Epstein-Barr virus latent membrane protein 2A blocks calcium mobilization in B lymphocytes.

LMP2A is expressed in latent Epstein-Barr virus (EBV) infection and interacts with LMP1 and members of the src tyrosine kinase family in the plasma membrane. Since tyrosine kinase mediate receptor-induced changes in intracellular free calcium, the effect of LMP2A on receptor-mediated intracellular calcium mobilization was evaluated by stably expressing LMP2A in an EBV-negative Burkitt tumor cell line (BJAB) or in LMP1-converted BJAB cells. LMP2A significantly blocked calcium mobilization following class II, CD19, or immunoglobulin M cross-linking. LMP2A effects were partially reversed in LMP1-

Stimulation of murine intestinal intraepithelial lymphocytes by the bacterial superantigen staphylococcal enterotoxin B.

To gain insight into the specificity and function of intestinal intraepithelial lymphocytes (IEL), we have examined their response to staphylococcal enterotoxin B (SEB), a significant cause of food poisoning and a potent T cell mitogen. IEL include two populations of TCR alpha beta+ T cells. One of these resembles the T cells found in the Peyer's patch and is thymus dependent. The other subset is characterized by both TCR alpha beta and gamma delta+ IEL bearing a unique form of the CD8 molecule, expressed as an alpha alpha homodimer. CD8 alpha+ beta- IEL are thymus independent and appear to ma

HIV induces IL-6 production by human B lymphocytes. Role of IL-4.

In vitro, normal B cells can produce TNF-alpha and IL-6 when activated with a first signal, and cytokines and B lymphocytes from some HIV-infected individuals spontaneously secrete TNF-alpha and IL-6, although the direct involvement of HIV has not been fully explored. In this study, we examined the effects of HIV (purified virus and a recombinant envelope protein) and various IL on TNF-alpha and IL-6 in vitro production by highly purified normal B cells. HIV alone did not induce IL-6 or TNF-alpha production by B cells from healthy subjects. HIV induced IL-6 production (500 to 1500 pg) in the p

Virus-specific B-lymphocytes are probably the primary targets for Aleutian disease virus.

368 1- to 5-year-old mink of wild-type or black genetic background were infected with Aleutian disease virus (ADV) naturally or using virus-containing immune complexes or purified virus. Thirty of the mink were immunized with dinitrophenol-conjugated ovalbumin (DNP-OA) before and during infection. Blood samples were taken at monthly intervals. We found that weak (and transient) monoclonal or oligoclonal immunoglobulin components were present in the plasma or serum approximately 1 month after infection, as judged by zone electrophoresis. In a few cases, we found quite stable myeloma-like hyperg

Effect of interferon-gamma on B lymphocytes of patients with systemic lupus erythematosus.

We studied the effect of interferon-gamma (IFN-gamma) on B cells in systemic lupus erythematosus (SLE). Low density B cells, which were fractionated on density gradients of Percoll, increased, and high density B cells decreased in number in SLE. Proliferative response of the high density B cells to interleukin 4 was reduced by IFN-gamma in normal controls, but not in SLE. IgG production of whole B cells induced by interleukin 2 or phytohemagglutinin induced T cell factors was enhanced by IFN-gamma in both normal controls and SLE in which activated B cells were thought to be increased in number

Human severe combined immunodeficiency disease: phenotypic and functional characteristics of peripheral B lymphocytes.

Human severe combined immunodeficiency disease (SCID) includes an X-chromosome-linked type characterized by a complete absence of mature T cells, hypogammaglobulinemia but normal or elevated number of B cells, suggesting that the disease results from a block in early T cell differentiation. It has been shown that B cells from obligate carrier women of this disorder exhibit the preferential use of the nonmutant X chromosome as the active X (as shown for T cells), suggesting that the SCID gene product has a direct effect on B cells as well as on T cells. To examine this question, we analyzed the

Demonstration and partial characterization of the interferon-gamma receptor on human B lymphocytes.

