Causal associations between primary sclerosing cholangitis and systemic lupus erythematosus: Evidence from Mendelian randomization and transcriptomic analyses.

Overlap between primary sclerosing cholangitis (PSC) and systemic lupus erythematosus (SLE) has been documented in previous studies, but the precise causal relationships between them remain elusive. This study aims to clarify the potential causal links between PSC and SLE using Mendelian randomization (MR) and transcriptomic analyses. Genome-wide association study (GWAS) summary data for PSC and SLE were obtained from the IEU OpenGWAS database. The inverse variance weighted (IVW) method was the primary approach for assessing the causal links between PSC and SLE. Potential horizontal pleiotropy

DIAGNOSTIC MANAGEMENT OF PATIENTS WITH ONYCHOMYCOSES.

INTRODUCTION: Onychomycosis is a fungal infection of the nail, causing discoloration and thickening of the affected nail plate, and is the most common nail infection worldwide. Microscopy and fungal culture are the gold standard techniques for onychomycosis diagnosis. At the same time, the culture method is long-term and requires significant costs. In this regard, a promising direction in laboratory diagnostics of onychomycosis is the detection of genetic markers of onychomycosis pathogens using polymerase chain reaction. The goal of current work is optimization of diagnostic care for patients

Deficiency of adenosine deaminase 2 skews adaptive immune repertoires toward specific sets of T- and B-cell receptors.

BACKGROUND: Adenosine deaminase 2 deficiency (DADA2) is a genetic disorder caused by biallelic hypomorphic or loss-of-function mutations in the ADA2 gene, which encodes a protein deaminase regulating extracellular adenosine metabolism. Clinical features encompass inflammatory vasculopathy, early-onset strokes, and a complex presentation involving both immunodeficiency and autoinflammation/autoimmunity. OBJECTIVE: Our aim was to determine a DADA2-specific adaptive immune architecture. METHODS: We profiled immunoglobulin levels and peripheral B- and T-cell phenotypes in 47 previously reported

Autosomal dominant in cis D4Z4 repeat array duplication alleles in facioscapulohumeral dystrophy.

Facioscapulohumeral dystrophy (FSHD) has a unique genetic aetiology resulting in partial chromatin relaxation of the D4Z4 macrosatellite repeat array on 4qter. This D4Z4 chromatin relaxation facilitates inappropriate expression of the transcription factor DUX4 in skeletal muscle. DUX4 is encoded by a retrogene that is embedded within the distal region of the D4Z4 repeat array. In the European population, the D4Z4 repeat array is usually organized in a single array that ranges between 8 and 100 units. D4Z4 chromatin relaxation and DUX4 derepression in FSHD is most often caused by repeat array c

Ultrasound attenuation imaging as a strategy for evaluation of early and late ambulatory functions in Duchenne muscular dystrophy.

BACKGROUND: Duchenne muscular dystrophy (DMD) is a genetic neuromuscular disorder that leads to mobility loss and life-threatening cardiac or respiratory complications. Quantitative ultrasound (QUS) envelope statistics imaging, which characterizes fat infiltration and fibrosis in muscles, has been extensively used for DMD evaluations. PURPOSE: Notably, changes in muscle microstructures also result in acoustic attenuation, potentially serving as another crucial imaging biomarker for DMD. Expanding upon the reference frequency method (RFM), this study contributes to the field by introducing the

Exploring prognostic biomarkers in pathological images of colorectal cancer patients via deep learning.

Hematoxylin and eosin (H&E) whole slide images provide valuable information for predicting prognostic outcomes in colorectal cancer (CRC) patients. However, extracting prognostic indicators from pathological images is challenging due to the subtle complexities of phenotypic information. We trained a weakly supervised deep learning model on data from 640 CRC patients in the prostate, lung, colorectal, and ovarian (PLCO) cancer screening trial dataset and validated it using data from 522 CRC patients in the cancer genome atlas (TCGA) dataset. We created the colorectal cancer risk score (CRCRS) t

Gradient Boosting Prediction of Overlapping Genes From Weighted Co-expression and Differential Gene Expression Analysis of Wnt Pathway: An Artificial Intelligence-Based Bioinformatics Study.

Introduction The Wnt (wingless-related integration site) signalling pathway is crucial for bone formation and remodelling, regulating the commitment of mesenchymal stem cells (MSCs) to the osteoblastic lineage. It triggers the transcriptional activation of Wnt target genes and promotes osteoblast proliferation and survival. Weighted co-expression network analysis (WGCNA) and differential gene expression analysis help researchers understand gene roles. Gradient boosting, a machine learning technique, enhances understanding of genetic and molecular mechanisms contributing to overlap genes, impro

A robust and validated integrated prognostic index for defining risk groups in adult acute lymphoblastic leukemia: an EWALL collaborative study.

