Relationship between Chlamydia-Ureaplasma-Mycoplasma Genital Detection with Semen Concentration and Motility among Greek Men.

One hundred and seventy two men at the State of Thessaly, Greece, inquiring semen analysis were enrolled in the study in order to investigate the incidence of Chlamydia, Ureaplasma and Mycoplasma (C-U-M) genera in respect to total sperm number (TSN), progressive motility (grades a and b) and total motility (grades a, b and c). Putative relation of C-U-M acquirement with sexual behavior was also investigated. Incidence of C-U-M among non-oligozoospermic and oligozoospermic men was similar. Νο correlation of C-U-M carriage to either oligozoospermia or asthenozoospermia was found. The tested seme…

The dose-response relationship between seminal plasma metal mixtures and oligo-astheno-teratozoospermia: a hospital-based case-control study.

BACKGROUND: Oligo-astheno-teratozoospermia (OAT), a leading cause of male infertility, affects approximately 30 % of reproductive-aged men. While trace elements influence semen quality, prior investigations predominantly focused on single-element effects. Critical knowledge gaps persist regarding the combinatorial impacts of multi-element exposures on OAT pathogenesis. METHODS: Our study enrolled 743 OAT cases and 179 controls. The seminal plasma element concentrations were quantified via inductively coupled plasma-mass spectrometry (ICP-MS). Analytical approaches mainly included restricted c

Mechanism of Shu-Mu Brain-Kidney Acupoint in Treating Oligoasthenospermia in Rats via the Hypothalamic-Pituitary-Testicular Axis.

Oligoasthenospermia is a common cause of male infertility. The hypothalamic-pituitary-testicular (HPT) axis regulates gonadal differentiation and maturation through reproductive hormone synthesis and release, playing a vital role in male fertility. Disrupting HPT axis stability impairs sperm production, reducing semen quality. Investigating electroacupuncture's effect on HPT axis regulation may provide insights into treating oligoasthenospermia. Fifty 8-week-old male SD rats were randomly divided into blank, model, Shu-Mu Brain-Kidney, non-acupoint, and L-carnitine groups (n = 10 per group). E

Novel mutations in FSIP2 cause male infertility through multiple morphological abnormalities of the sperm flagella.

Infertility is a global concern, and oligoasthenoteratozoospermia (OAT) is the most severe form of male infertility, characterized by reduced sperm count, decreased motility, and increased abnormal morphology. Multiple morphological abnormalities of the sperm flagella (MMAF) characterize the most severe type of OAT and are usually caused by loss-of-function mutations in the genes essential for vital aspects of sperm biology, including concentration, motility, and morphology. The fibrous sheath interacting protein 2 (FSIP2) plays an essential role in sperm flagellar structure and function by re

Novel biallelic DNHD1 variants associated with male infertility with severe MMAF phenotype.

Multiple morphological abnormalities of the sperm flagella (MMAF), characterized by severe morphological sperm defects, such as absent, short, irregular caliber, and coiled flagella with extreme asthenoteratozoospermia, are the most prevalent cause of human male infertility. Previous studies have identified several genes linked to MMAF; however, the increasing incidence of infertility indicates that most affected individuals remain undiagnosed, prompting further investigation to uncover novel mutations and genes. Whole-exome sequencing (WES) was conducted on a consanguineous infertile family f

The possible association of clusterin fucosylation changes with male fertility disorders.

In the seminal plasma (n = 118) and serum (n = 90) clusterin (CLU) the fucosylation and the expression of selected fucosyltransferases (FUTs) were analyzed. Samples from infertile men were divided into groups based on the results of the standard semen analysis: normozoospermic (N), teratozoospermic (T), asthenoteratozoospermic (AT) and oligoasthenoteratozoospermic (OAT). The CLU fucosylation was analyzed using lectin-ELISAs with biotinylated lectins specific to α1,3-, α1,2-linked antennary fucose, and α1,6-linked core fucose (LTA, UEA, and LCA, respectively). The concentrations of FUT3 and FUT

Proteomic analysis reveals the negative modulator of sperm function glycodelin as over-represented in semen exosomes isolated from asthenozoospermic patients.

STUDY QUESTION: Are there differences in the proteomic profile of exosomes isolated from seminal plasma of normozoospermic (NSP) and severe asthenozoospermic (SA) men, potentially contributing to sperm features? SUMMARY ANSWER: A relevant group of proteins known to positively regulate sperm functions were over-represented in seminal exosomes of NSP men, i.e. cysteine-rich secretory protein-1 (CRISP1), while the inhibitory protein glycodelin was enriched in exosomes of SA subjects. WHAT IS KNOWN ALREADY: Exosomes are secreted along the male reproductive tract and are thought to be involved in

Histone modification signatures in human sperm distinguish clinical abnormalities.

PURPOSE: Alternations to the paternal epigenome, specifically the components of sperm chromatin, can lead to infertility in humans and potentially transmit aberrant information to the embryo. One key component of sperm chromatin is the post-translational modification of histones (PTMs). We previously identified a comprehensive profile of histone PTMs in normozoospermic sperm; however, only specific histone PTMs have been identified in abnormal sperm by antibody-based approaches and comprehensive changes to histone PTM profiles remain unknown. Here, we investigate if sperm with abnormalities of

Impaired embryogenesis following intracytoplasmic sperm injection with an Arrdc5-/- mouse model of oligoasthenoteratospermia†.

Oligoasthenoteratospermia (OAT) describes male infertility due to a combination of low sperm count, reduced sperm motility, and abnormal sperm morphology. A common course of action to overcome infertility for males with this condition is intracytoplasmic sperm injection (ICSI). Here we used the Arrdc5 knockout mouse model of OAT to examine the ramifications of bypassing natural barriers preventing fertilization by defective sperm via ICSI on the viability of embryos and health of offspring. ARRDC5 is expressed specifically in the male germline where it has an essential role in sperm morphogene

2,8-Dihydroxyquinoline ameliorates oligoasthenozoospermia by inhibiting MAPK14-mediated spermatogonial ferroptosis.

BACKGROUND: Oligoasthenozoospermia (OAS) is a major cause of male infertility. 2,8-Dihydroxyquinoline (2,8-DHQ), an endogenous antioxidant metabolite, remains unexplored in OAS. This study identified 2,8-DHQ as a bioactive XHBG metabolite and elucidated its mechanisms in improving OAS. METHODS AND RESULTS: Male BALB/c mice received glycosides of Tripterygium wilfordii (GTW) (140 mg/kg/day, oral gavage) for 4 weeks to induce OAS, confirmed by marked reductions in testicular/epididymal weights, sperm concentration, serum testosterone, and severe seminiferous tubule disruption. Then, mice were t…

Homozygous CCDC146 mutation causes oligoasthenoteratozoospermia in humans and mice.

Infertility represents a significant health concern, with sperm quantity and quality being crucial determinants of male fertility. Oligoasthenoteratozoospermia (OAT) is characterized by reduced sperm motility, lower sperm concentration, and morphological abnormalities in sperm heads and flagella. Although variants in several genes have been implicated in OAT, its genetic etiologies and pathogenetic mechanisms remain inadequately understood. In this study, we identified a homozygous nonsense mutation (c.916C>T, p.Arg306*) in the coiled-coil domain containing 146 ( CCDC146) gene in an infertile
