Primary localized cutaneous amyloidosis (PLCA) is caused by the deposition of keratin amyloids in the skin and leads to severe symptoms. It is difficult to cure PLCA completely; therefore, prevention is essential. In this study, amyloid formation of a keratin-derived amyloidogenic peptide, KT5-6, was successfully inhibited by rosmarinic acid (RA), a polyphenol present in plants such as rosemary. A catalytic enantioselective conjugate addition of cyanide to alpha,beta-unsaturated N-acylpyrroles was developed using the chiral gadolinium catalyst generated from Gd(OiPr)3 and d-glucose-derived ligand 2. Generally high enantioselectivity was obtained from a wide range of substrates; substrates with beta-aryl and beta-vinyl substituents and alpha,beta-disubstituted substrates can now be used. Using this reaction as a key step, short-step syntheses of several pharmaceuticals and their lead compounds were achieved, including the beta-phenyl-substituted GABA analogue and pregabalin.

Lusutrombopag, a small molecule thrombopoietin receptor agonist, has been approved for the treatment of chronic liver disease-associated thrombocytopenia due to hypersplenism in patients scheduled to undergo elective invasive procedures in Japan. We performed partial splenic embolization (PSE) after administration of lusutrombopag in two patients with thrombocytopenia due to cirrhosis. In this study, a new degradation path of sunscreen active ingredient, 2-ethylhexyl-4-methoxycinnamate (EHMC) and 4-methoxycinnamic acid (MCA) in the presence of sodium hypochlorite (NaOCl), was discussed.
