Discovery of a novel nonsteroidal selective glucocorticoid receptor modulator by virtual screening and bioassays.

Synthetic glucocorticoids (GCs) have been widely used in the treatment of a broad range of inflammatory diseases, but their clinic use is limited by undesired side effects such as metabolic disorders, osteoporosis, skin and muscle atrophies, mood disorders and hypothalamic-pituitary-adrenal (HPA) axis suppression. Selective glucocorticoid receptor modulators (SGRMs) are expected to have promising anti-inflammatory efficacy but with fewer side effects caused by GCs. Here, we reported HT-15, a prospective SGRM discovered by structure-based virtual screening (VS) and bioassays. HT-15 can selectiv

Sensitivity of Haemonchus contortus to anthelmintics using different in vitro screening assays: a comparative study.

BACKGROUND: Helminthiasis and resistance to commercial anthelmintic compounds are major causes of economic losses for livestock producers, resulting in an urgent need for new drugs and reliable in vitro screening tests capable of detecting potentially active products. Considering this, a series of novel benzimidazole derivatives (5-methylbenzimidazole 1,2-disubstituted, 5-carboxybenzimidazole, 5-methylbenzimidazole 2-one) was screened on exsheathed L3 (xL3) and on the adult stage of Haemonchus contortus (Kirby anthelmintic-susceptible McMaster isolate). METHODS: This work presents the set-up

Real-time quaking-induced conversion assay is accurate for Lewy body diseases: a meta-analysis.

OBJECTIVE: Real-time quaking-induced conversion (RT-QuIC) is a novel in vitro acellular seed amplification analysis and has been widely used to detect prion diseases. Due to the similar mechanism of abnormal aggregation of α-synuclein, RT-QuIC has great potential for diagnosing Lewy body diseases. This meta-analysis was performed to evaluate the diagnostic accuracy of RT-QuIC for Lewy body diseases. METHODS: This study followed the PRISMA statement. We searched six databases for relevant studies published until February 20, 2022. Meta-analysis was conducted using RevMan 5.3, Stata 17.0, and M

Methotrexate Detection in Serum at Clinically Relevant Levels with Electrochemically Assisted SERS on a Benchtop, Custom Built Raman Spectrometer.

Therapeutic drug monitoring (TDM) is an essential clinical practice for optimizing drug dosing, thereby preventing adverse effects of drugs with a narrow therapeutic window, slow clearance, or high interperson pharmacokinetic variability. Monitoring methotrexate (MTX) during high-dose MTX (HD-MTX) therapy is necessary to avoid potentially fatal side effects caused by delayed elimination. Despite the efficacy of HD-MTX treatment, its clinical application in resource-limited settings is constrained due to the relatively high cost and time of analysis with conventional analysis methods. In this w

3D host cell and pathogen-based bioassay development for testing anti-tuberculosis (TB) drug response and modeling immunodeficiency.

Tuberculosis (TB) is a global health threat that affects 10 million people worldwide. Human Immunodeficiency Virus (HIV) remains one of the major contributors to the reactivation of asymptomatic latent tuberculosis (LTBI). Over the recent years, there has been a significant focus in developing in-vitro 3D models mimicking early events of Mycobacterium tuberculosis (Mtb) pathogenesis, especially formation of the granuloma. However, these models are low throughput and require extracellular matrix. In this article, we report the generation of a matrix-free 3D model, using THP-1 human monocyte/mac

Identification of non-validated endocrine disrupting chemical characterization methods by screening of the literature using artificial intelligence and by database exploration.

BACKGROUND: Exposure to endocrine disrupting chemicals (EDCs) represents a critical public health threat. Several adverse health outcomes (e.g., cancers, metabolic and neurocognitive/neurodevelopmental disorders, infertility, immune diseases and allergies) are associated with exposure to EDCs. However, the regulatory tests that are currently employed in the EU to identify EDCs do not assess all of the endocrine pathways. OBJECTIVE: Our objective was to explore the literature, guidelines and databases to identify relevant and reliable test methods which could be used for prioritization and reg

Bruce Nathan Ames - Paradigm shifts inside the cancer research revolution.

Dr. Bruce Ames turned 92 on December 16, 2020. He considers his most recent work linking adequate consumption of 30 known vitamins and minerals with successful aging to be his most important contribution. With the passage of time, it is not uncommon for the accomplishments of a well-known scientist to undergo a parsimonious reductionism in the public mind - Pasteur's vaccine, Mendel's peas, Pavlov's dogs, Ames' test. Those of us in the research generation subsequent to Dr. Ames' are undoubtedly affected by our own unconscious tendencies toward accepting the outstanding achievements of the past

Determination of Chemical Inhibitor Efficiency against Intracellular Toxoplasma Gondii Growth Using a Luciferase-Based Growth Assay.

Toxoplasma gondii is a protozoan pathogen that widely affects the human population. The current antibiotics used for treating clinical toxoplasmosis are limited. In addition, they exhibit adverse side effects in certain groups of people. Therefore, discovery of novel therapeutics for clinical toxoplasmosis is imperative. The first step of novel antibiotic development is to identify chemical compounds showing high efficacy in inhibition of parasite growth using a high throughput screening strategy. As an obligate intracellular pathogen, Toxoplasma can only replicate within host cells, which pro

Leptospira spp. Toolbox for Chemotaxis Assay.