The expression of interferon-gamma (IFN-gamma) receptors on normal human B cells and four B cell lines was studied. Recombinant human IFN-gamma was labeled with [gamma-32P]ATP using the catalytic subunit of a cAMP-dependent protein kinase. All four B cell lines, although differing in their responsiveness to IFN-gamma, were found to express high-affinity receptors (1,000-11,000 receptors/cell). Normal unactivated B lymphocytes were also found to express constitutively high-affinity receptors, approximately 1,400 receptors per cell with an estimated affinity of 295 pM. Activation of the normal B

Occurrence of mature B (IgM+, B220+) and T (CD3+) lymphocytes in scid mice.

Scid mice with and without detectable serum Ig (scid Ig+ and scid Ig- mice, respectively) were examined for the presence of mature "leaky" lymphocytes by flow microfluorimetry with the use of antibodies to B (IgM, B220) and T (CD3, CD4, CD8) lymphocyte surface Ag. The data showed that leaky scid mice are more frequent than is evident from serum Ig analysis and that the incidence of detectable B and T cells increases with age. IgM+ B220+ cells were not detectable in young adult mice (3 mo old), but in old mice (greater than or equal to 1 yr old) they were routinely present in the peritoneal cav

The negative surface charge density is a maturation marker of human B lymphocytes.

Small resting B lymphocytes were highly enriched and completely depleted of all preactivated large B lymphocytes using countercurrent centrifugal elutriation and free flow electrophoresis. They required T lymphocytes, monocytes, and a mitogen to produce antibodies after 5 days of preincubation. Large activated B lymphocytes were obtained in cell fractions which were free of resting ones. They produced antibodies even in the absence of a mitogen. Two groups were distinguished, differing in their stage of differentiation and their negative surface charge density. The cells of one group had an el

Differential sulphation of chondroitins in murine T and B lymphocytes and lymphoma cells.

Splenic B and T lymphocytes metabolically labelled in vitro with [35S]-H2SO4 show differential sulphation of chondroitin sulphate, the major sulphated glycosaminoglycan in both cell-retained and secreted fractions. In B cells, 6-O-sulphation is more abundant than 4-O-sulphation, whereas in T cells sulphation occurs predominantly in the 4-O-position. The murine lymphoma lines AKTB-1b and EL-4, of B and T origin respectively, show even more marked differences. AKTB-1b follows the pattern of normal B cells, whilst in EL-4 only chondroitin 4-sulphate is synthesized.

Production of chemokinetic inhibitory factor (CIF) by normal blood and spleen B lymphocytes.

We have recently reported the partial purification and characterization of of a new lymphokine, the heat-labile chemokinetic inhibitory factor (CIF) which inhibits neutrophil movement. We have also shown that this lymphokine is produced and secreted by cultured B-chronic lymphocytic leukaemia (CLL) cells in vitro. The present study shows that highly purified resting normal B lymphocytes from blood and spleen have the capacity to produce CIF spontaneously. After activation with anti-IgM or EBV-infection the lymphocytes produced a number of other factors, heat-stable chemokinetic inhibitory fact

In vitro production of interleukin 1 by normal and malignant human B lymphocytes.

In this study, the capacity of normal and neoplastic B lymphocytes to release interleukin 1 (IL 1) has been investigated. Peripheral blood B cells from normal donors were isolated by depletion of E rosetting cells and by positive selection of cells expressing surface immunoglobulin (sIg) or the B1 marker. Peripheral blood B cells from patients with B cell chronic lymphocytic leukemia (B-CLL) were purified by removal of E rosetting cells followed by complement-mediated cytotoxicity with selected monoclonal antibodies. All of the normal B cell suspensions and the large majority of the B-CLL cell

Selective partial IgM deficiency: functional assessment of T and B lymphocytes in vitro.

Seven patients with selective IgM deficiency (SIgMD) were studied for cell surface immunoglobulin (SmIg), T-cell subpopulations, and immunoglobulin (Ig) synthesis in vitro by peripheral blood lymphocytes (PBL). Serum IgM levels were less than 25 mg/dl, while IgA, IgG, and IgD were within normal levels. The patients had respiratory or urinary tract infections and two were diagnosed as having systemic lupus erythematosus (SLE). T/B-cell ratios in PBL were within normal ranges. Percentage ratios of B cells bearing SmIg were normal in five patients and decreased in two; however, normal values were

A p50 surface antigen restricted to human urinary bladder carcinomas and B lymphocytes.

