A new holistic 3D non-invasive analysis of cellular distribution and motility on fibroin-alginate microcarriers using light sheet fluorescent microscopy.

Cell interaction with biomaterials is one of the keystones to developing medical devices for tissue engineering applications. Biomaterials are the scaffolds that give three-dimensional support to the cells, and are vectors that deliver the cells to the injured tissue requiring repair. Features of biomaterials can influence the behaviour of the cells and consequently the efficacy of the tissue-engineered product. The adhesion, distribution and motility of the seeded cells onto the scaffold represent key aspects, and must be evaluated in vitro during the product development, especially when the

Immunoisolation to prevent tissue graft rejection: Current knowledge and future use.

This review focuses on the concept of immunoisolation and how this method has evolved over the last few decades. The concept of immunoisolation came out of the need to protect allogeneic transplant tissue from the host immune system and avoid systemic side effects of immunosuppression. The latter remains a significant hurdle in clinical translation of using tissue transplants for restoring endocrine function in diabetes, growth hormone deficiency, and other conditions. Herein, we review the most significant works studying the use of hydrogels, specifically alginate and poly (ethylene glycol),

Heme Iron Release from Alginate Beads at In Vitro Simulated Gastrointestinal Conditions.

Heme iron (Fe) release from alginate beads at in vitro simulated gastrointestinal conditions for potential use as oral heme Fe supplement was studied. Five beads at different ratios of sodium alginate (SA)-to-spray-dried bovine blood cells (SDBC) with weight ratios of 1:1.25, 1:2.5, 1:5, 1:10, and 1:15 (w/w) were prepared. Release characteristics of these beads were investigated at in vitro simulated gastrointestinal conditions. Release media pH strongly influenced the controlled Fe release from the beads. The heme Fe-beads in simulated gastric fluid (pH 2) remained in a shrinkage state and Fe

A 3D system for culturing human articular chondrocytes in synovial fluid.

Cartilage destruction is a central pathological feature of osteoarthritis, a leading cause of disability in the US. Cartilage in the adult does not regenerate very efficiently in vivo; and as a result, osteoarthritis leads to irreversible cartilage loss and is accompanied by chronic pain and immobility (1,2). Cartilage tissue engineering offers promising potential to regenerate and restore tissue function. This technology typically involves seeding chondrocytes into natural or synthetic scaffolds and culturing the resulting 3D construct in a balanced medium over a period of time with a goal of

Release of furosemide from multiple-unit and single-unit preparations containing different viscosity grades of sodium alginate.

The release characteristics and the effect of viscosity of sodium alginate on the release rate of furosemide (a rather poorly soluble drug) from hard gelatin capsules (single-unit), and minitablets (multiple-unit) filled in hard gelatin capsules, were evaluated. Swelling and erosion experiments showed a different behavior for each viscosity grade. Polymer characteristics influenced significantly the release of the drug from the preparations prepared and tested. The results indicate that erosion plays a significant role, accelerates release rate and shortens duration of drug release. Low viscos

Mild-temperature conjugation of pea protein isolate with oxidized sodium alginate: structural, interfacial, and emulsion properties.

Pea protein isolate (PPI) has limited food applications because of poor aqueous solubility and emulsifying performance. Here, sodium alginate (SA) was oxidized to generate reactive aldehyde groups, enabling Schiff-base-mediated conjugation with PPI under mild conditions (40 °C, 90 min). By varying reaction parameters, conjugates with grafting degrees (GD) of 10.67-22.87% were obtained. Increasing GD was accompanied by increased β-sheet content (32.81-41.52%), tertiary structural reorganization, enhanced surface charge, a water contact angle of up to 79.35°, and a > 50% reduction in oil-water i

Management of upper gastrointestinal symptoms in patients with autoimmune gastritis.

PURPOSE OF REVIEW: Autoimmune gastritis is characterized by atrophy of acid secreting parietal cells resulting in achlorhydria. Upper gastrointestinal symptoms are common in autoimmune gastritis and frequently result in prescriptions for acid suppressant medications despite the inability of the stomach to secrete acid. Evidence-based recommendations for management of gastrointestinal symptoms in autoimmune gastritis are lacking. RECENT FINDINGS: The most common symptoms in patients with autoimmune gastritis are dyspepsia, heartburn, and regurgitation. Gastroesophageal reflux should be confirm

Negative impact of soluble, gel-forming dietary fibres on the bioaccessibility of β-carotene, lutein, and lycopene.

Carotenoids, including β-carotene, lutein and lycopene, the 3 main carotenoids present in our body, are lipophilic phytochemicals associated with several health benefits. Dietary fibre may influence the bioavailability of carotenoids by altering their release from the food matrix and their solubilisation into mixed micelles during digestion and thus their subsequent absorption by enterocytes. We examined the dose-dependent influence of major soluble (pectin, guar, alginate, fructooligosaccharides,) and insoluble dietary fibres (cellulose, type-2 resistant starch) at nutritional relevant doses

Functional Recovery and Safety of a Novel Hyaluronic Gel/Alginate Sheet for Complex Hand Injuries.

INTRODUCTION: Adhesion formation following hand and microsurgical procedures is a relatively common but often substantial complication, potentially limiting mobility and impairing recovery. A bioresorbable hyaluronic acid (HA)/alginate hydrogel was developed to reduce this risk. This study aimed to 1) evaluate functional recovery after use of this novel hydrogel with grip strength and range of motion (ROM), 2) determine its effect on patient-reported outcomes and pain, and 3) assess its safety and complication profile. METHODS: A prospective, single-center cohort study was conducted in 17 pat

A 3D-printed scaffold composed of Alg/HA/SIS for the treatment of diabetic bone defects.

BACKGROUND: Diabetic bone healing remains a great challenge due to its pathological features including biochemical disturbance, excessive inflammation, and reduced blood vessel formation. In previous studies, small intestine submucosa (SIS) has been demonstrated for its immunomodulatory and angiogenic properties, which are necessary to diabetic bone healing. However, the noticeable drawbacks of SIS such as fast degradation rate, slow gelling time, and weak mechanical property seriously impede the 3D printing of SIS for bone repair. METHOD: In this study, we developed a novel kind of 3D-printe

Rapidly reducing cadmium from contaminated farmland soil by novel magnetic recyclable Fe3O4/mercapto-functionalized attapulgite beads.

Reducing cadmium (Cd) content from contaminated farmland soils remains a major challenge due to the difficulty in separating commonly used adsorbents from soils. This study synthesized novel millimeter-sized magnetic Fe3O4/mercapto-functionalized attapulgite beads (MFBs) through a facile one-step gelation process incorporating alginate. The MFBs inherit the environmental stability of alginate and enhance its mechanical strength by hybridizing Fe3O4 and clay mineral components. MFBs can be easily separated from flooded soils by magnets. When applied to 12 Cd-polluted paddy soils and 14 Cd-pollu

A Three-Dimensional Printable Hydrogel Formulation for the Local Delivery of Therapeutic Nanoparticles to Cervical Cancer.

Cervical cancer is the fourth most common malignancy among women. Compared to other types of cancer, therapeutic agents can be administrated locally at the mucosal vaginal membrane. Thermosensitive gels have been developed over the years for contraception or for the treatment of bacterial, fungal, and sexually transmitted infections. These formulations often carry therapeutic nanoparticles and are now being considered in the arsenal of tools for oncology. They can also be three-dimensionally (3D) printed for a better geometrical adjustment to the anatomy of the patient, thus enhancing the loca
