Calexcitin: a signaling protein that binds calcium and GTP, inhibits potassium channels, and enhances membrane excitability.

A previously uncharacterized 22-kDa Ca(2+)-binding protein that also binds guanosine nucleotides was characterized, cloned, and analyzed by electrophysiological techniques. The cloned protein, calexcitin, contains two EF-hands and also has homology with GTP-binding proteins in the ADP ribosylation factor family. In addition to binding two molecules of Ca2+, calexcitin bound GTP and possessed GTPase activity. Calexictin is also a high affinity substrate for protein kinase C. Application of calexcitin to the inner surface of inside-out patches of human fibroblast membranes, in the presence of Ca

Effector Fg34 triggers TaHRC-R-mediated calcium signaling to bolster Fusarium head blight resistance in wheat.

Fusarium head blight, one of the most devastating diseases affecting wheat, is primarily caused by Fusarium graminearum. The TaHRC gene (als … Fusarium-derived effector. The TaHRC-R/Fg34 complex elevates intracellular Ca2+ levels, activating calcium signaling pathways that reinforce Fusarium head blight resistance. Furthermore, Fg34 also binds to TaCBL4, forming a ternary complex with TaCIPK5 that enhances TaCIPK5 kinase activity, leading to increased phosphorylation of TaRBOHB. Collectively, these findi …

Direct targeting of ORAI1 by ginsenoside Rg3 modulates calcium signaling and senescence-associated AMPK-NRF2 activation.

BACKGROUND: 20(S)-ginsenoside Rg3 (Rg3(S)), a major saponin derived from red ginseng, exhibits diverse biological activities, including antioxidant and anti-senescence effects. However, the direct molecular targets through which Rg3(S) regulates calcium signaling and its role at membrane contact sites during cellular senescence remain largely unknown. METHODS: Plasma membrane (PM)-endoplasmic reticulum (ER) contacts and related protein interactions were analyzed using proximity ligation assays and co-immunoprecipitation. Direct binding of Rg3(S) to ORAI1 was validated using cellular thermal s…

TRPM7-mediated calcium signaling contributes to Hyperglycemia-induced mitochondrial dysfunction and apoptosis in retinal Müller cells.

Calcium signaling dysregulation is a critical trigger of mitochondrial dysfunction in metabolic disorders, yet the upstream mechanisms linking hyperglycemic stress to organellar Ca2+ overload remain poorly defined. The transient receptor potential melastatin 7 (TRPM7) channel functions as a Ca2+-permeable signaling node with unique kinase activity, but its role in hyperglycemia-induced glial injury is unknown. Here, we investigated whether TRPM7 mediates mitochondrial dysfunction and apoptosis in retinal Müller cells under hyperglycemic stress. Using a streptozotocin/high-fat diet-induced diab…

Mediodorsal thalamus of alcohol-dependent mice shows genetic and physiological adaptations and alcohol-biased calcium signaling.

Alcohol Use Disorder (AUD) is a significant health concern characterized by cognitive dysfunction and an inability to control alcohol intake, leading to severe social and health consequences. It is crucial to uncover neuroadaptations and cellular mechanisms responsible for poor decisions surrounding alcohol drinking. Although the mediodorsal thalamus (MD) is an essential brain region for cognitive function and reward-guided choices, the effects of alcohol dependence on MD neuroadaptations and how dependence alters MD activity during choice behaviors for alcohol over natural rewards (i.e., sucr

6-Hydroxyflavone Attenuates Inflammatory Osteolysis by Inhibiting Osteoclast Activation via the Nrf2 and Calcium Signaling Pathways.

Inflammatory osteolysis (IO) is a pathological bone disorder characterized by excessive osteoclast activation and bone resorption driven by inflammatory mediators and oxidative stress. 6-Hydroxyflavone (6-HOF), a natural flavonoid with potent antioxidant and anti-inflammatory properties, has not been fully investigated in the context of IO. In this study, we explored the therapeutic effects and underlying mechanisms of 6-HOF in LPS-induced osteoclastogenesis and inflammatory bone loss. Network pharmacology analysis predicted that 6-HOF primarily targets oxidative stress and calcium signaling p…

TLR-mediated calcium signaling regulates killing capacity of expanded NK cells: challenges for potential AML immunotherapy.

Natural killer (NK) cells are central to cancer immunosurveillance and immunotherapy. Their ability to engage in killing is critical for efficient target elimination and depends on tightly regulated Ca2+ signaling controlling granzyme degranulation. Toll-like receptor (TLR) engagement has been linked to Ca2+ signaling in other immune cell types, while a specific role in NK cells remains unresolved. Given that NK cells express a broad range of TLRs, and that presence of TLR ligands-including damage- and pathogen-associated molecular patterns-can influence clinical scenarios including adoptive N

From calcium signaling to cardiovascular pharmacology: TRPM7 as a key mechanistic regulator and emerging pharmacological focus.

Cardiovascular diseases remain the leading global cause of death and disability, prompting the need for precise mapping of molecular drivers that couple ionic flux to signaling. Because of its regulatory role in ionic balance and signaling, transient receptor potential melastatin 7 (TRPM7) channel-kinase has emerged as a key molecular determinant of cardiovascular physiology and pathophysiology. Following an overview of Ca2+ signaling in cardiovascular biology, we summarize current knowledge of TRPM7 from its structure to its function, including channel architecture, gating properties, and kin

Feeder cell-free production of CAR-NK cells via activation of PKC and calcium signaling pathways.

