Effects of high hydrostatic pressure, ultraviolet light-C, and far-infrared treatments on the digestibility, antioxidant and antihypertensive activity of α-casein.

Alpha-casein is the most important bioactive protein in processing technologies. This study investigated the digestibility, antioxidant and antihypertensive activities of α-casein when treated by high hydrostatic pressure (HPP), ultraviolet light-C (UV-C), and far-infrared radiation (FIR). The in vitro digestibility was modified after treatments, especially after 5min/200MPa HHP treatment. Ultra-performance liquid chromatography-tandem mass spectrometry analysis showed that one 5min/200MPa HHP treatment resulted in the highest yield of peptides. Based on the in vitro gastrointestinal digestion

Influence of pigments and protein aging on protein identification in historically representative casein-based paints using enzyme-linked immunosorbent assay.

A systematic study on the influence of pigments and sample aging on casein identification was performed on 30 reconstructed paints. The protein in all the paints was extracted into solution for analysis. The amount of protein that can be retrieved for solution-based analysis in each of the reconstructed paints was studied with a well-developed NanoOrange method before and after artificial aging. The results showed that in the paints with calcium phosphate (in bone black) and copper carbonate, hydroxide, or acetate (in verdigris and azurite), the amount of protein that can be retrieved for liqu

Casein kinase 1delta activates human recombinant deoxycytidine kinase by Ser-74 phosphorylation, but is not involved in the in vivo regulation of its activity.

Deoxycytidine kinase (dCK) is a key enzyme in the salvage of deoxynucleosides and in the activation of several anticancer and antiviral nucleoside analogues. We recently showed that dCK was activated in vivo by phosphorylation of Ser-74. However, the protein kinase responsible was not identified. Ser-74 is located downstream a Glu-rich region, presenting similarity with the consensus phosphorylation motif of casein kinase 1 (CKI), and particularly of CKI delta. We showed that recombinant CKI delta phosphorylated several residues of bacterially overexpressed dCK: Ser-74, but also Ser-11, Ser-15

(-)-epigallocatechin-3-gallate (EGCG) maintains kappa-casein in its pre-fibrillar state without redirecting its aggregation pathway.

The polyphenol (-)-epigallocatechin-3-gallate (EGCG) has recently attracted much research interest in the field of protein-misfolding diseases because of its potent anti-amyloid activity against amyloid-beta, alpha-synuclein and huntingtin, the amyloid-fibril-forming proteins involved in Alzheimer's, Parkinson's and Huntington's diseases, respectively. EGCG redirects the aggregation of these polypeptides to a disordered off-folding pathway that results in the formation of non-toxic amorphous aggregates. Whether this anti-fibril activity is specific to these disease-related target proteins or i

Acid resistance of enamel subsurface lesions remineralized by a sugar-free chewing gum containing casein phosphopeptide-amorphous calcium phosphate.

The aim of this clinical study was to investigate the acid resistance of enamel lesions remineralized in situ by a sugar-free chewing gum containing casein phosphopeptide-amorphous calcium phosphate nanocomplexes (CPP-ACP: Recaldent). The study utilized a double-blind, randomized, crossover design with two treatments: (i) sugar-free gum containing 18.8 mg of CPP-ACP, and (ii) sugar-free gum not containing CPP-ACP as control. Subjects wore removable palatal appliances with insets of human enamel containing demineralized subsurface lesions and chewed the gum for 20 min 4 times per day for 14 day

Casein kinase I-dependent phosphorylation within a PEST sequence and ubiquitination at nearby lysines signal endocytosis of yeast uracil permease.

