Western Anthelmintics in Early Twentieth-Century China Colonial Practices and Knowledge on "Tropical Diseases" of the In/between.

Protestant (medical) missionaries were the main proponents of Western medicine in China after the Opium Wars. Several studies have highlighted how they used spectacular surgery as a means of gaining public trust. As well as surgery, they also administered anthelmintic drugs such as santonin as a tool of persuasion and conversion. Many anthelmintic drugs of the European materia medica had a colonial history. My paper analyses how coloniality materialised in medical practice and anthelmintics in China. For the late nineteenth century, I will examine the colonial practices in which the drug santo…

The activity of drug-metabolizing enzymes and the biotransformation of selected anthelmintics in the model tapeworm Hymenolepis diminuta.

The drug-metabolizing enzymes of some helminths can deactivate anthelmintics and therefore partially protect helminths against these drugs' toxic effect. The aim of our study was to assess the activity of the main drug-metabolizing enzymes and evaluate the metabolism of selected anthelmintics (albendazole, flubendazole, mebendazole) in the rat tapeworm Hymenolepis diminuta, a species often used as a model tapeworm. In vitro and ex vivo experiments were performed. Metabolites of the anthelmintics were detected and identified by HPLC with spectrofluorometric or mass-spectrometric detection. The …

Studies in potential filaricides. 18. Synthesis of 2,2'-disubstituted 5,5'-dibenzimidazolyl ketones and related compounds as potential anthelmintics.

A series of 2,2'-disubstituted 5,5'-dibenzimidazolyl ketones and related compounds have been synthesized of which 2,2'-bis(carbomethoxyamino)-5,5'-dibenzimidazolyl ketone exhibited a broad spectrum of anthelmintic activity in experimental animals. At doses of 10-50 mg/kg given intraperitoneally, 5 killed 100% of the adult worms of Litomosoides carinii, Dipetalonema viteae, and Brugia malayi. By the oral route the macrofilaricidal efficacy of 5 was 97-100% at 100-200 mg/kg X 5 days. The treated animals showed gradual disappearance of microfilariae and before autopsy they became amicrofilariaemi

Studies on the effect of treatment with anthelmintics on weight gain in calves, naturally infected with gastrointestinal nematodes.

To avoid disease outbreaks caused by gastro-intestinal nematodes different prophylactic measures are recommended. In this connection pasture hygienic achievements and anthelmintics are used. In practice it is often difficult to realize pasture hygienic steps as move the animals to parasite free areas and to avoid that they come back to the infected. In many cases the parasite control is limited to routine deworming. Our intention was to study if anthelmintic treatment without other measures had any effect on weight gain and number of eggs in faeces in calves naturally infected with Ostertagia …

Anthelmintic resistance in canine parasitology: a critical review of an emerging global challenge.

Anthelmintic resistance (AR) in canine parasitology is rapidly emerging as a significant health challenge. Growing reports of multidrug resistance in the canine hookworm Ancylostoma caninum, as well as macrocyclic lactone resistance in the heartworm Dirofilaria immitis, and isolated cases in Dipylidium caninum, highlight the increasing importance of AR. Unlike livestock, AR in dogs develops under different dynamics, shaped by unique epidemiological and biological features, including lower treatment frequencies, smaller parasite populations, and distinct husbandry practices. Nevertheless, frequ

Evaluation of nematode susceptibility and resistance to anthelmintic drugs with a WMicrotracker motility assay.

Grazing ruminants suffer from various helminth infections particularly those caused by gastrointestinal nematode (GIN) parasites, which have a considerable impact on their welfare and productivity. To treat these infections, the intensive use of macrocyclic lactone (ML) anthelmintics has led to the emergence of drug-resistant parasite populations worldwide. The standard method for detecting resistance, the Faecal Egg Count Reduction Test (FECRT), is susceptible to misinterpretation, leading to flawed management decisions that undermine parasite control efforts. Thus, there is a pressing need f…

Anthelmintic and Antibiotic Therapy Resolves Intestinal Inflammatory Infiltration in Experimental Trichuriasis.

Despite significant advances in human health, soil-transmitted helminths (STH) continue to pose a major public health challenge, particularly in impoverished regions. Albendazole has been used to treat STH for over 40 years and remains widely utilized in mass drug administration programs. However, it is estimated that over 1.5 billion people are still infected globally, with Brazil reporting a prevalence of 5.41% for human trichuriasis. The nematode Trichuris muris is widely used in murine models to study trichuriasis due to its impact on the epithelial mucosa, including tissue damage, dysbios

Paraherquamides - A new hope and great expectations of anthelmintic agents: Computational studies.

Nematode infections impose a significant health and economic burden, particularly as parasites develop resistance to existing treatments and evade host defenses. This study explores the efficacy of 48 paraherquamide analogs, a class of polycyclic spiro-oxindole alkaloids with unique structural features, as potential anthelmintic agents. Employing advanced computational methods, including molecular docking, MM-GBSA, and molecular dynamics simulations, we assessed the interaction of these analogs with the Ls-AchBP receptor, a model for nematode neurotransmission. Among the analogs studied, Parah

Modelling bodyweight to avoid anthelmintic underdosing of goats in resources-limited settings.

Improved management of livestock in resource-limited settings can provide a means towards improved human nutrition and livelihoods. However, gastrointestinal nematodes (GIN) are a significant production-limiting factor. Anthelmintics play a role in GIN management; however, few anthelmintic classes are available in many low-middle-income countries. Utilising a limited range of classes may increase selection for anthelmintic resistance; therefore, strategies to reduce other selective pressures are of heightened importance. Avoiding anthelmintic underdosing is one such strategy, but it can be cha…

Interactions of anthelmintic veterinary drugs with the soil microbiota: Toxicity or enhanced biodegradation?

Anthelmintic (AH) compounds are used to control gastrointestinal nematodes (GINs) in livestock production. They are only partially metabolized in animals ending in animal excreta whose use as manures leads to AH dispersal in agricultural soils. Once in soil, AHs interact with soil microorganisms, with the outcome being either detrimental, or beneficial. We aimed to disentangle the mechanisms of these complex interactions. Two soils previously identified as « fast » or « slow», regarding the degradation of albendazole (ABZ), ivermectin (IVM), and eprinomectin (EPM), were subjected to repeated a

Role of the Abcg2 Transporter in Secretion into Milk of the Anthelmintic Clorsulon: Interaction with Ivermectin.

Clorsulon is a benzenesulfonamide drug that is effective in treating helminthic zoonoses such as fascioliasis. When used in combination with the macrocyclic lactone ivermectin, it provides high broad-spectrum antiparasitic efficacy. The safety and efficacy of clorsulon should be studied by considering several factors such as drug-drug interactions mediated by ATP-binding cassette (ABC) transporters due to their potential effects on the pharmacokinetics and drug secretion into milk. The aim of this work was to determine the role of ABC transporter G2 (ABCG2) in clorsulon secretion into milk and

Anthelmintic niclosamide attenuates pressure-overload induced heart failure in mice.

The heart is a high energy demand organ and enhancing mitochondrial function is proposed as the next-generation therapeutics for heart failure. Our previous study found that anthelmintic drug niclosamide enhanced mitochondrial respiration and increased adenosine triphosphate (ATP) production in cardiomyocytes, therefore, this study aimed to determine the effect of niclosamide on heart failure in mice and the potential molecular mechanisms. The heart failure model was induced by transverse aortic constriction (TAC) in mice. Oral administration of niclosamide improved TAC-induced cardiac hypertr
