Microbial succession-potential influence mechanism on flavor modulation in spontaneously fermented Moringa oleifera leaves: An integrative multi-omics approach.

In this study, the relationship between flavor composition and microbial succession in Moringa oleifera pickles (MOPs) at different stages of spontaneous fermentation was systematically investigated. The results demonstrated a significant increase in the content of organic acids and amino acids during fermentation including malonic acid, citric acid, valine (Val), and asparagine (Asn). These compounds not only enhanced the overall flavor profile but also provided favorable nutritional conditions that supported microbial succession. Furthermore, an integrated aroma network was established throu

Integrating multi-omics in bile for biomarker discovery in cholangiocarcinoma.

BACKGROUND: Cholangiocarcinoma (CCA) is an aggressive cancer with a poor prognosis. Histopathology evaluation of brushings and biopsies obtained during endoscopic retrograde cholangiopancreatography (ERCP) currently remains the main method of diagnosis, which has limited sensitivity for malignancy detection. Our study aimed to identify human bile-derived biomarkers to improve CCA diagnosis. Bile samples were collected from patients during ERCP for primary sclerosing cholangitis, CCA, or benign biliary disease. METHODS: Bile samples were collected from patients undergoing ERCP for biliary obst

Multi-omic analysis reveals lysine acylation and biofilm formation induced by central metabolites in methicillin-resistant Staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major multidrug-resistant pathogen responsible for severe infections, such as pneumonia and bloodstream infections. These infections are often associated with high mortality rates and pose a significant burden on public health. Studies have shown that biofilm formation is a key factor contributing to its enhanced drug resistance. Recent evidence also implicates core metabolites in biofilm regulation. However, their specific regulatory mechanisms remain unclear. In this study, we initially assessed how key metabolites from glycolysis and t

BACKGROUND: Bladder cancer (BLCA) continues to be a significant cause of cancer mortality in the urinary tract, with therapeutic resistance representing a major barrier to improving patient outcomes. Within the tumor microenvironment (TME), cancer-associated fibroblasts (CAFs) are pivotal drivers of BLCA progression, contributing to immune evasion and therapy resistance. This study leverages single-cell analysis to delineate CAF subclusters and explore the immune characteristics of CAFs-based BLCA classification. MATERIALS AND METHODS: Signal-cell RNA sequencing (scRNA-seq) datasets were used

Integration of multi-omics and crowdsourcing assessment of placenta-brain axis biomarkers for predicting neurodevelopmental disorders.

Neurodevelopmental disorders (NDDs) are heterogeneous and multifactorial psychiatric disorders with abnormalities in multiple biological domains. It is increasingly recognized that the placenta profoundly influences fetal neurodevelopment due to the finding of the placenta-brain axis. However, few studies have investigated the interplay between placenta dysfunction and NDDs, especially autism spectrum disorder (ASD) symptoms, attention-deficit/hyperactivity disorder (ADHD) symptoms, and intellectual disability (ID) symptoms, by using integrative multi-omics data. Here, we performed an analysis

Multi-omics insights into the metabolic reprogramming of host cells triggered by Entamoeba histolytica Gal/GalNAc lectin intermediate subunit.

The Gal/GalNAc lectin in ‌Entamoeba histolytica‌ is pivotal for its pathogenicity, and its intermediate subunit (Igl) is a crucial virulence factor. Here, the interference of eukaryotically expressed Igl on normal metabolic processes within Caco-2 cells was explored. Targeted metabolomic analysis detected 232 metabolites in Caco-2 cells, revealing 63 with significant alterations after Igl treatment. Notably, the lactate content significantly increased, indicating that Igl activates aerobic glycolysis. Metabolic flux analysis demonstrated that Igl could inhibit aerobic respiration and the trica

Integrated multi-omics mapping of mitochondrial dysfunction and substrate preference in Barth syndrome cardiac tissue.

Barth syndrome (BTHS) is a rare X-linked recessively inherited disorder caused by variants in the TAFAZZIN gene, leading to impaired conversion of monolysocardiolipin (MLCL) into mature cardiolipin (CL). Accumulation of MLCL and CL deficiency are diagnostic markers for BTHS. Clinically, BTHS includes cardiomyopathy, skeletal myopathy, neutropenia, and growth delays. Severely affected patients may require early cardiac transplants due to unpredictable cardiac phenotypes. The pathophysiological mechanisms of BTHS are poorly understood, and treatments remain symptomatic. This study analyzed heart
