Response to hypercapnia and exercise hyperpnea in graded anesthesia.

We examined the relationship between response to hypercapnia and ventilatory response to exercise under graded anesthesia in eight dogs. The response to hypercapnia was measured by the CO2 rebreathing method under three grades of chloralose-urethan anesthesia. The degrees of response to hypercapnia (delta VE/delta PETCO2, 1 X min-1 X Torr-1) in light (L), moderate (M), and deep (D) anesthesia were 0.40 +/- 0.05 (mean +/- SE), 0.24 +/- 0.03, and 0.10 +/- 0.02, respectively, and were significantly different from each other. Under each grade of anesthesia, exercise was performed by electrically s…

Cardiovascular responses to opioid agonists injected into the nucleus of tractus solitarius of anesthetized cats.

It has been proposed that various opiate receptor subtypes mediate different cardiovascular responses to centrally administered opioids. We evaluated this hypothesis in chloralose-urethane anesthetized cats by monitoring the cardiovascular and respiratory responses to relative mu [morphine, morphiceptin, D-Ala2, MePhe4, Gly-ol5 enkephalin (DAGO)] and delta [D-Ala2, D-Leu5enkephalin (DADL)] agonists microinjected (0.5 ul/kg) into the caudal region of the Nucleus of Tractus Solitarius (NTS). Dynorphin (1-13), an endogenous opioid which exhibits selective affinity towards the kappa receptor, was …

Brain tissue pressure measurement during sodium nitroprusside infusion.

The effect of acute 25% decrease in mean arterial pressure (MAP) produced by sodium nitroprusside (SNP) infusion on mean intracerebral and cerebral perfusion pressures was examined in 7 swine with intracranial hypertension. All animals were anesthetized (alpha-chloralose), paralyzed (pancuronium bromide), orotracheally intubated and mechanically ventilated to maintain normocapnia. An epidural balloon was incrementally inflated to produce the desired brain tissue pressure (BTP) which was measured by an intracerebral microtransducer implanted contralateral to the balloon. MAP was measured from t…

A comparison of the cardiovascular effects of sodium nitroprusside and trimethaphan.

In dogs anesthetized with pentobarbital-chloralose, cardiac output and blood flows of four regional vascular beds (superior mesenteric, left renal, left circumflex coronary and left femoral) were continuously monitered with electromagnetic flowmeters. Arterial blood pressure and heart rate were also measured. Hypotension was induced with intravenous infusions of sodium nitroprusside and trimethaphan for 5-16 min to produce comparable reductions of mean arterial pressure (32 mm Hg or 26 per cent with nitroprusside and 37 mm Hg or 31 per cent with trimethaphan). Cardiac output also decreased, bu…

Assessment of muscarinic transmission in the superior cervical and ciliary ganglion of the cat.

This study was undertaken to determine if muscarinic mechanisms are involved in synaptic transmission in the parasympathetic ciliary ganglion as has been clearly shown for sympathetic ganglia. Cats were anesthetized, and following topical ephedrine, pupillary constrictions were elicited by electrical stimulation of the intracranial oculomotor nucleus. Nictitating membrane contractions were evoked by electrical stimulation of the preganglionic cervical nerve. Frequency-response curves were repeated after infusion with hexamethonium (0.6-1.0 mg/kg min-1) and after subsequent administration of at

Renin release, an artifact of anesthesia and its implications in rats.

In our attempt to find an anesthetic agent which did not influence the renin-angiotensin system in the rat, the effect of widely used injectable and gaseous anesthetics and narcotic agents on renin release was chacterized. All of the agents studied induced dose- and time-related increases in serum renin activity when administered in anesthetic doses. Preliminary experiments in anesthetic doses. Preliminary experiments indicated that cardiovascular effects were highly variable, giving little insight into the relationship between renin release and cardiovascular changes. Propranolol impaired mos

Impact of Mechanical Ventilation and Anesthesia on PET Tracer Kinetics for Combined PET/fMRI Studies in Rats.

