Prediction of T cell epitopes of Brucella abortus and evaluation of their protective role in mice.

Brucellae are Gram-negative intracellular bacteria that cause an important zoonotic disease called brucellosis. The animal vaccines are available but have disadvantage of causing abortions in a proportion of pregnant animals. The animal vaccines are also pathogenic to humans. Recent trend in vaccine design has shifted to epitope-based vaccines that are safe and specific. In this study, efforts were made to identify MHC-I- and MHC-II-restricted T cell epitopes of Brucella abortus and evaluate their vaccine potential in mice. The peptides were designed using online available immunoinformatics to

Recombinant gp120 vaccine-induced antibodies inhibit clinical strains of HIV-1 in the presence of Fc receptor-bearing effector cells and correlate inversely with HIV infection rate.

Nonneutralizing Abs may play a role in protecting animals and humans from lentiviral infections. We explored the Ab-dependent, cell-mediated virus inhibition (ADCVI) Ab response to recombinant gp120 (rgp120) vaccination in sera from 530 participants in the Vax 004 trial. Serum ADCVI activity was measured against a clinical R5 strain of HIV-1 using peripheral blood mononuclear effector cells from healthy donors. The level of vaccine-induced ADCVI activity correlated inversely with the rate of acquiring HIV infection following vaccination, such that for every 10% increase in ADCVI activity, ther

AS01B-, MF59-, and alum-based adjuvants and HIV vaccine immunogenicity: a post-hoc cross-protocol comparison of results from HVTN 100, 107, 120, and 702.

INTRODUCTION: Vaccine adjuvants are crucial in HIV vaccine development, enhancing immune responses. Immune responses generated by different adjuvanted vaccines are rarely compared directly with the same vaccine antigens. METHODS: This retrospective cross-protocol, cross-sectional analysis examined four randomized, controlled, double-blinded, multicenter trials (HVTN 100, 107, 120, and 702) of adults without HIV who received the ALVAC-HIV (vCP2438) vaccine and a bivalent gp120 protein booster with either MF59, Alum, or AS01B adjuvant. Participants received ALVAC-HIV at months 0, 1, 3, 6, and 1

Effect of Fc Fusion on Folding and Immunogenicity of Middle East Respiratory Syndrome Coronavirus Spike Protein.

Middle East respiratory syndrome coronavirus (MERS-CoV) induces severe respiratory impairment with a reported mortality rate of ~36% in humans. The absence of clinically available MERS-CoV vaccines and treatments to date has resulted in uncontrolled incidence and propagation of the virus. In vaccine design, fusion with the IgG Fc domain is reported to increase the immunogenicity of various vaccine antigens. However, limited reports have documented the potential negative effects of Fc fusion on vaccine antigens. To determine whether Fc fusion affects the immunogenicity of MERS-CoV antigen, we c

Folate Receptor Alpha Peptide Vaccine Generates Immunity in Breast and Ovarian Cancer Patients.

Purpose: Folate receptor alpha (FR) is overexpressed in several cancers. Endogenous immunity to the FR has been demonstrated in patients and suggests the feasibility of targeting FR with vaccine or other immune therapies. CD4 helper T cells are central to the development of coordinated immunity, and prior work shows their importance in protecting against relapse. Our previous identification of degenerate HLA-class II epitopes from human FR led to the development of a broad coverage epitope pool potentially useful in augmenting antigen-specific immune responses in most patients.Patients and Met

Designing a multi-epitope peptide based vaccine against SARS-CoV-2.

COVID-19 pandemic has resulted in 16,114,449 cases with 646,641 deaths from the 217 countries, or territories as on July 27th 2020. Due to multifaceted issues and challenges in the implementation of the safety and preventive measures, inconsistent coordination between societies-governments and most importantly lack of specific vaccine to SARS-CoV-2, the spread of the virus that initially emerged at Wuhan is still uprising after taking a heavy toll on human life. In the present study, we mapped immunogenic epitopes present on the four structural proteins of SARS-CoV-2 and we designed a multi-ep

Plant Hsp90 is a novel adjuvant that elicits a strong humoral and cellular immune response against B- and T-cell epitopes of a Toxoplasma gondii SAG1 peptide.