We have previously described the derivation of a monoclonal antibody, S2C6, to a novel 50 Kdalton antigen associated with human urinary bladder carcinoma. No reactions were obtained with carcinomas of unrelated origin or with normal urothelial cells. However, the antibody also reacted with a similar antigen on some cell lines of B lymphocyte origin. Using large panels of target cells we have now shown that this reactivity was entirely restricted to cells of the B lineage within the haematopoietic system. As opposed to its apparent restriction to malignant cells of the urothelium, the S2C6 anti

Distribution of T and B lymphocytes in lymphoid tissue of infants and children.

Normal lymphoid tissue from children undergoing elective surgery was examined for T and B lymphocyte distribution. Although established for peripheral blood and bone marrow, T and B lymphocyte distributions have not been previously reported for lymph nodes, appendix, thymus, and spleen tissues in children. Thymus-dependent T cells were determined by the sheep erythrocyte rosette technique, and thymus-independent B cells were determined by the fluorescent labeling of surface immunoglobulins A (IgA), G (IgG), and M (IgM). Fifty percent of lymph node cells were either T or B cells; 65% of these c

B-cell monoclonal lymphocytosis and B-cell abnormalities in the setting of familial B-cell chronic lymphocytic leukemia.

BACKGROUND: Among all hematologic malignancies, B-cell chronic lymphocytic leukemia (BCLL) has the highest familial clustering (three- to sevenfold increase), strongly suggesting a genetic component to its etiology. Familial BCLL can be used as a model to study the early pathogenesis of this disease. METHODS: We examined nine kindreds from the National Cancer Institute's Familial BCLL Registry, consisting of 19 affected members with BCLL and 33 clinically unaffected first-degree relatives. Flow cytometric immunophenotyping to detect a B-cell monoclonal lymphocytosis (BCML) was performed. Mono

B Lymphocytes in Acute Ischemic Stroke: From Immune Tolerance to Neuroprotection and Autoimmunity.

Whereas the pathobiology of stroke is traditionally thought to be mediated mainly by innate immune cells, the recent discovery that immune tolerance to brain-specific antigens takes place in the meninges suggests that the CNS makes use of unique pathways to respond to brain-derived antigens. By virtue of their adaptive and innate-like functions, B lymphocytes can play multiple roles in stroke. Following acute ischemic stroke, B cells home to the site of injury and produce cytokines that can either promote or exacerbate inflammation. They also secrete autoantibodies against neuronal and/or vasc

Identifying Negative Biomarkers of CD4 Cytotoxic T Lymphocytes via Granzyme B Exclusion Analysis.

CD4 cytotoxic T lymphocytes (CTLs) are a distinct subset of CD4+ T cells that can mediate antigen-specific cytotoxicity. However, their specific surface markers have yet to be defined. In this study, we used gene-plasticity-based mutual exclusion analysis to identify negative biomarkers for CD4 CTLs, using granzyme B (GZMB) expression as the pivotal reference. We analyzed bulk RNA-seq data from 768 human CD4+ T cell samples, calculated gene plasticity scores, and applied cosine similarity to detect genes that are anti-correlated with GZMB. Several genes encoding cell surface membrane proteins

Bexarotene signaling in human B and T lymphocytes induces gut-homing receptor expression.

BACKGROUND: Retinoic acid (RA) receptors (RARs) in human lymphocytes modulate the humoral and intestinal immune response by regulating target genes, including CD38, TGM2, and gut-homing markers. The impact of retinoid X receptors (RXRs) on this process is elusive. OBJECTIVE: To determine the impact of the RXR ligand bexarotene (BXR) on the activation and differentiation of human B and T lymphocytes. METHODS: In vitro BXR stimulation of human CD19+ B cells and CD4+ T helper cells was investigated regarding retinoid target gene expression using qPCR and flow cytometry and validated in peripher
