Combined exposure to atrazine and phoxim exacerbated the alterations of enzyme activity and abnormal gene expression in earthworms (Eisenia fetida).

Multiple pesticides often coexist in soil, potentially leading to interactions among their components, these may detrimentally impact soil o … oxidative damage. Elevated expression levels of translationally controlled tumor protein (tctp) and calreticulin (crt) genes were observed in most exposed groups compared to the control. The synergistic effects of ATR and PHO on earthworms observed in this study may pose ecological risks to the soil ecosystem; thus, more attention should be paid to the joint e …

Sunitinib induces immunogenic cell death through eIF2α phosphorylation to potentiate immunotherapy in HCC.

Hepatocellular carcinoma (HCC) prognosis remains poor due to its immunosuppressive tumor microenvironment. Inducing immunogenic cell death (ICD) represents a promising strategy to remodel this environment. Herein, we developed an ICD-related risk signature (ICDRS) to stratify patients and predict immunotherapy response. Through integrated database screening, sunitinib was repurposed as an ICD inducer. It elicited hallmark features of ICD, including calreticulin exposure, ATP release, and HMGB1 secretion, and effectively suppressed organoid growth. Sunitinib-pretreated HCC cells promoted dendri…

Boosting immunogenic tumour cell death via nanotherapeutic targeting of the Stanniocalcin 1 phagocytosis checkpoint for enhanced cancer immunotherapy.

Eliciting calreticulin (CRT) surface exposure is essential for triggering immunogenic cell death (ICD). However, stanniocalcin 1 (STC1) suppresses CRT translocation by sequestering it within mitochondria, limiting ICD induction in tumours. Here, we show that silencing STC1 enhances CRT surface exposure in Lewis lung carcinoma (LLC) cells when combined with paclitaxel (PTX), converting dying tumour cells into an in-situ vaccine that drives immunoprevention of tumour growth. To maximize this therapeutic synergy, we engineered a nanoplatform co-delivering siSTC1 and PTX, in which PTX is covalentl…

Investigation of the in vitro anticancer potential of bis(imino)acenaphthene-N-heterocyclic carbene transition metal complexes revealed TrxR inhibition and triggering of immunogenic cell death (ICD) for allyl palladates.

Immunogenic cell death (ICD) is a regulated form of cell death that activates an immune response through the release of danger-associated molecular patterns (DAMPs), including calreticulin, ATP, and HMGB1. Gold complexes are known to induce ICD, but the ICD-inducing potential of palladium complexes remains largely unexplored. We report the first examples of palladium compounds capable of inducing ICD, specifically allyl palladates bearing bis(imino)acenaphthene-NHC (BIAN-NHC) ligands. Cytotoxicity tests on human cancer cell lines revealed that allyl palladates outperform their cinnamyl analogu…

Philadelphia chromosome-negative myeloproliferative neoplasms: clinical aspects and treatment options.

Clinical studies of Philadelphia chromosome-negative myeloproliferative neoplasms (MPN) have progressed greatly with the discovery of mutations in three driver genes: JAK2, MPL, and calreticulin. Other genes that may play important roles in pathogenesis and progression of MPN have also been identified. Several prognostic prediction systems based on various risk factors including these genetic factors have been developed and utilized in clinical practice. All mutations of the three driver genes result in JAK2 activation, and JAK inhibitors have indeed improved clinical outcomes for primary myel…

The stem/progenitor landscape is reshaped in a mouse model of essential thrombocythemia and causes excess megakaryocyte production.

Frameshift mutations in CALR (calreticulin) are associated with essential thrombocythemia (ET), but the stages at and mechanisms by which mutant CALR drives transformation remain incompletely defined. Here, we use single-cell approaches to examine the hematopoietic stem/progenitor cell landscape in a mouse model of mutant CALR-driven ET. We identify a trajectory linking hematopoietic stem cells (HSCs) with megakaryocytes and prospectively identify a previously unknown intermediate population that is overrepresented in the disease state. We also show that mutant CALR drives transformation prima…

X chromosome inactivation analysis reveals a difference in the biology of ET patients with JAK2 and CALR mutations.

Calreticulin mutations (CALR(MUT)) are found in a significant proportion of patients with essential thrombocythemia (ET) lacking JAK2(V617F) or MPL mutations. They are associated with substantially different hematological and clinical features and define a distinct subtype of ET. We show here that their presence is significantly correlated with a clonal X chromosome inactivation pattern (XCIP). Of 105 female ET patients investigated, 61 had an interpretable XCIP, and a clonal pattern was observed in 88% of CALR(MUT) patients compared with 26% of JAK2(V617F) (P = .0002) and 9% of JAK2(V617F)/MP…

Endoplasmic Reticulum Chaperones in Viral Infection: Therapeutic Perspectives.

Viruses are intracellular parasites that subvert the functions of their host cells to accomplish their infection cycle. The endoplasmic reticulum (ER)-residing chaperone proteins are central for the achievement of different steps of the viral cycle, from entry and replication to assembly and exit. The most abundant ER chaperones are GRP78 (78-kDa glucose-regulated protein), GRP94 (94-kDa glucose-regulated protein), the carbohydrate or lectin-like chaperones calnexin (CNX) and calreticulin (CRT), the protein disulfide isomerases (PDIs), and the DNAJ chaperones. This review will focus on the ple…

Melatonin, hormone of darkness and more: occurrence, control mechanisms, actions and bioactive metabolites.

In its role as a pineal hormone, melatonin is a pleiotropic, nocturnally peaking and systemically acting chronobiotic. These effects are largely explained by actions via G protein-coupled membrane receptors found in the suprachiasmatic nucleus, but also in numerous other sites. Nuclear (ROR/RZR), cytoplasmic (quinone reductase-2, calmodulin, calreticulin) and mitochondrial binding sites and radical-scavenging properties contribute to the actions of melatonin. Regulation of pineal melatonin biosynthesis is largely explained by control mechanisms acting on arylalkylamine N-acetyltransferase, at …

Exercise inhibits doxorubicin-induced cardiotoxicity by preventing cardiac and endothelial cell senescence by activating FNDC5/Irisin signaling.

Exercise's effect on Dox-induced cardiac senescence in the heart was investigated using p16-3MR transgenic mice. Mice treated with doxorubicin (Dox) had decreased cardiac function, increased p16INK4a + and b-Galactosidase+ senescent cells in the heart, decreased HMGB1 and calreticulin expression, endothelial cell senescence, and decreased TUJ1 expression (indicating reduced axonal density), increased expression of Semaphorin3A and p-cofilin with impaired F-actin depolarization and cytoskeleton disorganization. None of these changes were seen in mice treated with Dox + Exercise. Exercise sustai…
