Physiology in microgravity.

Studies of physiology in microgravity are remarkably recent, with almost all the data being obtained in the past 40 years. The first human spaceflight did not take place until 1961. Physiological measurements in connection with the early flights were crude, but, in the past 10 years, an enormous amount of new information has been obtained from experiments on Spacelab. The United States and Soviet/Russian programs have pursued different routes. The US has mainly concentrated on relatively short flights but with highly sophisticated equipment such as is available in Spacelab. In contrast, the So

Ocular torsion in upright and tilted positions during hypo- and hypergravity of parabolic flight.

Four subjects considered resistant to motion sickness were tested in KC-135 parabolic flight to examine ocular torsion at hypo- and hypergravity. Three of these showed no significant torsion at zero G in either the upright position or when tilted 30 degrees to right or left. At 1.8 G in the tilted positions they showed greater ocular counterrolling than at 1 G. None of these three subjects became motion sick. The fourth subject showed eye torsion toward his left in all positions at zero G. This leftward bias could also be seen at 1.8 G when tilted left ear down, the side that induces rightward

Space motion sickness during 24 flights of the space shuttle.

The incidence and severity of Space Motion Sickness (SMS) were determined from 24 flights of the Space Shuttle. A standardized questionnaire developed at the NASA-Johnson Space Center (JSC) was administered to all crewmembers postflight during an oral debriefing with the examining flight surgeon. Cases of SMS were graded mild, moderate or severe using criteria developed at the JSC. The incidence of SMS during a first Shuttle flight for 85 crewmembers was 67% (57 cases). There were 26 mild cases (30%), 20 moderate (24%), and 11 severe (13%). Differences were found between males and females, cre

Carotid Body Chemoreceptors: from Mountain to Space.

Human acclimatization to both high-altitude and microgravity is crucial for terrestrial and space exploration. Although these extreme environments elicit several similar physiological responses, including autonomic imbalance, metabolic stress, and reduced exercise capacity, the underlying central mechanisms remain unclear. However, these effects appear to be linked to chemoreflex control. This review summarizes current evidence and proposed potential similarities and bioconvergent mechanisms underlying human responses to both terrestrial and extraterrestrial environments.

Musculoskeletal perturbations of deep space radiation: Assessment using a Gateway MRI.

Human space exploration expansion from Low-Earth Orbit to deep space is accelerating the need to monitor and address the known health concerns related to deep space radiation. The human musculoskeletal system is vulnerable to these risks (alongside microgravity) and its health reflects the well-being of other body systems. Multiparametric magnetic resonance imaging (MRI) is an important approach for assessing temporal physiological changes in the musculoskeletal system. We propose that ultra-low-field MRI provides an optimal low Size Weight and Power (SwaP) solution for non-invasively monitori

Responses to spaceflight of mouse mandibular bone and teeth.

OBJECTIVE: To determine if spaceflight and microgravity affect non-weight bearing bones and development and mineralization of teeth, reasoning that combining an organ and a cellular level approach can lead to greater insights about these effects. DESIGN: Mandibles and incisors of mice flown on the US STS-135 space shuttle mission and the Russian Bion-M1 satellite were studied using micro-computed tomography and immunohistochemistry. Ground controls were mice housed in standard vivarium cages and flight habitats. RESULTS: Incisor length was greater in the 13-day STS-135 flight mice than in ei

Preparative enrichment of human tissue cells capable to change a site of growth in vitro or in vivo - Recent developments.

Human cells are heterogeneous in regard to their biochemical features and functions. Detailed knowledge about each single cell type is important to understand the whole organism. In order to get a deeper insight in the concert of life, it has to be considered that cell populations such as thyroid cells, epithelial breast cells, endothelial cells, or chondrocytes are heterogeneous in regard to function, RNA expression patterns and protein content. This is true for normal cells and even more relevant for cancer cells. A number of sophisticated methods were developed to enrich cohorts of cells ge

Ultrasound evaluation of sinus fluid levels in swine during microgravity conditions.

BACKGROUND: Acute rhinosinusitis is a common problem that could occur in space secondary to absence of gravity-dependent drainage or odontogenic or external sources of infection. The purpose of this study was to determine the efficacy of ultrasound to determine sinus fluid distribution levels in swine and to assess the accuracy of ultrasound in the animal during normal and microgravity conditions. METHODS: Anesthetized swine had a catheter placed through a frontal bone window to allow aliquots of a viscous solution to be injected at 1 G (N = 4) or during brief microgravity parabolic flights (

Percutaneous bladder catheterization in microgravity.

INTRODUCTION: Urinary obstruction (UO) or failure to void has been observed during several episodes of short-duration spaceflight, necessitating bladder catheterization. It should be considered a possible medical condition in long-duration space missions as well. Antiemetics used early in space flight add to the risk and severity of voiding problems, along with the sensory and psychological peculiarities of voiding without gravity and in the unusual setting of a spacecraft. Urolithiasis due to the above-normal calcium excretion increases the risk of UO in long duration space missions. Finally,

3D realtime echography and echography assisted by a robotic arm for investigating astronauts in the ISS from the ground.

