Low cost tuberculosis vaccine antigens in capsules: expression in chloroplasts, bio-encapsulation, stability and functional evaluation in vitro.

Tuberculosis (TB) caused by Mycobacterium tuberculosis is one of the leading fatal infectious diseases. The development of TB vaccines has been recognized as a major public health priority by the World Health Organization. In this study, three candidate antigens, ESAT-6 (6 kDa early secretory antigenic target) and Mtb72F (a fusion polyprotein from two TB antigens, Mtb32 and Mtb39) fused with cholera toxin B-subunit (CTB) and LipY (a cell wall protein) were expressed in tobacco and/or lettuce chloroplasts to facilitate bioencapsulation/oral delivery. Site-specific transgene integration into the

Immunological cytokine correlates of protective immunity and pathogenesis in leprosy.

The in vitro production of interferon (IFN)-gamma, interleukin (IL)-5, tumour necrosis factor (TNF)-alpha and IL-10 by blood mononuclear cells in response to whole Mycobacterium leprae and polyclonal stimulii of 23 individuals, representing a variety of conditions in relation to exposure/susceptibility to M. leprae, was assayed. In most cases, healthy household contacts of newly diagnosed multibacillary leprosy patients, designated exposed household contacts (EC), showed low-to-undetectable in vitro IFN-gamma production in addition to substantial TNF-alpha production in response to M. leprae.

Seven decades after the Asian influenza pandemic: A historical review about immunity and vaccines against H2N2.

In 1957, a reassortant influenza A virus (IAV) H2N2 subtype emerged in humans and encountered a population that was antigenically naïve to this subtype. The lack of pre-existing immunity to the H2 hemagglutinin (HA) facilitated efficient human-to-human transmission, and by the end of the Summer of 1957, most countries around the world reported increasing influenza cases caused by the new influenza virus subtype. The pandemic lasted until 1958, resulting in millions of infections globally, with 1-4 million estimated deaths. The first vaccines targeting specifically the H2N2 subtype were availab

Leish-F1 Multi-epitope Lentiviral Vaccine as a Novel Candidate Against Leishmania major in Balb/c Mice.

The growing trend of Leishmania major (L. major) infections around the world has raised global concerns due to the lack of efficient vaccines and antiparasitic drugs. In addition, the drug resistance crisis and side effects of chemotherapy remain to be addressed. In the present study, the designed multi-epitope lentiviral vaccine from Leish-F1 (consisting of TSA, Leif, LmSTI1 proteins) was produced. For this purpose, multi-epitope construct was sub-cloned into the pCDH513 lentiviral vector, previously designed in silico. Subsequently, to produce a recombinant lentivirus, PCDH513B-Leish-F1 vect

In silico design of a multi-epitope vaccine targeting DENV-1 and DENV-3.

The co-infection of DENV-1 and DENV-3 during endemic outbreaks can be potentially fatal and complicate the diagnostic process. In our study, we have focused on the development of a multiepitope vaccine against DENV-1 and DENV-3 co-infection, utilizing non-structural protein 1 (NS1) and envelope protein (E) as key antigens. B cell and T cell epitopes were predicted for their immunogenicity, antigenicity, and ability to elicit an IFN-γ response. The final construct showed predicted stability (Instability Index: 30.63), antigenicity (0.5509), non-allergenicity, and hydrophilic character (GRAVY: -

Immunogenic Profiling Reveals Promising RV-Identified Antigens as Vaccine Candidates Against Klebsiella pneumoniae.

Multidrug-resistant Klebsiella pneumoniae is an increasing global threat, and the limited availability of new antibiotics highlights vaccination as a promising strategy for infection prevention. Although different vaccine candidates have been evaluated, none is currently approved, mainly due to capsular heterogeneity among strains. Protein-based vaccines may overcome this limitation by targeting conserved epitopes. In this study, we assessed the immunogenicity of five outer membrane proteins (ChiP, LamB, RafY, OmpW, PagP) previously selected by reverse vaccinology (RV), comparing them with the

Assessment of Mycobacterium tuberculosis dodecin scaffold as a multimerization platform on the immunogenicity of HPV L2 antigens.