PURPOSE: Positron Emission Tomography (PET), a crucial tool in molecular brain imaging, has evolved into a hybrid system through integration … e under mechanical ventilation. The anesthesia type also strongly influenced the tracer kinetics: α-chloralose anesthesia reduced brain uptake, whereas isoflurane led to a more rapid equilibrium. CONCLUSIONS: These findings underscore the profound impact of mechanical ventilation and anesthesia selection on PET tracer kinetics in hybrid PET/fMRI studies. Th …

Intraurethral stimulation evokes bladder responses via 2 distinct reflex pathways.

PURPOSE: Recent animal studies have shown that selective activation of pudendal nerve branches can evoke bladder responses through 2 distinct reflex pathways. We examined intraurethral electrical stimulation as a minimally invasive means of selectively activating these pathways in the cat. MATERIALS AND METHODS: Bladder responses evoked by intraurethral electrical stimulation were measured in alpha-chloralose anesthetized male cats at different stimulation frequencies, stimulation intensities and intraurethral locations. RESULTS: Intraurethral electrical stimulation evoked inhibitory and exc…

Acetazolamide induces indomethacin and ischaemia-sensitive pial arteriolar vasodilation in the piglet.

AIM: Acetazolamide (AZD) produces cerebral vasodilation. The underlying mechanism is unclear, but it is assumed to be largely due to CO2 retention and acidosis. We tested if cerebrovascular effects of AZD were similar to hypercapnia in the newborn pig. METHODS: We used the closed cranial window/intravital microscopy technique to determine pial arteriolar diameters simultaneously with laser-Doppler flowmetry (LDF) to monitor cortical blood perfusion. Anaesthetized (Na-thiopenthal +alpha-chloralose), ventilated, 1-day-old instrumented piglets (n=38) were divided into five experimental groups: t…

The effect of dexmedetomidine on nutrient organ blood flow.

The alpha 2-adrenergic agonist dexmedetomidine decreases not only heart rate, myocardial contractility, and oxygen demand, but also cardiac output (Q). To investigate whether this reduction in Q could critically impair perfusion of individual organs, we studied the effect of dexmedetomidine on nutrient blood flow to the heart, brain, kidney, spleen, skin, intestine, liver, and arteriovenous anastomoses using the radioactive microsphere technique. Studies were conducted in 14 dogs with an open chest and anesthetized with either chloralose/urethane (CU) or fentanyl/halothane (FH), to create diff…

Respiratory and acid-base parameters during salicylic intoxication in dogs.

This paper examines the mechanism responsible for hyperventilation and accompanying respiratory alkalosis during acute salicylism. Sodium salicylate (250 mg/kg) was administered to 8 spontaneously breathing anesthetized dogs (alpha-chloralose, 50 mg/kg, and urethane, 500 mg/kg, iv). The trachea was sectioned and connected to a pneumotachograph. A catheter was placed in the cisterna magna for sampling cerebrospinal fluid (CSF) and a femoral artery was cannulated for blood sampling and pressure determinations. Once the cardiorespiratory steady-state was obtained, air flow, tidal volume, arterial…

Ex vivo NMR study of lactate metabolism in rat brain under various depressed states.

Brain endogenous lactate metabolism was investigated by ex vivo nuclear magnetic resonance (NMR) spectroscopy study after the infusion of rats with a solution of glucose and lactate labeled as either [3-(13)C]lactate or [1-(13)C]glucose, when their cerebral activity was more or less depressed under the influence of either pentobarbital, alphachloralose, or morphine. We found that: (1) the ratio between the enrichment of alanine C3 and that of glutamate C4, gamma-aminobutyric acid (GABA) C2, glutamine C4, or aspartate C3 decreased from pentobarbital to alphachloralose and morphine whatever the …

Oxytocin increased intragastric pressure in the forestomach of rats via the dorsal vagal complex.

We previously reported that appetite-enhancing peptides facilitated phasic contractions of the distal stomach and relaxed the forestomach via the dorsal vagal complex (DVC). The present study investigated the effects of anorectic substances on gastric reservoir function. The effects of oxytocin on the motility of the forestomach were examined in rats anesthetized with urethane-chloralose. Gastric motor responses were measured using an intragastric balloon. The fourth ventricular administration of oxytocin (0.1 - 1.0 nmol) increased intragastric pressure (IGP) in the forestomach in a dose-depen…
