Study on the physicochemical indexes, nutritional quality, and flavor compounds of Trichiurus lepturus from three representative origins for geographical traceability.

Trichiurus lepturus (hairtail) is an important economic component of China's marine fishing industry. However, due to the difficulty in identifying the appearance of hairtail from different geographical distributions, hairtails with geographical indication trademarks were imitated by general varieties. In this study, the texture characteristics, color, basic nutrients, amino acids, mineral, fatty acids, and volatile flavor substances were used as indicators for multivariate statistical analysis to determine whether three origins of hairtails from the habitats of Zhoushan (East China Sea, T.Z),

The Role of Branched-Chain Amino Acids and Branched-Chain α-Keto Acid Dehydrogenase Kinase in Metabolic Disorders.

Branched-chain amino acids (BCAAs), composed of leucine, isoleucine, and valine, are important essential amino acids in human physiology. Decades of studies have revealed their roles in protein synthesis, regulating neurotransmitter synthesis, and the mechanistic target of rapamycin (mTOR). BCAAs are found to be related to many metabolic disorders, such as insulin resistance, obesity, and heart failure. Also, many diseases are related to the alteration of the BCAA catabolism enzyme branched-chain α-keto acid dehydrogenase kinase (BCKDK), including maple syrup urine disease, human autism with e

[Peritoneal dialysis in a neonate with maple syrup urine disease. A case report].

Maple syrup disease is an autosomal recessive entity caused by a congenital error in the metabolism of three essential branchedchain amino acids: valine, leucine and isoleucine. The neonatal form of this disease is expressed by a severe and progressive neurological compromise, associated with a peculiar smell of urine, a consequence of the elimination of the excess of these amino acids. This smell of burnt sugar mimics the molasses obtained from maples, which gives its name to this disease. The best method to eliminate these toxins is hemodiafiltration, but in centers where this practice is no

Enhancement of growth performance, muscle nutrient composition, and nutritional value in rainbow trout (Oncorhynchus mykiss) by guanidinoacetic acid-enriched diets.

The role of guanidinoacetic acid (GAA) in modulating nutritional value and muscle quality warrants attention, particularly given the scarcity of mechanistic studies in aquatic animals. A total of 450 healthy juvenile rainbow trout (Oncorhynchus mykiss, aged approximately 76 d post-hatch, initial body weight 7.77 ± 0.14 g) were randomly distributed into 5 groups (3 replicates per group, 30 fish per replicate) to investigate the effects of dietary GAA supplementation at concentrations of 0, 300, 600, 1200, and 2400 mg/kg on muscle nutritional value and quality, designated as groups G0, G3, G6, G

Free amino acid levels in serum and small intestine during the post-natal development of normal and sparse-fur mutant mice.

Ornithine transcarbamylase (OTC) is a mitochondrial enzyme which catalyzes the conversion of ornithine into citrulline. In sparse-fur (spf) mutant mice, in which both hepatic and small intestinal OTC are deficient, only small amounts of citrulline are released into the blood. To evaluate the fate of ornithine as well as the levels of substrates and products of these metabolic pathways in normal and OTC-deficient mice, we measured the concentration of free amino acids in small intestinal mucosa and serum during post-natal development. In control animals, ornithine, alanine and citrulline concen

Comparative metabolomic assessment of bioconverted traditional and black wheat via the in vitro piglet digestion model.

Colored wheat is gaining significant attention in the food and feed industry due to its components’ potential health benefits. As weaning piglets experience major discomfort in their early life, the quality and bioavailability of the feed nutrients become crucial for their survival. Fermentation of the feed aims to be a cheap and efficient stage of the feed production, as it is able to overcome major shortcomings of the cereals. The aim of this study was to investigate the differences in metabolites between traditional (variety “Herkus”) and black (variety “DS8558-1”) (DS) wheat cereals (both

Structural insight of the 5-(Hydroxyethyl)-methylthiazole kinase ThiM involving vitamin B1 biosynthetic pathway from the Klebsiella pneumoniae.

Thiamin pyrophosphate (TPP) is an essential co-factor in amino acid and carbohydrate metabolic pathways. The TPP-related vitamin B1 biosynthetic pathway is found in most bacterial, plant and lower eukaryotic processes; however, it is not present in humans. In bacterial thiamin synthesis and salvage pathways, the 5-(hydroxyethyl)-methylthiazole kinase (ThiM) is essential in the pathway forming TPP. Thus, ThiM is considered to be an attractive antibacterial drug target. Here, we determined the crystal structures of ThiM from pathogenic Klebsiella pneumoniae (KpThiM) and KpThiM in complex with it

Analysis of Shigella flexneri wzz (Rol) function by mutagenesis and cross-linking: wzz is able to oligomerize.

