Micromolding of photocrosslinkable chitosan hydrogel for spheroid microarray and co-cultures.

Bioengineering approaches, such as co-cultures of multiple cell types, that aim to mimic the physiological microenvironment may be beneficial for optimizing cell function and for engineering tissues in vitro. This study describes a novel method for preparing a spheroid microarray on microfabricated hydrogels, alone or in co-cultures. Photocrosslinkable chitosan was synthesized and utilized for fabricating hydrogel microstructures through a micromolding process. The chitosan surface was initially cell repellent but became increasingly cell adhesive over time. By using this unique property of ch

Self-assembly of an amphiphilic derivative of chitosan and micellar solubilization of puerarin.

A kind of amphiphilic derivatives of chitosan (2-hydroxyl-3-butoxyl)-propylcarboxymethyl-chitosan (HBP-CMCHS), has been synthesized, and the critical micelle concentration (cmc) of HBP-CMCHS was detected by the fluorescence method. The puerarin-loaded HBP-CMCHS micellar system was prepared by physical entrapped method. Result showed that when adding the same amount of puerarin, the solubilizing capacity was enhanced by increasing the concentration of HBP-CMCHS and temperature. Puerarin-loaded micellar system of HBP-CMCHS was characterized by TEM and DLS. TEM photograph revealed that the micell

Influence of molecular weight on oral absorption of water soluble chitosans.

Chitosan, a cationic polysaccharide, has been widely employed as dietary supplement and in pharmacological and biomedical applications. Although numerous studies have focused on its applications as pharmaceutical excipients or bioactive reagents, molecular weight (MW)-dependent pharmaceutical and bioactive properties remain unclear. As a preliminary study, we investigated the MW-dependent Caco-2 cell layer transport phenomena (in vitro) and intestinal absorption patterns after oral administration (in vivo) of water-soluble chitosans (WSCs). The absorption of chitosan was significantly influenc

Anti-ulcer effects of chitin and chitosan, healthy foods, in rats.

In this study, we compared the effects of low molecular weight (LMW) chitosan (MW: 25,000-50,000), high molecular weight (HMW) chitosan (MW: 500,000-1000,000) and chitin on ethanol-induced gastric mucosal injury and on the healing of acetic acid-induced gastric ulcers in rats. Oral administration of LMW chitosan (250, 500 and 1000 mg/kg) dose-dependently prevented ethanol-induced gastric mucosal injury. Repeated oral administration of LMW chitosan (100, 200 and 400 mg/kg twice daily) also dose-dependently accelerated the gastric ulcer healing. However, the effects of HMW chitosan and chitin on

Enhancing Mucosal Immune Response of Newcastle Disease Virus DNA Vaccine Using N-2-Hydroxypropyl Trimethylammonium Chloride Chitosan and N,O-Carboxymethyl Chitosan Nanoparticles as Delivery Carrier.

Because mucosal sites are the entry ports of pathogens, immunization via mucosal routes can extremely enhance the immunity. To elevate the potential of N-2-hydroxypropyl trimethylammonium chloride chitosan (N-2-HACC) and N,O-carboxymethyl chitosan (CMC) nanoparticles as a mucosal immune delivery carrier for DNA vaccines, we prepared the NDV F gene plasmid DNA with C3d6 molecular adjuvant (pVAX I-F(o)-C3d6) encapsulated in the N-2-HACC-CMC nanoparticles (N-2-HACC-CMC/pFDNA-C3d6 NPs). The N-2-HACC-CMC/pFDNA-C3d6 NPs had regular spherical morphology and low toxicity with a mean diameter of 309.7

Deep eutectic solvent-assisted extraction of chitosan for polyethylene microplastic coagulation.

Microplastic pollution has emerged as an important environmental challenge. Among existing remediation strategies, coagulation-flocculation offers a cost-effective and scalable approach. However, the reliance on conventional chemical coagulants raises concerns regarding sludge generation and secondary environmental impacts. Chitosan has therefore attracted interest as a bio-based coagulant, but the environmental advantages of its use are closely dependent on its extraction process. This study investigates the use of deep eutectic solvents (DESs) as alternative extractants for chitosan producti

Sulfated chitosan mitigates acute lung injury induced bone loss via immunoregulation.

