Transgenerational effects on intestinal inflammation status in mice perinatally exposed to bisphenol S.

Increasing evidence has highlighted the critical role of early life environment in shaping the future health outcomes of individuals in subsequent generations. Bisphenol S (BPS) has been widely used as a substitute for various plastic materials due to the limited application of Bisphenol A (BPA) which is an endocrine disruptor. However, the lack of efficient evaluation of BPS leaves doubts about the relevant substitute of BPA. Few studies of transgenerational inheritance have examined the effects of environmental exposures to endocrine disruptors on the immune system. In this study, we analyze

Developmental programming: Sex-specific programming of growth upon prenatal bisphenol A exposure.

In both human and animals, in utero exposure to bisphenol A (BPA), an endocrine-disrupting chemical used in the production of plastics and epoxy resins, has been shown to affect offspring reproductive and metabolic health during adult life. We hypothesized that the effect of prenatal exposure to environmentally relevant doses of BPA will be evident during fetal organogenesis and fetal/postnatal growth trajectory. Pregnant ewes were administered BPA subcutaneously from 30 to 90 days of gestation (term 147 days). Fetal organ weight, anthropometric measures, maternal/fetal hormones and postnatal

Fast and Reversible "Turn on" Fluorescent Sensors Based on Bisphenol-a for Zn2+ in Aqueous Solution.

Two novel bisphenol-A derivatives (R1 and R2) linked pyrene and napthylthiazole moieties were synthesized via condensation reaction, and positively applied for the selective recognition of Zn2+ ion in EtOH/H2O. Their optical properties were observed by using UV-vis and fluorescence measurements. R1 and R2 exhibited high selectivity and sensitivity towards Zn2+ over other metal ions. This fluorescence selectivity may be owing to inhibited excited-state intramolecular proton transfer (ESIPT) and photoinduced electron transfer (PET). The fluorescence titration analysis indicated detection limits

The ameliorative effect of quercetin on bisphenol A-induced toxicity in mitochondria isolated from rats.

Recent studies have demonstrated that bisphenol A (BPA) has an adverse or toxic effect on the kidney. This study was designed to evaluate the ability of quercetin (QUER) to prevent BPA-induced mitochondrial dysfunction. Thirty-two healthy adult male Wistar rats were randomly divided into four groups, as follows: control group (olive oil), BPA group (250 mg/kg), BPA þ QUER group (250 mg/kg + 75 mg/kg), and QUER group (75 mg/kg). All treatments were orally administered for 14 days. Kidney mitochondria were isolated by administration of the different centrifugation method. Uric acid and creatinin

Genetic evidence for estrogenicity of bisphenol A in zebrafish gonadal differentiation and its signalling mechanism.

Bisphenol A (BPA) can induce endocrine disorders in humans and animals. In this study, we used several zebrafish mutants deficient in estrogen production and signalling, which could be valuable for evaluating estrogenic activities and mechanisms of EDCs. With low endogenous estrogens, the all-male aromatase mutant (cyp19a1a-/-) is expected to be more responsive to estrogenic exposure, and mutants of nuclear estrogen receptors (nERs; esr1-/-, esr2a-/- and esr2b-/-) alone or in combination would allow us to evaluate the action mechanisms of estrogenic EDCs. Exposure to BPA could rescue the all-m

Interactive effects of nonylphenol and bisphenol A exposure with oxidative stress on fetal reproductive indices.

Nonylphenol (NP) and/or bisphenol A (BPA) may have reproductive effects. Although the mechanisms of action remain unclear, steroid hormones biosynthesis, hypothalamus pituitary adrenal axis activity, oxidative stress, and crosstalk interaction of NP and BPA mixture and its pathways may play a contributory role. This cross-sectional study examined whether the interactive effects of NP/BPA and oxidative stress biomarkers played a role in reproductive indices (penis length and anogenital distance (AGD)) in 244 mother-fetus pairs. Four biomarkers of oxidative stress, (8-hydroxy-2'-deoxyguanosine (

Characterization of free and immobilized laccase from Cyberlindnera fabianii and application in degradation of bisphenol A.

Recovery difficulty and inactivation of laccases are major challenges that hamper their application in biotechnology. In this study, laccase was purified from Cyberlindnera fabianii using ion-exchange and gel filtration chromatography with homogeneity confirmed by sodium dodecyl sulphate polyacrylamide gel electrophoresis. Purified laccase of 52 kDa was immobilized on calcium and copper alginate beads by entrapment method. Free and immobilized enzymes were characterized, and efficiency of bisphenol A (BPA) degradation was determined. pH optima for free and immobilized laccases were 5.0 and 6.0

Inhibition of L-type calcium channels by Bisphenol A in rat aorta smooth muscle.

Bisphenol A (BPA) is an endocrine disrupting chemical used on a wide range in industry. This compound has been used in the production of polycarbonate plastics and epoxy resins. For this reason and their global use, BPA is one of the most common environmental chemicals to which humans are exposed. This exposure can cause several adverse health outcomes, including at the cardiovascular level. The regulation of ion channels in vascular smooth muscle is pivotal and important for vasoreactivity, and changes in their flux can be involved in the pathophysiology of some cardiovascular diseases. This

Melatonin ameliorates bisphenol A-induced perturbations of the prostate gland of adult Wistar rats.