Risk stratification is crucial to the successful treatment of acute lymphoblastic leukemia (ALL). Although numerous risk factors have been identified, an optimal prognostic model for integrating variables has not been developed. We used individual patient data from 4 contemporary academic national clinical trials, UKALL14, NILG-ALL10/07, GIMEMA-LAL1913, and PETHEMA-ALL-HR2011, to generate and validate the European Working Group for Adult ALL prognostic index (EWALL-PI), which is based on white blood cell count, genetics, and end of induction minimal residual disease (MRD). Individual patient r

Non-conventional deep brain stimulation in a network model of movement disorders.

Conventional deep brain stimulation (DBS) for movement disorders is a well-established clinical treatment. Over the last few decades, over 200,000 people have been treated by DBS worldwide for several neurological conditions, including Parkinson's disease and Essential Tremor. DBS involves implanting electrodes into disorder-specific targets in the brain and applying an electric current. Although the hardware has developed in recent years, the clinically used stimulation pattern has remained as a regular frequency square pulse. Recent studies have suggested that phase-locking, coordinated rese

Imaging Features of Arrhythmogenic Cardiomyopathies.

Arrhythmogenic cardiomyopathy (ACM) is a genetic disease characterized by replacement of ventricular myocardium with fibrofatty tissue, predisposing the patient to ventricular arrhythmias and/or sudden cardiac death. Most cases of ACM are associated with pathogenic variants in genes that encode desmosomal proteins, an important cell-to-cell adhesion complex present in both the heart and skin tissue. Although ACM was first described as a disease predominantly of the right ventricle, it is now acknowledged that it can also primarily involve the left ventricle or both ventricles. The original rig

Truncated M13 phage for smart detection of E. coli under dark field.

BACKGROUND: The urgent need for affordable and rapid detection methodologies for foodborne pathogens, particularly Escherichia coli (E. coli), highlights the importance of developing efficient and widely accessible diagnostic systems. Dark field microscopy, although effective, requires specific isolation of the target bacteria which can be hindered by the high cost of producing specialized antibodies. Alternatively, M13 bacteriophage, which naturally targets E. coli, offers a cost-efficient option with well-established techniques for its display and modification. Nevertheless, its filamentous

A finite porous-viscoelastic model capturing mechanical behavior of human cervix under multi-step spherical indentation.

The cervix is a soft tissue exhibiting time-dependent behavior under mechanical loads. The cervix is a vital mechanical barrier to protect the growing fetus. The remodeling of the cervical tissue, characterized by an increase in time-dependent material properties, is necessary for a safe parturition. The failure of its mechanical function and accelerated tissue remodeling is hypothesized to lead to preterm birth, which is birth before 37 weeks of gestation. To understand the mechanism of the time-dependent behavior of the cervix under compressive states, we employ a porous-viscoelastic materia

The causality between systemic inflammatory regulators and chronic respiratory diseases: A bidirectional Mendelian-randomization study.

INTRODUCTION: Accumulative evidence suggests the associations between systemic inflammatory regulators and chronic respiratory diseases (CRDs). However, the intrinsic causation remains implicit. Therefore, this study aimed to examine causative associations by mendelian randomization (MR) and to identify valuable active factors. METHODS: Based on data from the GWAS database, we performed MR analyses of 41 serum cytokines from 8,293 Finnish and European descent cohorts from GBMI and UKBB for five major CRDs. We mainly applied inverse variance weighted regression, supplemented by MR-Egger regres

GutMap: A New Interface for Analysing Regional Motility Patterns in ex vivo Mouse Gastrointestinal Preparations.

Different regions of the gastrointestinal tract have specific functions and thus distinct motility patterns. Motility is primarily regulated by the enteric nervous system (ENS), an intrinsic network of neurons located within the gut wall. Under physiological conditions, the ENS is influenced by the central nervous system (CNS). However, by using ex vivo organ bath experiments, ENS regulation of gut motility can also be studied in the absence of CNS influences. The current technique enables the characterisation of small intestinal, caecal, and colonic motility patterns using an ex vivo organ ba

The Application of GSCM in Eliminating Healthcare Waste: Hospital EDC as an Example.

Eliminating unnecessary healthcare waste in hospitals and providing better healthcare quality are the core issues of green supply chain management (GSCM). Hence, this study used a hospital's emergency department crowding (EDC) problem to illustrate how to establish an emergency medicine service (EMS) simulation system to obtain a robust parameters setting for solving hospitals' EDC and waste problems, thereby increasing healthcare quality. Inappropriate resource allocation results in more serious EDC; more serious EDC results in increasing operating costs. Therefore, in the healthcare system,

Design optimization of high tibial osteotomy plates using finite element analysis for improved biomechanical effect.

BACKGROUND: High tibial osteotomy (HTO) is a common treatment for moderate osteoarthritis of the medial compartment in the knee joint by the translation of the force center toward the lateral compartment. However, the stability of a short plate such as Puddu used in this procedure was not as effective as other long plates such as Tomofix. No previous studies have used a rigorous and systematic design optimization method to determine the optimal shape of short HTO plate. Therefore, the purpose of this study is to evaluate the improved biomechanical stability of a short HTO plate by using design
