxygenation (OGD/R)-induced GPX4 degradation is primarily dependent on autophagy activation. Mechanistically, the E3 ubiquitin ligase KLHL8 tags GPX4 through ubiquitination and promotes its binding to the selective autophagy receptor TAX1BP1, ther…

Divergent Mechanisms of H2AZ.1 and H2AZ.2 in PRC1-Mediated H2A Ubiquitination.

The histone H2A variant H2AZ plays pivotal roles in shaping chromatin architecture and regulating gene expression. We recently identified H2AZ.2 in histone H2A lysine 119 ubiquitination (H2AK119ub)-enriched nucleosomes, but it is not known whether its highly related isoform H2AZ.1 also regulates this modification. In this study, we employed isoform-specific epitope-tagged knock-in mouse embryonic stem cell (ESC) lines to dissect the roles of each isoform in Polycomb Repressive Complex 1 (PRC1)-mediated H2AK119ub. Our results show that H2AZ.1 and H2AZ.2 share highly overlapping genomic binding …