As human will stay for long duration in isolated sites like ISS there will be a need to perform quick and reliable diagnosis to evaluate the gravity of the pathology in presence of clinical symptoms. Many pathological situations (abnormal heart rate, pericardic collection, mitral prolaps, cholecystis, renal lithiasis, normal and ectopic pregnancies, ovarian cyst, acute appendicitis, phlebitis ... ) may occur even if all the astronauts are absolutely normal and healthy preflight. Ultrasound echography and Doppler are non invasive methods easy to use in space and very well adapted and used in ro

Head movements in low and high gravitoinertial force environments elicit motion sickness: implications for space motion sickness.

Astronauts report that head movements in flight tend to bring on symptoms of space motion sickness (SMS). We evaluated how head movements in pitch, yaw, and roll--made both with normal vision and eyes-occluded--affect susceptibility to motion sickness in the zero G phase of parabolic flight maneuvers. The findings are clearcut: pitch head movements are most provocative, yaw least provocative, and roll intermediate. Susceptibility is greater with normal vision than with eyes occluded. The same susceptibility pattern emerged for head movements in the 1.8-2.0 G phase of parabolic flight. These ex

Vestibulospinal reflexes as a function of microgravity.

The idea that sustained microgravity will result in altered otolith input requiring a modification of postural control was explored by using the pathway that links the otolith organs and spinal motoneurons. Two related methods were used. First, the Hoffmann reflex was used to measure at specific times the excitability of the soleus-spinal motoneuron pool during a brief unexpected linear acceleration. Second, extensive dynamic postural testing with a moving platform was done before and after flight. The Hoffmann reflex amplitude, reflecting otolith-modulated motoneuron sensitivity, was low in f

Single-cell analysis identifies conserved features of immune dysfunction in simulated microgravity and spaceflight.

Microgravity is associated with immunological dysfunction, though the mechanisms are poorly understood. Here, using single-cell analysis of human peripheral blood mononuclear cells (PBMCs) exposed to short term (25 hours) simulated microgravity, we characterize altered genes and pathways at basal and stimulated states with a Toll-like Receptor-7/8 agonist. We validate single-cell analysis by RNA sequencing and super-resolution microscopy, and against data from the Inspiration-4 (I4) mission, JAXA (Cell-Free Epigenome) mission, Twins study, and spleens from mice on the International Space Stati

Vascular hyporesponsiveness in simulated microgravity: role of nitric oxide-dependent mechanisms.

Simulated microgravity depresses the ability of arteries to constrict to norepinephrine (NE). In the present study the role of nitric oxide-dependent mechanisms on the vascular hyporesponsiveness to NE was investigated in peripheral arteries of the rat after 20 days of hindlimb unweighting (HU). Blood vessels from control rats and rats subjected to HU (HU rats) were cut into 3-mm rings and mounted in tissue baths for the measurement of isometric contraction. Mechanical removal of the endothelium from carotid artery rings, but not from aorta or femoral artery rings, of HU rats restored the cont

Reduced spontaneous baroreflex response slope during lower body negative pressure after 28 days of head-down bed rest.

Effects of 28 days of continuous 6 degrees head-down tilt bed rest on spontaneous vagally mediated baroreflex response slope were evaluated from beat-by-beat relationships between R-R interval and systolic arterial blood pressure. Twelve healthy men (age 27-42 yr) were assigned to either countermeasure (CM) or no-countermeasure (no-CM) groups. CM consisted of strenuous short-term exercise once per day 6 days/wk from days 7 to 28 and lower body negative pressure (LBNP) for 15 min on days 16, 18, 20, and 22-28. Spontaneous baroreflex slope was evaluated by application of linear regression to seq

Anorexia in space and possible etiologies: an overview.

Space travelers experience a flight duration-dependent loss in weight and body mass while in a microgravity environment, despite the absence of increased energy expenditure. Anorexia in space can lead to in-flight caloric deficits of 1330 kcal per 70 kg astronaut per day in the presence of abundant food and has a critical effect on endurance and performance. Microgravity, alterations in the light-and-dark cycle, and exposure to radiation energy are the environmental stresses believed to influence appetite, food intake, and gastrointestinal function during space flight. Review of data and recen

Indomethacin attenuates post-suspension hypotension in Sprague-Dawley rats.

Orthostatic hypotension is a serious condition that is sometimes manifested in astronauts during standing postflight. These observations may be related to impairment of autonomic function and/or excessive production of endothelium-dependent relaxing factors. To evaluate the role of the cyclooxygenase inhibitor indomethacin as a countermeasure against the post-suspension reduction in mean arterial pressure (MAP), we examined the cardiovascular responses to 7-day 30 degrees tail-suspension and a subsequent 6-hr post-suspension period in conscious male Sprague-Dawley rats. Indomethacin (2 mg/kg)