Uracil uptake by Saccharomyces cerevisiae is mediated by the FUR4-encoded uracil permease. The modification of uracil permease by phosphorylation at the plasma membrane is a key mechanism for regulating endocytosis of this protein. This modification in turn facilitates its ubiquitination and internalization. Following endocytosis, the permease is targeted to the lysosome/vacuole for proteolysis. We have previously shown that uracil permease is phosphorylated at several serine residues within a well characterized N-terminal PEST sequence. In this report, we provide evidence that lysine residues

Identification of casein kinase II phosphorylation sites in Max: effects on DNA-binding kinetics of Max homo- and Myc/Max heterodimers.

Myc proteins have been implicated in the regulation of cell growth and differentiation. The identification of Max, a basic region/helix-loop-helix/leucine zipper protein, as a partner for Myc has provided insights into Myc's molecular function as a transcription factor. Recent evidence indicates that the relative abundance of Myc and Max is important to determine the level of specific gene transcription. In this report we have identified two major in vivo phosphorylation sites in Max (Ser-2 and -11) which can be modified in vitro by casein kinase II (CKII). Phosphorylation of these sites modul

Beta-casein gene promoter activity is regulated by the hormone-mediated relief of transcriptional repression and a mammary-gland-specific nuclear factor.

Transcription from the beta-casein milk protein gene promoter is induced by the synergistic action of glucocorticoid and prolactin hormones in the murine mammary epithelial cell line, HC11. We analyzed the binding of nuclear proteins to the promoter and determined their binding sites. Site-directed mutagenesis was used to determine the function of nuclear factor binding. During lactogenic hormone induction of HC11 cells, the binding of two nuclear factors increased. The binding of two other nuclear factors, present in uninduced cells, decreased. The basal activity of the promoter could be incr

Peptide profiling of casein peptide reveals its anti-diabetic effects via DPP-IV inhibition and IRS1/PI3K/AKT signaling.

Type 2 diabetes mellitus (T2DM) is a growing global health burden, increasing demand for safe food-derived strategies for glycemic control. Casein-derived peptides have potential metabolic benefits, but their antidiabetic mechanisms remain unclear. In this study, casein peptide (CNP), enriched in low-molecular-weight peptides (<3 kDa), was investigated as the principal bioactive intervention for T2DM regulation. LC-MS/MS identified 880 peptides; among the 50 most abundant, all were predicted to inhibit DPP-IV, 26 to inhibit α-glucosidase, and one to inhibit α-amylase. CNP inhibited DPP-IV in v

A multifunctional injectable, adhesive and self-healing carboxymethyl cellulose/alginate hydrogel reinforced with clove oil-caseinate nanoparticles for diabetic wound healing.

Diabetic wound healing is frequently impaired by a persistently inflammatory wound microenvironment accompanied by impaired angiogenesis. Therefore, fabrication of advanced wound dressings to restore a favorable wound microenvironment is essential to accelerate the healing process. This study developed a carboxymethyl cellulose/sodium alginate (CMC/ALG) hydrogel reinforced with clove oil-loaded sodium caseinate nanoparticle (CO-CAS NPs) as a wound dressing with multifunctional properties for diabetic wound repair. Clove oil was extracted by hydro-distillation and characterized by GC-MS, reveal

Dental surface restoration using Ca-caseinate bio/nano colloids: Converged roughness parameters reveal heterogeneous tooth surface reactivity.

Dental erosion, the chemical dissolution of the tooth surface structures, is an increasing problem in the modern world due to growing trends toward acidic food consumption and can cause permanent dental hard tissue loss. A detailed understanding of the reactivity of dental surfaces to chemicals invading the oral cavity is necessary to devise effective countermeasures. We introduce the use of converged roughness parameters Sqconv, i.e., roughness parameters calculated at a reduced field of view, to reflect the critical surface building blocks controlling the reactivity, as measured by white lig

Comparative evaluation of remineralization and microhardness of demineralized primary enamel treated with casein phosphopeptide amorphous calcium phosphate and chitosan hydrogel: An in vitro study.