BACKGROUND: The 90-kDa heat-shock protein (Hsp90) from Nicotiana benthamiana (NbHsp90.3) is a promising adjuvant, especially for those vaccines that require a T cell-mediated immune response. Toxoplasma gondii SAG1 is considered one of the most important antigens for the development of effective subunit vaccines. Some epitopes located in the SAG1 C-terminus region have showed a strong humoral and cellular immune response. In the present study, we aimed to assess the efficacy of NbHsp90.3 as carrier/adjuvant of SAG1-derived peptide (SAG1HC) in a T. gondii infection murine model. METHODS: In th

Long-Term Protective Immune Responses Induced by rBCG-RBD/rRBD Heterologous Prime/Boost Immunization Strategy: Fusion of RBD-Wuhan with LTB Adjuvant Induces Cross-Reactivity with SARS-CoV-2 Variant Omicron.

Background/Objectives: SARS-CoV-2, the causative agent of COVID-19, has been responsible for more than seven million deaths worldwide since its emergence. The Bacillus Calmette-Guérin (BCG) vaccine, used for over 100 years to prevent tuberculosis, induces a Th1-prominent immune response that is important for protection against Mycobacterium tuberculosis, other mycobacteria, and intracellular pathogens. BCG has also been shown to induce innate immune memory and heterologous protection against non-related infections. Additionally, BCG has been used as a vector to express heterologous proteins, s

Topographic differences in EEG microstates: distinguishing juvenile myoclonic epilepsy from frontal lobe epilepsy.

UNLABELLED: Pseudomonas aeruginosa (P. aeruginosa) is a critical healthcare challenge due to its ability to cause persistent infections and the acquisition of antibiotic resistance mechanisms. Lack of preventive vaccines and rampant drug resistance phenomenon has rendered patients vulnerable. As new antimicrobials are in the preclinical stages of development, mining for the unexploited drug targets is also crucial. In the present study, we designed a B- and T-cell multi-epitope vaccine against P. aeruginosa using a subtractive proteomics and immunoinformatics approach. A total of five proteins

Design, purification, and biophysical characterization of a multi-epitope vaccine construct made from outer membrane proteins of Pseudomonas aeruginosa.

Pseudomonas aeruginosa is increasingly becoming resistant to multiple drugs and is held responsible for high rate of mortality and morbidity across the globe. This study aims to examine proteins from outer membrane such as OprE, OprF, OprC, and OprG, for the multi-epitope vaccine design. In this article, the prediction of epitopes for helper T-cells, cytotoxic T-cells, and B-cell epitopes was carried out by the application of various immune-informatics tools. All the predicted epitopes were aligned and assembled into a peptide sequence along with linkers and adjuvant sequences. Further, second

Design of Multi-Epitope DIVA-Compatible Vaccine Candidate Against Canine Parvovirus 2.

Canine parvovirus type 2 (CPV-2) is a highly contagious and often fatal canine pathogen, with antigenic variants (CPV-2a, 2b, 2c), maternally derived antibody (MDA) interference, and lack of Differentiating Infected from Vaccinated Animals (DIVA) capability limiting current vaccines. Using an immunoinformatics-based reverse vaccinology approach, we identified conserved B-cell, cytotoxic T lymphocyte (CTL), and helper T lymphocyte (HTL) epitopes from CPV-2 structural (VP1, VP2) and nonstructural (NS1, NS2) proteins. These were screened for antigenicity, immunogenicity, safety, and stability. Th

Protective Effects of rLPG3 Plus Freund's Incomplete Adjuvant on Parasitism, Hepatic Function, and Immune Modulation in Experimental Visceral Leishmaniasis.

Visceral leishmaniasis (VL) is a neglected tropical disease affecting humans and dogs, particularly in urban settings. Current therapies are limited by toxicity, lengthy regimens, and emerging drug resistance. No human vaccine is available, and only a few licensed formulations exist for canine use. Here, we evaluated a recombinant Leishmania infantum lipophosphoglycan-3 (rLPG3) antigen formulated with Freund's incomplete adjuvant (FIA) against Leishmania infantum challenge in BALB/c mice. The formulation reduced hepatic parasitism, increased antioxidant enzyme activities (superoxide dismutase,

Bioinformatics-driven design and protective efficacy assessment of a multi-epitope vaccine for Toxoplasma gondii.

