The effect on amebal virulence of Escherichia coli, Clostridium symbiosum and a mixture of lactobacilli was investigated. Amebas from HM1 and HK axenic strains were incubated with a single bacterial strain or lactobacilli for short, intermediate, or long periods and analyzed for their erythrophagocytosis, hemolytic activity and cytotoxicity in vitro and hepatic abscesses formation in vivo. It was found that in vitro virulence was enhanced by all studied bacteria in HK9 but not in HM1 strains, which only showed an increase in cytotoxicity. The virulence in vivo was solely increased in trophozoi…

Shigella flexneri, a Gram negative bacillus, causes bacillary dysentery, an ulcerative disease of the human colon, by invading intestinal ep … ial genes necessary to achieve entry are located on five contiguous loci covering 30 kb on a 220 kb virulence plasmid in S. flexneri. Locus 2 has been particularly studied. Six genes organized as an operon encode highly immunogenic proteins among which IpaB (62 kD) and IpaC (48 kD) are the invasins of this microorganism which subsequently grows very rapidly …

Endemic nosocomial Legionnaires disease has occurred at our medical center for several years. Two subtypes of Legionella pneumophila serogroup 1 (UH-1 and RH-1) have been isolated in approximately equal numbers from hospital potable water. However, almost all clinical isolates have been UH-1. To assess potential differences in virulence, 50% lethal doses (LD50) and 50% infective doses (ID50) of UH-1 and RH-1 were determined by intraperitoneal infection in guinea pigs. The UH-1 LD50 was 7.41 X 10(6) CFU, which was significantly lower than the RH-1 LD50 of 9.12 X 10(7) CFU (P = 0.0001). The mean…

The comparative virulence of thermotolerant Mucorales was determined for cortisone-treated and untreated Swiss mice by intravenous administration of spores. The measure of virulence was based on an LD50 value, calculated after the 30-day observation period. Of the known etiological agents of mucormycosis, Mucor meihei, M. pusillus, Rhizopus arrhizus, R. chinensis, R. cohnii, R. microsporus, R. oryzae, R. rhizopodiformis and Cunninghamella elegans were able to produce fatal infections in mice; whereas, Mucor alternans, M. ramosissimus and Syncephalastrum racemosum were avirulent at dosages of u…

The pattern of redistribution of human virulence in epidemic strains of influenza viruses in recombination with human avirulent strains was studied. In the course of recombination of epidemic and attenuated influenza virus strains variants with different human virulence were obtained. Some recombinants manifested enforced reactogenic properties as compared with the epidemic strain (X/28, M/35, and 0/26--H1N1, and 2P--H3N2). At the same time, recombinants No 19 (H1N1) and 1P (H3N2) with a similar set of surface antigens were innocuous for man after intranasal administration. Using the observed …

1. Rabbits are very susceptible to infection by inhalation of Type I pneumococci. 2. When rabbits are exposed to a pneumococcus spray the bacteria readily penetrate into the lower respiratory tract. The pneumococci which reach the periphery of the lungs as a result of this procedure usually disappear within a few hours but a generalized and fatal septicemia frequently later appears. Pneumococci may then be recovered from the periphery of the kidney, liver, and spleen. In the animals which die pleurisy and pericarditis are common but pneumonia does not occur. 3. Rabbits may recover from pneumoc

Orientia tsutsugamushi is an obligate intracellular bacterium found in Leptotrombidium mites that causes the human disease scrub typhus. A distinguishing feature of O. tsutsugamushi is its extensive strain diversity, yet differences in virulence between strains are not well defined nor well understood. We sought to determine the bacterial drivers of pathogenicity by comparing seven strains using murine infections combined with epidemiological human data to rank each strain in terms of relative virulence. Murine cytokine expression data revealed that the two most virulent strains, Ikeda and Kat…

Recent isolation of the non-K1 Escherichia coli neonatal meningitis strain S286, belonging to phylogroup C, which is closely related to major group B1, and producing an extended-spectrum beta-lactamase, encouraged us to seek the genetic determinants responsible for its virulence. We show that S286 belongs to the sequence O type ST23O78 and harbors 4 large plasmids. The largest one, pS286colV (~120 kb), not related to resistance, contains genes characteristic of a Conserved Virulence Plasmidic (CVP) region initially identified in B2 extra-intestinal avian pathogenic E. coli (APEC) strains and i…

1. As regards these strains, at least, there is no consistent change, either increase or decrease, of rabbit pathogenicity during progressive rabbit passage. 2. There is no difference in pathogenicity between these different strains, although some of them were isolated from condylomata, some from chancres, one from a mucous patch, and one by Nichols from the nervous system. 3. We do not believe that there is any consistency of difference in speed of incubation between these various strains. 4. We think that the nature of the lesion produced by any individual strain, and the incubation time as

Vibrio vulnificus is a highly virulent gram-negative aquatic bacterium found in warm brackish waters and in molluscum shellfish worldwide that can cause sepsis after ingestion of raw oysters or undercooked molluscum shellfish as well as deadly skin and soft-tissue infections from immersion in contaminated seawater. Populations at a higher risk of infections are men older than 60 years with diabetes mellitus, liver cirrhosis, renal disease, or elevated iron levels and those who are immunocompromised. Three severe syndromes caused by V. vulnificus are gastroenteritis, primary septicemia, and wou

BACKGROUND: Species of Candida are the main cause of aggressive fungal infections in preterm newborns. These infections are linked to significant illness and death. Neonatal thrush is diagnosed by observing white patches on the surfaces of the mucosa of the mouth. Yeast is often associated with it. This study aimed to apply different techniques to characterize the yeasts associated with neonatal mouth thrush. Laboratory-based yeast species identification techniques (morphological and molecular) have been employed. Virulence genes were tested to compare experimental virulence assays and the pre…

Pathogenic Leptospira are spirochete bacteria which cause leptospirosis, a re-emerging zoonotic disease of global importance. Here, we use a recently described lineage of environmental-adapted leptospires, which are evolutionarily the closest relatives of the highly virulent Leptospira species, to explore the key phenotypic traits and genetic determinants of Leptospira virulence. Through a comprehensive approach integrating phylogenomic comparisons with in vitro and in vivo phenotyping studies, we show that the evolution towards pathogenicity is associated with both a decrease of the ability t…