Human papillomavirus infection, the primary cause of cervical cancer (the fourth most common cancer in women), is preventable through prophylactic HPV vaccines. As an alternative to current HPV vaccines, all based on a cocktail of L1 virus-like particles (VLPs) from multiple HPV types, the L2 capsid protein offers cross-protection via a conserved epitope residing at the N-terminus between residues 20-38. The Trx-L2(20-38)-8mer (Trx-8mer) is an L2-based antigen composed of a polytope of L2(20-38) epitopes from eight HPV types inserted into thioredoxin from the hyperthermophilic archaeon Pyrococ

Efficacy of an anti-fertility vaccine based on mammalian gonadotrophin releasing hormone (GnRH-I)--a histological comparison in male animals.

A N-terminal modified gonadotrophin releasing hormone (GnRH-I, tetanus toxoid-CHWSYGLRPG-NH2) conjugate was evaluated histologically in a number of male animal species (mice, dogs and sheep). The immunogen has previously been shown to be highly effective in rats, by suppressing both steroidogenesis and spermatogenesis. However, cross-species efficacy of peptide vaccines is known to be highly variable. Therefore, a comparative evaluation of reproductive tissues from animals immunized against this immunogen adsorbed onto an alum-based adjuvant was made. The sheep and dogs were chosen, as use of

Identification of a LolC homologue in Burkholderia pseudomallei, a novel protective antigen for melioidosis.

Melioidosis is an emerging disease of humans in Southeast Asia and tropical Australia. The bacterium causing this disease, Burkholderia pseudomallei, is also considered a bioterrorism agent, and as yet there is no licensed vaccine for preventing B. pseudomallei infection. In this study, we evaluated selected proteins (LolC, PotF, and OppA) of the ATP-binding cassette systems of B. pseudomallei as candidate vaccine antigens. Nonmembrane regions of the B. pseudomallei proteins were expressed and purified from Escherichia coli and then evaluated as vaccine candidates in an established mouse model

A trivalent gastric cancer antigen conjugated to a diphtheria toxin epitope synergizes with an NKT cell agonist to boost antitumor immunity.

The gastric cancer-associated antigen MG7-Ag is aberrantly expressed in multiple gastrointestinal malignancies and represents a promising broad-spectrum target for pan-gastrointestinal tumor peptide vaccines; however, its clinical application is hampered by low immunogenicity. To overcome this limitation, we covalently conjugated a diphtheria toxin-derived T-helper epitope to the trivalent antigen MG3, which was constructed by tandem repetition of the monovalent MG1 peptide from the MG7-Ag. A liposomal vaccine was subsequently developed by incorporating DT-MG3 together with the natural killer

Iron-oxide nanoparticles (SPIONs) enhance malaria vaccine antibody response.

Protein-subunit malaria vaccines typically elicit only modest and short-lived antibody responses without an effective delivery system. To address this limitation, carboxylated superparamagnetic iron-oxide nanoparticles (Fe3O4-COOH SPIONs) has been assessed as a carrier for the recombinant 19-kDa C-terminal fragment of Plasmodium falciparum merozoite surface protein-1 (PfMSP-1). Process began by synthesizing and functionalizing the SPIONs, followed by the conjugation of PfMSP-1 to the nanoparticle surface, and quantification of the antigen loading. BALB/c mice (n = 8/group) received one of four

Optimization and production of a GLP-grade recombinant Sm29 vaccine against schistosomiasis: From expression to preclinical assessment.

Schistosomiasis remains a major neglected tropical disease, and the development of a safe, effective vaccine is a global priority. Sm29, a Schistosoma mansoni surface antigen associated with naturally acquired resistance, has emerged as a promising vaccine candidate; however, its translational advancement requires both high-quality antigen production and evaluation with clinically acceptable adjuvants. Here, we report a Good Laboratory Practice-aligned process for generating tag-free recombinant Sm29 in Escherichia coli, including multi-step anion-exchange chromatography followed by subsequent

A chimeric trivalent Echovirus vaccine designed by loop substitution elicits cross-neutralizing immunity against serotypes 11, 18, and 30.

Echoviruses, members of the Enterovirus B genus in the Picornaviridae family, comprise numerous serotypes that frequently cause aseptic meningitis, myocarditis, and severe neonatal infections. The coexistence of multiple antigenically distinct serotypes poses a significant challenge for vaccine development, underscoring the clinical need for a broad-spectrum multivalent vaccine. In this study, we focused on three prevalent serotypes-Echovirus 11 (E11), 18 (E18), and 30 (E30)-as representative models to explore multivalent vaccine design. We first predicted and experimentally identified dominan