Bisphenol A (BPA) is an endocrine disrupting chemical (EDC) that has been demonstrated to induce alterations in reproductive organs while melatonin (ML), an antioxidant, present in plants and animals, is capable of protecting against EDC-induced alterations. Adult male Wistar rats (average weight, 240 + 10 g) were divided into four groups of ten animals each: Rats in group I (control) received oral 0.2 ml 1% dimethyl sulfoxide (DMSO)/99% canola oil as vehicle; group II received intra-peritoneal 10 mg/kg BW/day ML. Group III received oral BPA dissolved in DMSO and solubilized in canola oil at 1

Low-dose effect of developmental bisphenol A exposure on sperm count and behaviour in rats.

Bisphenol A is widely used in food contact materials and other products and is detected in human urine and blood. Bisphenol A may affect reproductive and neurological development; however, opinion of the European Food Safety Authority (EFSA) on bisphenol A (EFSA J, 13, 2015 and 3978) concluded that none of the available studies were robust enough to provide a point of departure for setting a tolerable daily intake for bisphenol A. In the present study, pregnant Wistar rats (n = 17-21) were gavaged from gestation day 7 to pup day 22 with bisphenol A doses of 0, 25 μg, 250 μg, 5 mg or 50 mg/kg b

Bisphenol S Induces Adipogenesis in Primary Human Preadipocytes From Female Donors.

Human exposure to bisphenol A has been associated with negative health outcomes in humans and its use is now regulated in a number of countries. Bisphenol S (BPS) is increasingly used as a replacement for bisphenol A; however, its effects on cellular metabolism and potential role as an endocrine disruptor have not been fully characterized. In the current study, we evaluated the effect of BPS on adipogenesis in primary human preadipocytes. The effect of BPS on the differentiation of human preadipocytes was determined after treatment with BPS at concentrations ranging from 0.1 nM to 25 μM by qua

Prenatal exposure to bisphenol A alters mouse fetal pancreatic morphology and islet composition.

BACKGROUND: Exposure to bisphenol A (BPA), an endocrine disrupting chemical, during gestation is associated with a variety of metabolic dysfunctions in adulthood, including hyperinsulinemia, glucose intolerance and insulin resistance. These modifications in glucose homeostasis largely stem from alterations in pancreatic function. However, the effects of BPA on the fetal pancreas have never been explored. The present study addressed this important question by examining the effects of prenatal BPA exposure on the mouse fetal pancreatic development. MATERIALS AND METHODS: Pregnant mice were fed

Maternal transfer of bisphenol A impacts the ontogeny of cortisol stress response in rainbow trout.

Bisphenol A (BPA) is widespread in the aquatic environment, and early life-stage exposure to this chemical affects growth and development in fish. We tested the hypothesis that BPA accumulation in eggs, mimicking maternal transfer, impacts the development and function of the hypothalamus-pituitary-interrenal (HPI) axis in rainbow trout (Oncorhynchus mykiss). To mimic maternal transfer, oocytes were loaded with 0 (control), 1, 4 and 40ng BPA embryo(-1). We measured the temporal changes in whole body cortisol content, transcript levels of corticosteroidogenesis-related genes and glucocorticoid r

Parental urinary biomarkers of preconception exposure to bisphenol A and phthalates in relation to birth outcomes.

BACKGROUND: Bisphenol A (BPA) and phthalates are ubiquitous non-persistent endocrine disrupting chemicals whose relation with infant birth size is not clearly understood. METHODS: We examined associations between maternal and paternal preconception urinary concentrations of total BPA and 14 phthalate metabolites and birth size for 233 infants. Multiple linear regression models were used to estimate parental quartiles of BPA and phthalates in relation to birth weight, length, head circumference, and ponderal index with separate models run for each parent adjusting for age, smoking, body mass i

Effect of bisphenol A on Drosophila melanogaster behavior--a new model for the studies on neurodevelopmental disorders.

Developmental disorders such as autism and attention deficit hyperactivity disorder (ADHD) appear to have a complex etiology implicating both genetic and environmental factors. Bisphenol A (BPA), a widely used chemical in the plastic containers and in the linings of food and beverage cans, has been suggested to play a possible causative role in some developmental disorders. Here, we report behavioral modifications in Drosophila melanogaster following early exposure to BPA, which may suggest BPA as an environmental risk factor for the behavioral impairments that are the basis of diagnosis of au

Operation of fill-and-draw reactors fed with bisphenol A as sole carbon and energy source.

While there is ample evidence in the literature that many organic xenobiotics can be biodegraded as sole carbon and energy source by pure batch cultures of selected microorganisms, the same evidence is very limited for continuous or semi-continuous mixed-culture processes simulating biological wastewater treatment plants. This study investigates for the first time whether Bisphenol A (BPA) can be removed and used as sole carbon and energy source by mixed microbial cultures in a semi-continuous process. Four fill and draw bioreactors were inoculated with unacclimated soil and operated at variou