BACKGROUND: Dental caries is a chronic microbial disease, characterized by progressive enamel demineralization. Improved understanding of its pathogenesis has shifted dentistry toward minimally invasive biomimetic strategies such as Casein phosphopeptide-amorphous calcium phosphate (CPP-ACP). Chitosan, a natural polysaccharide with antibacterial and biomimetic mineralization properties, has recently emerging as a promising alternative. Hence, this study compared the remineralization efficacy of CPP-ACP and chitosan hydrogel on artificially induced enamel lesions on primary enamel. METHODOLOGY

Regulation of peptide hydrophobicity by self-assembly of casein peptides with ferulic acid to potentially reduce peptide bitterness and improve in vitro digestive activity.

The main purpose of this study was to investigate the structural properties, mechanism of action, bitter taste changes, and γ-aminobutyric acid transaminase (GABA-T) inhibitory activity of CH-FA complexes in vitro digestion by self-assembly of casein hydrolysate (CH) and ferulic acid (FA). Structural analysis revealed that CH-FA was formed spontaneously, driven primarily by hydrophobic interactions and hydrogen bonding. The electron tongue results demonstrated that the bitter value of CH was reduced by 30.89 %, which may be due to the improvement of the hydrophobic amino acid microenvironment

Milk Powder Formulations with Varying Casein to Whey Ratios and Calcium Addition: Physico-Chemical and Structural Properties and the Effect of Low-Frequency Ultrasound.

This study examined the effect of low-frequency ultrasound (20 kHz, 1 and 5 min) on the physiochemical and structural properties of milk powder formulations with varying casein to whey ratios (0:100, 60:40, and 50:50) and calcium addition (30 mM). The ultrasound treatment led to changes in particle size, with an initial increase in aggregation followed by fragmentation. Calcium addition resulted in looser packing, as evidenced by a decrease in both bulk and tapped densities. DSC analysis indicated that calcium addition stabilized the protein-lactose matrix by increasing the glass transition te

pH Cycling-Induced Formation of Chickpea Protein-Casein Dual-Protein Complexes: Interaction Mechanisms, Structure, Function, and Bioactive Molecule Protection.

Single proteins have certain limitations in terms of functional properties, while composite products combining plant and animal proteins offer a practical and sustainable strategy to address this issue. This study aims to establish a dual-protein complex, the pH cycling-induced chickpea protein (CP)-casein (CA) complex (pH-CP/CA), to load curcumin and enhance its functionality. Results showed that pH-CP/CA (CP/CA = 2:1, mass ratio) exhibited a smaller particle size and a higher absolute value of zeta potential. Hydrogen-bonding, electrostatic, and hydrophobic interactions drove the pH-CP/CA fo

A comparative study of the composition of microorganisms and metabolites in different β-casein genetic types of dairy cows based on metagenomics and non-targeted metabolomics.

β-Casein is the main component of cow's milk protein, with A1 and A2 β-casein being the most common. Of these, A1 β-casein hydrolysate produces BCM-7, which can cause lactose intolerance, while A2 β-casein milk is more gentle on the gut. However, there is limited research on the composition of rumen microbiota, metabolites, and host metabolites in different genotype cows using metagenomics and metabolomics. In this study, we used multi-omics analysis techniques to perform enrichment analysis of differential metabolites, identifying three key metabolic pathways in all three groups: Arachidonic

Alleviation effect of lutein Pickering emulsion formed by casein-dextran conjugates through Maillard reaction against blue light retinal degeneration.

With the increasing prevalence of electronic devices, awareness of the risks linked to blue light exposure has significantly heightened. Lutein, a powerful antioxidant, safeguards eye tissue by filtering blue light, while supplementation with docosahexaenoic acid (DHA) enhances retinal function. Adequate intake of these nutrients can help reduce the potential damage from prolonged blue light exposure. The protective effects of lutein and algal oil stabilized with Pickering emulsion were investigated using casein-dextran (CD) conjugates via Maillard reaction. Microstructural analysis revealed a