Respiratory inflammatory diseases disrupt bone metabolism and cause pathological bone loss. The lung-bone axis is established in chronic diseases like asthma and cystic fibrosis but is less studied in acute lung injury (ALI), recently implicated in COVID-19-induced bone loss. This study examined the effects of LPS-induced ALI on bone phenotype and explored the role of 2-N, 6-O sulfated chitosan (26SCS) in mitigating pneumonia-induced bone loss via inflammatory response modulation. Our findings show that 26SCS effectively reaches bone tissue after oral administration. It promotes macrophage pol

Chitosan and polycaprolactone blended PDMS coatings improve biocompatibility of magnetic elastomers.

PDMS/NdFeB composites are promising materials for soft magnetic actuators, but NdFeB particles corrode in body fluids and release toxic metal ions, limiting their biomedical use. We developed ~ 100 µm spin-coated PDMS-chitosan (PDMS-CHIT) and PDMS-polycaprolactone (PDMS-PCL) coatings that solve this problem. Over 24 weeks of immersion, these coatings reduced neodymium and iron release by more than 95%, keeping ion concentrations well below cytotoxicity thresholds. Importantly, the PDMS-PCL coating fully preserved magnetorheological actuation (ΔG' ≈ 61 kPa under 0.5 T, comparable to uncoated co

Physically crosslinked sodium carboxymethyl cellulose/chitosan antifungal coating for litchi preservation.

Polysaccharide-based antimicrobial coatings have garnered significant attention for preservation in fruits and vegetables, but the flavor impact of essential oils as antimicrobial agents remains an issue. Herein, natamycin (NAT), which is odorless and has broad-spectrum antifungal activity, was incorporated into a physically crosslinked network of carboxymethyl cellulose sodium salt (CMCNa) and chitosan quaternary ammonium salt (HACC) to prepare antimicrobial coatings for litchi preservation. The optimal CMC-Na/HACC film formulation (CH5) exhibits outstanding barrier properties. Compared to th

Anti-Inflammatory Effects of Artocarpin-Loaded Chitosan Microparticles on Inflammatory Chondrocytes.

Artocarpin is a promising bioactive compound with anti-inflammatory potential; however, its limited aqueous solubility may restrict its direct biological application. Therefore, artocarpin-loaded chitosan microparticles (CSPs/AE) were prepared as a biocompatible carrier to improve artocarpin delivery. This study aimed to evaluate the cytocompatibility, antioxidant activity, and anti-inflammatory effects of CSPs/AE in lipopolysaccharide (LPS)-stimulated chondrocytes. CSPs/AE maintained high cell viability (>80% at 0.125 and 0.5 mg/mL) and showed significantly lower cytotoxicity than free artoca

Harnessing 5-fluorouracil-loaded chitosan nanoparticles for targeted colon cancer therapy.

Colon cancer remains a significant global health burden, and although 5-fluorouracil (5-FU) is a cornerstone chemotherapeutic agent, its clinical utility is constrained by systemic toxicity and poor tumor selectivity. This review highlights recent advances in chitosan-based nanoparticles as targeted delivery systems for 5-FU in colon cancer therapy. Chitosan, a biocompatible and biodegradable polymer, offers advantages such as mucoadhesion and pH sensitivity, making it well-suited for colon-specific drug delivery. The review covers formulation strategies, physicochemical characterization, and

Enhanced enzymatic hydrolysis of chitosan via MOF-mediated activation of chitosanase.

Chitosanases are attractive biocatalysts for producing bioactive chito-oligosaccharides (COS), but their applications are limited by insufficient catalytic efficiency, poor stability, and challenging recovery. Although many chitosanases can be activated by metal ions, the direct addition of free ions into reaction systems suffers from low efficiency and separation challenges. This study presented a MOF-based strategy to create a Cu-enriched coordination microenvironment, enabling simultaneous immobilization and activation of marine polysaccharide hydrolases. Taking chitosanase OUC-CsnCA as a m
