The nature of cancer initiation by fumonisin B(1) (FB(1)) was investigated in rat liver by monitoring the effect of phenobarbital (PB) as cancer promoter and evaluating the involvement of spontaneously initiated cells. A PB promoting regimen (0.05% in the diet) stimulated the outgrowth of FB(1)-induced placental glutathione S-transferase (GSTP) positive initiated hepatocytes. The antihistamine methapyrilene (MP) has been shown to be a potent hepatocarcinogen in rats. However, it has demonstrated little genotoxic activity in a wide variety of short-term tests. Experimental mutagenic effect of acrylamide on thyroid gland cells was studied by an extended micronucleus test. Acrylamide in doses corresponding to 0.004-0.1 LD(50) increased the incidence of thyroid follicular cells (A-cells) with micronuclei and other karyological parameters in exposed rats after hemithyroidectomy. This cytogenetic effect allows regarding acrylamide as a mutagen for the thyroid gland and as a carcinogen for this organ.

The inhibition of gap junctional intercellular communication (GJIC) is a common effect of nongenotoxic carcinogens and might be a biomarker for these agents.