INTRODUCTION: Toxoplasma gondii can cause toxoplasmosis. It is an important type of pathogen within the broad category of emerging and re-emerging zoonoses. As an infectious disease featuring a complex multi-host transmission cycle, it poses an increasingly severe threat to global public health. No licensed vaccines are currently available for pets and humans, and thus a novel high-efficiency vaccine is urgently required. METHODS: Six antigens (GRA1, MIC17A, OWP2, LEA880, LEA870, and a hypothetical protein LEA530) representing different stages of the parasite lifecycle were selected from Toxo

Adjuvant-driven epitope hierarchy correlates with the protective efficacy of FimA vaccine against Klebsiella pneumoniae.

Hypervirulent Klebsiella pneumoniae (hvKP) variants represent urgent multidrug-resistant threats against which no licensed vaccine exists. FimA is indispensable for KP mucosal colonization and is thus a promising vaccine target; however, its protective immunodominant B-cell epitopes in vaccines with different adjuvants against KP remain undefined. Here, we revealed how adjuvants shape different hierarchies of the immunodominant B-cell epitopes of FimA and studied the humoral anti-FimA response and protective efficacy in a murine model of acute pneumonia. BALB/c mice were intramuscularly immuni

Intranasal immunization in mice with spore-associated serine-aspartate repeat-containing protein D from Staphylococcus aureus secreted in Bacillus subtilis.

Staphylococcus aureus is a major human pathogen causing diseases ranging from mild skin infections to life-threatening conditions due to its diverse virulence factors. The emergence of antibiotic-resistant strains underscores the urgent need for an effective vaccine, though none has yet achieved clinical success. The nasal mucosa serves as a natural reservoir in about 30% of individuals and represents a key source of subsequent infections, particularly in immunocompromised patients. Among its virulence factors, SdrD, a surface-anchored MSCRAMM adhesin associated with methicillin-resistant stra

Immunoinformatics design of a novel multiepitope vaccine candidate against non-typhoidal salmonellosis caused by Salmonella Kentucky using outer membrane proteins A, C, and F.

The global public health risk posed by Salmonella Kentucky (S. Kentucky) is rising, particularly due to the dissemination of antimicrobial resistance genes in human and animal populations. This serovar, widespread in Africa, has emerged as a notable cause of non-typhoidal gastroenteritis in humans. In this study, we used a bioinformatics approach to develop a peptide-based vaccine targeting epitopes from the outer membrane proteins A, C, and F of S. Kentucky. Additionally, we employed flagellin protein (fliC) from Salmonella Typhimurium (S. Typhimurium) as an adjuvant to enhance the vaccine's

Preclinical Efficacy of Clumping Factor A in Prevention of Staphylococcus aureus Infection.

UNLABELLED: Treatment of Staphylococcus aureus infections has become increasingly difficult because of the emergence of multidrug-resistant isolates. Development of a vaccine to prevent staphylococcal infections remains a priority. To determine whether clumping factor A (ClfA) is a good target protein for inclusion in a multivalent vaccine, we evaluated its efficacy in a variety of relevant staphylococcal infection models, challenging with different S. aureus strains. ClfA adsorbed to Alhydrogel and mixed with Sigma Adjuvant System was more immunogenic and stimulated a more robust Th17 respons

Amplifying immune responses via a parenteral prime-intranasal boost strategy using a lipidated chitosan nanoparticle-based multi-epitope COVID-19 vaccine.

As COVID-19 continues to spread and new SARS-CoV-2 variants emerge, the immunity provided by first-generation vaccines has been waning, highlighting the need for new strategies to enhance antigen-specific immune responses and overall vaccine efficacy. Nasal vaccination is a promising approach for inducing mucosal immune responses, offering protection at the primary sites of SARS-CoV-2 infection. In this study, we report the development of a novel vaccine formulation, CSOA-LipoCov, consisting of oleic acid-functionalized chitosan nanoparticles loaded with a lipidated multi-epitope peptide-conju

Cytotoxic and immunomodulatory activity of CD151-LEL-based peptides in breast cancer and THP-1 cells.

CD151, a cell surface oncoprotein belongs to the tetraspanin family, drives metastasis, drug resistance, and immune escape, and is recognized as a tumor-associated antigen (TAA). In the screening phase, a positive correlation between CD151 expression and amplification of CCND1, ERBB2, and MYC genes as well with breast cancer (BC) metastasis was observed using data from different GEO datasets. This study identified three immunogenic peptides within the large extracellular loop (LEL) of CD151 using in silico approaches: one each of CTL and HTL epitope, which bind strongly to class I and II MHC m
