Tissue-specific dysregulation of S100A8 in synovium and cartilage reveals limitations of NSAID therapy in inflammatory arthritis.

Inflammatory arthritis involves complex immune responses in both synovial and cartilage tissues, yet the mechanisms underlying tissue-specific inflammatory regulation remain unclear. Using a carrageenan-induced rat arthritis model, we found that synovium and cartilage displayed distinct transcriptional responses, with synovium showing broader immune activation. Gene ontology and KEGG analyses revealed enrichment of inflammatory and immune pathways in both tissues, more prominently in synovium. Cross-species validation using peripheral leukocyte transcriptomes from rheumatoid arthritis (RA) pat

S100A8 regulated by estrogen improves injured endometrial epithelium reconstruction by promoting tight junction formation and stromal cell transformation.

Estrogen is used for endometrial repair; however, it has limited effectiveness. Cytokine S100A8 expression is affected by estrogen and is involved in regulating the damage repair. We aimed to confirm the effects of S100A8 and explore its mechanisms during the reconstruction of the injured endometrium. We investigated the effects of estrogen on S100A8 expression in healthy endometrium. A rat model of endometrial injury and cultured primary endometrial cells were used to determine the pleiotropic effects of S100A8 on endometrial epithelial repair. Estrogen regulated the recruitment of S100A8-pos

Proteomics of plasma-derived extracellular vesicles reveals S100A8 as a novel biomarker for Alzheimer's disease: A preliminary study.

Extracellular vesicles (EVs) act as mediators for intercellular transfer of Aβ and tau proteins, promoting the propagation of these pathological misfolded proteins throughout the brain in Alzheimer's disease (AD). Levels of blood exosomal Aβ42, total Tau (t-Tau) and phosphorylated Tau (p-Tau) had a high correlation with their concentrations in cerebrospinal fluid (CSF), demonstrating that exosomal biomarkers have equal contribution as those in CSF for the diagnosis of AD. We aimed to comprehensively characterize the proteome of plasma-derived EVs to identify differentially expressed proteins (

Translocation of intrauterine-infused bacterial lipopolysaccharides to the mammary gland in dexamethasone-treated goats.

Our previous study showed that intrauterine-infused lipopolysaccharide (LPS) can be translocated to the mammary gland to induce weak inflammation. This study aimed to determine whether dexamethasone treatment facilitated the translocation of LPS from the uterus to the mammary gland to induce a heavy inflammatory response. Sixteen goats were divided into control and LPS groups, subjected to daily dexamethasone administration before saline or LPS infusion. Milk and blood samples were collected before and after LPS infusion to determine the milk yield and somatic cell count (SCC) and blood leucoc

Alterations of circulating endogenous secretory RAGE and S100A9 levels indicating dysfunction of the AGE-RAGE axis in autism.

An excess accumulation of advanced glycation end products (AGEs) has been reported in autism brains. Through their interaction with their putative receptor RAGE, AGEs can promote neuroinflammation, oxidative stress and neuronal degeneration. To shed more light on the possible alterations of the AGEs-RAGE axis in autism, hereto we measured plasma levels of endogenous secretory RAGE (esRAGE) and its proinflammatory ligand S100A9 in 18 young adults with autistic spectrum disorder (ASD) and 18 age- and gender-matched healthy comparison subjects. The Childhood Autism Rating Scale (CARS) was used to

CKD disrupts cardiac energy metabolism and aggravates cardiac inflammation, oxidative stress, and dysfunction post-infarction.

AIMS: Patients with chronic kidney disease (CKD) display a reduced survival following myocardial infarction (MI). As the underlying mechanisms remain unclear, we examined the impact of CKD on cardiac remodeling and function post-MI using a mouse model of adenine-induced CKD. METHODS AND RESULTS: After MI, CKD mice showed a stronger cardiac dysfunction compared to non-CKD controls. While immunohistochemical and immunofluorescence analyses did not reveal changes in cardiomyocyte apoptosis, infarction size, or myofibroblast content, CKD mice exhibited an increased number of circulating myeloid c

The Innate Immune Protein S100A9 Protects from T-Helper Cell Type 2-mediated Allergic Airway Inflammation.

Calprotectin is a heterodimer of the proteins S100A8 and S100A9, and it is an abundant innate immune protein associated with inflammation. In humans, calprotectin transcription and protein abundance are associated with asthma and disease severity. However, mechanistic studies in experimental asthma models have been inconclusive, identifying both protective and pathogenic effects of calprotectin. To clarify the role of calprotectin in asthma, calprotectin-deficient S100A9-/- and wild-type (WT) C57BL/6 mice were compared in a murine model of allergic airway inflammation. Mice were intranasally c

The Quest for Antiinflammatory and Immunomodulatory Strategies in Heart Failure.

Intensive research over the last 3 decades has unequivocally demonstrated the relevance of inflammation in heart failure (HF). Despite our current and ever increasing knowledge about inflammation, the clinical success of antiinflammatory and immunomodulatory therapies in HF is still limited. This review outlines the complexity and diversity of inflammation, its reciprocal interaction with HF, and addresses future perspectives, calling for immunomodulatory therapies that are specific for factors that activate the immune system without the risk of nonspecific immune suppression.

Myeloid related protein induces muscle derived inflammatory mediators in juvenile dermatomyositis.