The modal length or degree of polymerization (dp) of the Shigella flexneri O-antigen is determined in an unknown manner by the Wzz/Rol protein. The Wzz protein is anchored into the cytoplasmic membrane by two transmembrane domains (TM1 amino acids 32-52; TM2 amino acids 295-315) with the central loop of the protein located in the periplasm. Plasmids were constructed encoding hybrid Wzz proteins consisting of regions of S. flexneri Wzz (WzzSF) and Salmonella typhimurium Wzz (WzzST). These imparted O-antigen modal chain lengths that implied that the carboxy-terminal region of Wzz was involved in

The Health Benefits of Egg Protein.

Once the general public accepts that dietary cholesterol is not a concern for cardiovascular disease risk, foods that have been labeled as high-cholesterol sources, including eggs, may be appreciated for their various other dietary components. One of the nutrients in eggs that deserves further discussion is egg protein. Egg protein has been recognized to be highly digestible and an excellent source of essential amino acids, with the highest attainable protein digestibility-corrected amino acid score. Egg protein has been shown to decrease malnutrition in underdeveloped countries, possibly incr

A Limited Number of Amino Acid Permeases Are Crucial for Cryptococcus neoformans Survival and Virulence.

One unique attribute of Cryptococcus neoformans is its ability to procure essential monomers from its surroundings to survive in diverse environments. Preferentially, sugars are the energy sources for this opportunistic pathogenic fungus under the carbon catabolite repression (CCR); however, sugar restriction induces alternative use of low molecular weight alcohol, organic acids, and amino acids. The expression of transmembrane amino acid permeases (Aaps) allows C. neoformans to utilize different amino acids and their conjugates, notwithstanding under the nitrogen catabolite repression (NCR).

The complex role of branched chain amino acids in diabetes and cancer.

The obesity and diabetes epidemics are continuing to spread across the globe. There is increasing evidence that diabetes leads to a significantly higher risk for certain types of cancer. Both diabetes and cancer are characterized by severe metabolic perturbations and the branched chain amino acids (BCAAs) appear to play a significant role in both of these diseases. These essential amino acids participate in a wide variety of metabolic pathways, but it is now recognized that they are also critical regulators of a number of cell signaling pathways. An elevation in branched chain amino acids has

Noscapine-Amino Acid Conjugates Suppress the Progression of Cancer Cells.

Lung cancer is the leading cause of cancer deaths globally; 1 in 16 people are diagnosed with lung cancer in their lifetime. Microtubules, a critical cytoskeletal assembly, have an essential role in cell division. Interference with the microtubule assembly leads to genetic instability during mitosis and cancer cell death. Currently, available antimitotic drugs such as vincas and taxanes are limited due to side effects such as alopecia, myelosuppression, and drug resistance. Noscapine, an opium alkaloid, is a tubulin-binding agent and can alter the microtubule assembly, causing cancer cell deat

Rapid development of oedema and defective brain histidine metabolism in young guinea pigs fed protein-energy deficient diet.

Male weanling guinea pigs were fed ad libitum a 3% pelleted casein diet supplemented with ascorbic acid while control animals were pair-fed a standard guinea-pig chow. Oedema was observed in some of the former group as early as the 2nd week and became very prominent by the end of the 3rd week. The malnourished group was characterized by reduced plasma albumin level and some disaggregation of hepatic polyribosomes but these showed no correlation in time with development of oedema. The pool of plasma essential amino acids were markedly reduced as early as Day 7, and most prominently affected wer

Amino acid biosynthetic routes as drug targets for pulmonary fungal pathogens: what is known and why do we need to know more?

Amongst 1.5 million fatal mycoses of humans occurring annually [1], the vast majority involve the human lung as the primary site of pathogenesis, and are derived from organisms which occupy environmental niches. On entry into the respiratory system pathogenic fungi must draw upon metabolic versatility for survival and proliferation as the mammalian lung is a nutritionally limiting environment. The nutritional stresses encountered have exposed vulnerabilities which have long been viewed as potential antifungal targets, since humans lack several of the metabolic pathways which fungi rely upon fo

Induced mechanism of phosphatase hormesis by Cd ions and rhizosphere metabolites of Trifolium repens L.

Rhizosphere phosphatases can exhibit hormetic effects in response to cadmium (Cd) ion stimulation. However, understanding the mechanisms underlying hormesis effects on soil ecosystems is challenging as studies on hormesis are usually specific to an organism, cell, or organ. To comprehensively investigate the mechanism of phosphatase hormesis, this study utilized in situ zymography and metabolomics to analyze the rhizosphere of Trifolium repens L. (white clover). Zymograms showed that rhizosphere phosphatase displayed a hormetic effect in 10 mg kg-1 Cd contaminated soil, with a hotspot area 1.8
