Enamel mineralisation is a highly controlled process in which the deposition, growth, and maturation of inorganic crystallites are regulated by secreted matrix proteins at the molecular and cellular level. Maxillary and mandibular first molars from the col1-caPPR mutants as well as normal controls aged for 12 weeks were observed by SEM and nanoindentation, respectively. Several types of aberrations in enamel distribution and crystal organisation were encountered in the transgenic molars. Also, the gene alteration resulted in degradation by as much as 23.42% in hardness and 17.56% in the elasti

The clinico-pathological records of 30 unicystic ameloblastomas collected over a period of 10 years were studied. The mean age at diagnosis was 18.0 years (SD +/- 8.1), most lesions were located in the mandible and were frequently associated with impacted teeth, root resorption and tooth displacement. The unicystic ameloblastomas in 11 patients (4 females and 7 males) exhibited invasion of the fibrous wall, 4 cases (1 female and 3 males) showed intra-luminal proliferation and the remaining 15 specimens (9 females and 6 males) were lined by non-proliferating ameloblastic epithelium. Two cases r

Comparative analysis of tooth development in the main vertebrate lineages is needed to determine the various evolutionary routes leading to … er (IDE) and outer dental epithelium being separated by stellate reticulum. IDE differentiates into ameloblasts, which produce enamel matrix components. In the region facing differentiating IDE, mesenchymal cells differentiate into dental papilla and give rise to odontoblasts, which first deposit a layer of predentin matrix. The first elements of the enamel m …

Ameloblastoma or adamantinoma is the rarest of the three forms of tumor of the odontogenic type. They are benign, locally aggressive neoplasms arising from ameloblasts, which typically occur at the angle of the mandible, and are often associated with an un-erupted tooth and must, therefore, be differentiated from a dentigerous cyst which will be centered on the crown. When in the maxilla (less common), they are located in the premolar region, and can extend up in the maxillary sinus. Ameloblastoma is reported to constitute about 1-3% of tumors and cysts of the jaws. The tumor is by far more co…

Deafblindness is part of several genetic disorders. We investigated a consanguineous Egyptian family with two siblings affected by congenita … led white matter disease in both. We show that Pex6 localizes to the apical extensions of secretory ameloblasts and differentiated odontoblasts at early stages of dentin synthesis in mice, and to cilia of retinal photoreceptor cells. We propose PEX6, and possibly other peroxisomal genes, as candidate for the rare cooccurrence of deafblindness and enamel dyspl …

Fluoride plays a dual role in dental health-preventing caries at optimal levels but causing fluorosis when excessive. While most animal studies focus on young mice, age-related susceptibility to fluoride remains poorly understood. This study presents the first comprehensive analysis of developmental stage-dependent differences in fluoride toxicity, focusing on enamel formation and systemic fluoride clearance. Male C57BL/6J mice-adolescent (5-9 weeks) and mature (16-20 weeks)-were exposed to fluoride in drinking water (0, 50, 100, or 125 ppm) for 6 weeks. Adolescent mice developed pronounced de

Amelogenins and tuftelins are highly specialized proteins secreted into the developing enamel matrix during mammalian enamel formation. Both tuftelins and amelogenins have been associated with various functions during nucleation and maturation of the developing enamel matrix. In this study we conducted experiments to investigate whether tuftelins and portions of the amelogenin molecule were deposited and processed in spatially distinguished portions of the developing enamel matrix, using antibodies specific against tuftelin or amelogenins. The amelogenin antibodies were raised against recombin

This article reviews odontogenic and developmental oral lesions encountered in the gnathic region of pediatric patients. The process of odontogenesis is discussed as it is essential to understanding the pathogenesis of odontogenic tumors. The clinical presentation, microscopic features, and prognosis are addressed for odontogenic lesions in the neonate (dental lamina cysts/gingival cysts of the newborn, congenital (granular cell) epulis of the newborn, melanotic neuroectodermal tumor, choristoma/heterotopia, cysts of foregut origin), lesions associated with unerupted/erupting teeth (hyperplast

Whether the interference by fluoride (F-) with secretory amelogenesis in vitro could be modulated by altering the levels of calcium (Ca) and … of pre-exposure enamel. High Ca levels also considerably reduced the cellular changes in secretory ameloblasts induced by 52 microM of F- and slightly counteracted the inhibition of matrix secretion, as measured biochemically. Some of the effects of F- on secretory amelogenesis in vitro can thus be reversed by raising Ca levels in the medium. Therefore, the …

Proximal renal tubular acidosis (pRTA) is a syndrome caused by abnormal proximal tubule reabsorption of bicarbonate resulting in metabolic acidosis. Patients with mutations to the SLC4A4 gene (coding for the sodium bicarbonate cotransporter NBCe1), have pRTA, growth delay, ocular defects, and enamel abnormalities. In an earlier report, we provided the first evidence that enamel cells, the ameloblasts, express NBCe1 in a polarized fashion, thereby contributing to trans-cellular bicarbonate transport. To determine whether NBCe1 plays a critical role in enamel development, we studied the expressi…

Dentin matrix protein 1 (DMP1) is an Arg-Gly-Asp-containing acidic phosphoprotein that was originally identified from a rat incisor cDNA library and was thought to be a dentin-specific protein. DMP1 was later shown to express in a number of hard tissue-forming cells, including osteoblasts, osteocytes, ameloblasts, and cementoblasts, and was considered to play important roles in mineralization. Further, DMP1 gene expression was also detected in fetal bovine brain and in newborn mouse brain. These findings indicate the possibility of DMP1 expression in other soft tissues. In the present study, t…

In vivo, RANKL and OPG were localized to odontoblasts, ameloblasts, and pulp cells in developing mouse teeth by immunohistochemistry. In a coculture system, we found the dental cells to be inhibitory to ocl formation from spleen and bone marrow precursors, despite their production of osteoclast stimulatory factors. …

The ultracytochemical acridine orange (AO) method has been employed to demonstrate DNA template activity within embryonic incisor tooth organs excised from New Zealand White rabbits during the 25th day of gestation. Survey ultrastructural examination revealed characteristic distribution patterns of AO positive cell populations in the inner and outer enamel epithelium and the adjacent ectomesenchyme of the cervical loop region and in populations of fibroblast-like cells connecting as a band the outer AO positive cell groups. With increasing differentiation of ameloblasts and odontoblasts the nu…

Dentinogenic ghost cell tumors are rare tumors, and few cases of them were reported in the literature. The presence of pigment in odontogenic lesions is a rare unexplained histological finding. In this report, we describe a unique case of a 7-year-old girl that was referred to the Department of Oral and Maxillofacial Surgery complaining of a left mandibular swelling. Clinical examination revealed a huge, ulcerated mass. Both incisional and excisional biopsies revealed a benign infiltrative odontogenic tumor with admixed ameloblast-like cells and pigmented ghost cells, consistent with a pigment

Hypoxia is characterized by a disparity between supply and demand of oxygen. The association between hypoxia and head and neck tumors is a topic of significant interest. Tumors frequently encounter areas with inadequate oxygen supply, resulting in a hypoxic microenvironment. Ameloblastoma is one of the most common benign odontogenic tumors of the maxillofacial region. It is a slow-growing but locally invasive tumor with a high recurrence rate. The literature has demonstrated the correlation between hypoxia and ameloblastoma, revealing a discernible link between the heightened expression of hyp