INTRODUCTION: The aetiopathogenesis of juvenile dermatomyositis (JDM) remains poorly understood. In particular the contribution of monocytes or macrophages, which are frequently observed to be an infiltrate within muscle tissue very early in the disease process, is unknown. We hypothesised that these cells secrete the pro-inflammatory myeloid related protein (MRP) 8/14 which may then contribute to muscle pathology in JDM. METHODS: In this study of 56 JDM patients, serum MRP8/14 levels were compared with clinical measures of disease activity. Muscle biopsies taken early in disease were assesse

Brief episode of ischemia activates protective genetic program in rat heart: a gene chip study.

OBJECTIVE: Brief episodes of ischemia of 20 min or less have the potential to protect the heart. Such episodes are associated primarily with reversible ischemic injury yet they induce changes in gene expression. The purpose of the study was to determine whether activation of protective genes takes place within 4 h following a brief episode of ischemia that would mimic angina pectoris. METHODS: Three groups of rats were studied. In the control (Ctrl) group, hearts were immediately excised following anesthesia; in the sham-operated (SO) group, opened-chest rats received 4 h and 20 min of no int

Deproteinized bovine bone and gentamicin as an adjunct to GTR in the treatment of intrabony defects: a randomized controlled clinical study.

PURPOSE: Pterygium is a vision-impairing fibrovascular lesion that grows across the corneal surface and is associated with sunlight exposure. To increase our understanding of the cells types involved in pterygium, we have used expressed sequence tag analysis to examine the transcriptional repertoire of isolated pterygium and to identify marker genes for tissue origin and cell migration. METHODS: An unnormalized unamplified cDNA library was prepared from 15 pooled specimens of surgically removed pterygia as part of the NEIBank project. Gene expression patterns were compared with existing data

A TLR2/S100A9/CXCL-2 signaling network is necessary for neutrophil recruitment in acute and chronic liver injury in the mouse.

BACKGROUND & AIMS: Neutrophils are important immune effectors required for sterile and non-sterile inflammatory responses. However, neutrophils are associated with pathology in drug-induced liver injury, acute alcoholic liver disease, and ischemia-reperfusion injury. An understanding of the complex mechanisms that control neutrophil recruitment to the injured liver is desirable for developing strategies aimed at limiting neutrophil-mediated cellular damage. METHODS: Wt, tlr2(-/-), tlr4(-/-), and s100a9(-/-) mice were administered CCl4 either acutely (8, 24, 48, or 72 h) or chronically (8 week

SPP1+ macrophage-driven interactions shape the tumor microenvironment in lymph node metastatic acral melanoma.

Melanoma is a highly aggressive skin cancer with biologically distinct subtypes. Acral melanoma (AM), a rare but particularly aggressive form, often presents with lymph node (LN) metastasis at diagnosis. Despite its clinical severity, the mechanisms underlying its metastatic behavior remain unclear. Emerging studies suggest the tumor microenvironment as a key driver of metastatic niche formation, but its specific role in AM progression is not well characterized. To investigate the role of the tumor microenvironment in AM progression, we performed single-cell RNA sequencing (scRNA-seq) on tumor

Neutrophil S100a9 Deficiency Protects against Periodontal Bone Loss.

Periodontitis is an inflammatory disease driven by microbial dysbiosis, characterized by immune dysregulation and alveolar bone resorption. Although upregulation of S100A8/A9 levels was found in human periodontitis, its mechanism of regulating osteoimmunological homeostasis during periodontitis progression remains elusive. To investigate this, we constructed a ligature-induced periodontitis model using S100a9 knockout mice. Further studies showed that S100a9 knockout mice exhibited impaired neutrophil extracellular trap (NET) formation and ameliorated bone resorption. Mechanistic studies demon

Mitochondrial Proteomics Uncovers that Myeloid S100A9 Deficiency Promotes Liver Injury and Fibrosis Regression via Mitochondrial Metabolic Reprogramming of Macrophages.

Macrophage-mediated inflammation has been implicated in the pathogenesis of liver fibrosis. Metabolic reprogramming involves the transition of macrophages from a proinflammatory M1 phenotype toward an anti-inflammatory M2 phenotype. S100A9 is highly expressed in activated macrophages and promotes M1 polarization; however, it is unknown whether S100A9 alters the polarization state of macrophages by regulating metabolism. This study revealed enhanced oxidative phosphorylation (OXPHOS) in S100a9-deficient bone marrow-derived macrophages (BMDMs) based on mitochondrial proteomics, characterized by

CCR1+ monocytes facilitating bronchopulmonary dysplasia through regulation of S100A8 and MMP8.

Bronchopulmonary dysplasia (BPD) is the most common chronic lung disease in preterm infants and is characterized by aberrant late-stage lung development driven by multiple factors. Although previous studies have demonstrated that BPD is associated with prenatal and postnatal inflammatory stimuli, the inflammatory mechanisms involved immune cell populations have not been fully elucidated. Here, we systematically investigated the roles of myeloid cells in the inflammatory processes underlying BPD. Using single-cell transcriptomic data of normal and BPD mouse lung, we found that the proportions o

S100A9 promotes inflammasome-dependent autoinflammation by blocking the degradation of SYK tyrosine kinase.

Autoinflammatory diseases such as cryopyrin-associated periodic fever syndrome and familial Mediterranean fever involve an aberrant secretion of interleukin-1β due to genetic defects in the NLRP3 (NOD-, LRR-, and pyrin domain-containing protein 3) or pyrin inflammasomes. The regulatory mechanisms and possible interactions between inflammasome pathways remain unclear. In addition, these conditions show a high expression of the inflammatory alarmins S100A8/A9. Spleen tyrosine kinase is a known regulator of NLRP3 activity, but its connection to S100 proteins and pyrin-driven inflammation has not