BACKGROUND: Ex vivo manufacturing of CAR-NK cells is a complex, multistep process involving NK cell activation, genetic modification, and cellular expansion. Currently, tumor cell-derived feeder cells (e.g., K562) are commonly used to manufacture CAR-NK cells. However, the feeder cell-based (FCB) method presents several risks to the CAR-NK product, including contamination, residual impurities, immunogenicity, and lot-to-lot variability. Additionally, establishing and maintaining feeder cells increases costs, complicates scaling up production, and requires rigorous quality control to ensure saf

Grancalcin Hydrogel Microneedle Patches Alleviate Sepsis via Modulation of Calcium Signaling to Augment Immune Cell Phagocytosis.

Disorders in Ca2+ signaling contribute to many metabolic manifestations of sepsis and are one of the driving forces underlying multiorgan failure. Herein, a compensatory elevation in calcium-binding protein grancalcin (GCA) levels in monocytes and macrophages of patients and mice with sepsis is observed. Gca deletion in myeloid cells displays increased inflammation and organ damage in cecal ligation and puncture-induced sepsis. Mechanistically, GCA enhances the phagocytic function of immune cells by regulating intracellular calcium signaling. A GCA@Acrylate-PEG-NHS/weakly temperature-sensitive…

Induction of systemic resistance through calcium signaling in Arabidopsis exposed to air plasma-generated dinitrogen pentoxide.

Plasma technology, which can instantaneously transform air molecules into reactive species stimulating plants, potentially contributes to developing a sustainable agricultural system with high productivity and low environmental impact. In fact, plant immunity activation by exposure to a reactive gas mainly consisting of dinitrogen pentoxide (N2O5) was recently discovered, while physiological responses to N2O5 are rarely known. Here, we demonstrate early (within 10 min) physiological responses to N2O5 gas in Arabidopsis. Exposure to N2O5 gas induced an increase in cytosolic Ca2+ concentration w

Transcriptional reprogramming in SMA mouse hearts reveals signatures of early heart failure and dysregulated calcium signaling.

Spinal muscular atrophy (SMA) is an inherited neurodegenerative disease that leads to loss of motor neurons in the anterior horn of the spinal cord with consequent muscle atrophy. SMA results from the functional deletions of the SMN1 gene, resulting in insufficient production of the survival motor neuron (SMN) protein. It is not known why lower motor neurons are particularly sensitive to the loss of SMN function, but it is increasingly apparent that extraneuronal tissues, such as cardiac and skeletal muscle, are also affected by SMN deficiency. We have previously shown that SMN deficiency in a

Age-related calcium signaling disturbance restricted cAMP metabolism and induced ovarian oxidation stress in laying ducks.

The ovary is the main controller of female fertility, unfortunately, its onset of aging processes was earlier than other organs. Our previous studies showed calcium (Ca) deficiency reduced ovarian weight and declined numbers of dominant follicles in an avian model. However, whether Ca provided a functional role in follicle development of aged avian, and its further mechanism was still unknown. In this study, fifty180-day-old and fifty 700-day-old female Longyan ducks were divided into the young group and the aged group to illustrate the differences of Ca signaling and further mechanisms. We fo

Lead and calcium crosstalk tempted acrosome damage and hyperpolarization of spermatozoa: signaling and ultra-structural evidences.

BACKGROUND: Exposure of humans and animals to heavy metals is increasing day-by-day; thus, lead even today remains of significant public health concern. According to CDC, blood lead reference value (BLRV) ranges from 3.5 µg/dl to 5 μg/dl in adults. Recently, almost 2.6% decline in male fertility per year has been reported but the cause is not well established. Lead (Pb2+) affects the size of testis, semen quality, and secretory functions of prostate. But the molecular mechanism(s) of lead toxicity in sperm cells is not clear. Thus, present study was undertaken to evaluate the adverse effects o

Thapsigargin and its prodrug derivatives: exploring novel approaches for targeted cancer therapy through calcium signaling disruption.

Thapsigargin, a sesquiterpene lactone derived from Thapsia garganica L., has demonstrated mixed potential as an anticancer agent due to its potent ability to disrupt calcium signaling and induce apoptosis. This review evaluates the chemopreventive and chemotherapeutic potential of thapsigargin, focusing on its molecular mechanisms and toxicity. An extensive literature review of studies published since 2015 was conducted using databases such as PubMed/MedLine and Science Direct. Findings indicate that thapsigargin's primary mechanism is the inhibition of sarco/endoplasmic reticulum calcium ATPa…

The SOX4/EZH2/SLC7A11 signaling axis mediates ferroptosis in calcium oxalate crystal deposition-induced kidney injury.

Epigenetic regulation is reported to play a significant role in the pathogenesis of various kidney diseases, including renal cell carcinoma, acute kidney injury, renal fibrosis, diabetic nephropathy, and lupus nephritis. However, the role of epigenetic regulation in calcium oxalate (CaOx) crystal deposition-induced kidney injury remains unclear. Our study demonstrated that the upregulation of enhancer of zeste homolog 2 (EZH2)-mediated ferroptosis facilitates CaOx-induced kidney injury. CaOx crystal deposition promoted ferroptosis in vivo and in vitro. Usage of liproxstatin-1 (Lip-1), a ferrop

Nicotinic Acid Adenine Dinucleotide Phosphate (NAADP)-Mediated Calcium Signaling Is Active in Memory CD4+ T Cells.

Nicotinic acid adenine dinucleotide phosphate (NAADP), identified as one of the most potent calcium-mobilizing second messengers, has been studied in different eukaryotic cell types, including lymphocytes. Although aspects of NAADP-mediated calcium release in lymphocytes are still under debate, the organelles pertaining to NAADP-mediated calcium release are often characterized as acidic and related to lysosomes. Although NAADP-mediated calcium release in different subsets of T cells, including naïve, effector and natural regulatory T cells, has been studied, it has not been widely studied in m