A toolbox for chemotaxis assay adapted to Leptospira spp. has emerged in the recent years: soft agar assay, capillary assay, and videomicroscopy tracking. Those methods allow to demonstrate chemotaxis defect, identify diverse chemoattractants, or decipher motile behavior quantitatively. These experiments have demonstrated a role of motility and potentially chemotaxis in leptospirosis pathogenesis. We describe extensively the methods and provide the key steps to use this toolbox.

"Armed to the teeth": The multiple ways to survive insecticidal and predatory challenges in Aedes aegypti larvae.

Environmental pollutants, such as insecticides, can alter the equilibrium of aquatic ecosystems, particularly those closely located to human occupations. The use of such anthropogenic compounds frequently results in the selection of resistant individuals. However, how the underlying insecticide resistance mechanisms interplay with the abilities of the resistant individuals to cope with other environmental challenges (e.g., predators) has not received adequate attention. Here, we evaluated whether resistance to pyrethroid insecticides in larvae of the yellow fever mosquito, Aedes aegypti (Dipte

Spectrophotometric method for the quantitative assay of N-hydroxysulfosuccinimide esters including extinction coefficients and reaction kinetics.

A quantitative spectrophotometric method has been developed for the analysis of N-hydroxysulfosuccinimide (sulfo-NHS), a chromophore with a maximum absorbance at 268 nm. The extinction coefficients were determined between pH 6.0 and 8.0 and found to vary in a nonlinear manner. This spectrophotometric profile is not present in its esters which however release an equimolar amount of sulfo-NHS when they react with nucleophilic groups or hydrolyze in aqueous solution. This fact facilitates the determination in solution of the concentration and purity of bis(sulfosuccinimidyl) suberate (BS3) used a

Rabies in the insectivorous Pallas's mastiff bat (Molossus molossus) in northeastern Brazil.

We describe the pathologic and immunohistochemical findings in five cases of rabies in the insectivorous Pallas's mastiff bat (Molossus molossus) in the city of Patos, state of Paraíba, northeastern Brazil. All cases were found during the day, in different neighborhoods, prostrate and unable to fly. No significant lesions were observed at necropsy or on histologic examination. The brains of the five bats showed strong immunohistochemical labeling for rabies virus in the form of large corpuscles or multiple clusters of granules within the perikarya of neurons in different areas of the brain, ma

Development of a reference artificial sediment for chemical testing adapted to the MELA sediment contact assay.

Most persistent organic pollutants, due to their hydrophobic properties, accumulate in aquatic sediments and represent a high risk for sediment quality. To assess the toxicity of hydrophobic pollutants, a novel approach was recently proposed as an alternative to replace, refine and reduce animal experimentation: the medaka embryo-larval sediment contact assay (MELAc). This assay is performed with Japanese medaka embryos incubated on a natural sediment spiked with the compound being tested. With the aim of improving this assay, our study developed a reference exposure protocol with an artificia

Attraction to herbivore-induced plant volatiles by the host-foraging parasitoid fly Exorista japonica.

Responses of the tachinid fly Exorista japonica Townsend to odors from corn plants infested with the fly's host, the larvae of the noctuid moth Mythimna separata (Walker), were examined in a wind tunnel. Naïve female flies showed a higher rate of landing on M. separata-infested corn plants from which the host larvae had been removed than on artificially damaged or intact corn plants. When paper impregnated with a solution of headspace volatiles collected from host-infested plants was attached to intact plants, females landed on the plants at a high rate. Females also responded to intact plants

Use of bioassay and microplate assay to detect and measure insecticide resistance in field populations of Culex pipiens from filariasis endemic areas of Egypt.

Insecticide and resistance bioassays and microplate assays were performed on Culex pipiens mosquitoes to determine the level and mechanisms of resistance. Culex pipiens larvae were collected from three filariasis-endemic areas of Egypt and reared to adults for subsequent production and testing of F1 generation larvae and adults. Bioassays were performed using World Health Organization (WHO) methods with the diagnostic doses of 6 organophosphate insecticides for larvae and 1 organochlorine (OC), 4 pyrethroid, 2 organophosphate, and 2 carbamate insecticides for adults. Microplate assays were per

Transport of Schistosoma japonicum cercariae and the feasibility of niclosamide for cercariae control.

Field experiments were conducted in southwestern China to estimate the distances over which cercariae of Schistosoma japonicum are transported and remain infective in village irrigation ditches and to determine the feasibility of interrupting this transport using niclosamide, the molluscicide commonly used for snail control in this environment. The number of worms recovered from bioassay mice downstream from experimentally introduced cercariae, but without niclosamide, decreased about 90% over the first 100 m of ditch and then remained essentially constant for the remaining 300 m of the ditch

Biotoxicity of mercury as influenced by mercury(II) speciation.

Integration of physicochemical procedures for studying mercury(II) speciation with microbiological procedures for studying the effects of mercury on bacterial growth allows evaluation of ionic factors (e.g., pH and ligand species and concentration) which affect biotoxicity. A Pseudomonas fluorescens strain capable of methylating inorganic Hg(II) was isolated from sediment samples collected at Buffalo Pound Lake in Saskatchewan, Canada. The effect of pH and ligand species on the toxic response (i.e., 50% inhibitory concentration [IC50]) of the P. fluorescens isolated to mercury were determined
