Open science datasets from PREVENT-AD, a longitudinal cohort of pre-symptomatic Alzheimer's disease.

To move Alzheimer Disease (AD) research forward it is essential to collect data from large cohorts, but also make such data available to the global research community. We describe the creation of an open science dataset from the PREVENT-AD (PResymptomatic EValuation of Experimental or Novel Treatments for AD) cohort, composed of cognitively unimpaired older individuals with a parental or multiple-sibling history of AD. From 2011 to 2017, 386 participants were enrolled (mean age 63 years old ± 5) for sustained investigation among whom 349 have retrospectively agreed to share their data openly.

Modeling Neurodegeneration in silico With Deep Learning.

Deep neural networks, inspired by information processing in the brain, can achieve human-like performance for various tasks. However, research efforts to use these networks as models of the brain have primarily focused on modeling healthy brain function so far. In this work, we propose a paradigm for modeling neural diseases in silico with deep learning and demonstrate its use in modeling posterior cortical atrophy (PCA), an atypical form of Alzheimer's disease affecting the visual cortex. We simulated PCA in deep convolutional neural networks (DCNNs) trained for visual object recognition by r

Re-evaluating the role of TPJ in attentional control: contextual updating?

The right temporo-parietal junction (TPJ) is widely considered as part of a network that reorients attention to task-relevant, but currently unattended stimuli (Corbetta and Shulman, 2002). Despite the prevalence of this theory in cognitive neuroscience, there is little direct evidence for the principal hypothesis that TPJ sends an early reorientation signal that "circuit breaks" attentional processing in regions of the dorsal attentional network (e.g., the frontal eye fields) or is completely right lateralized during attentional processing. In this review, we examine both functional neuroimag…

Distinguishing how from why the mind wanders: a process-occurrence framework for self-generated mental activity.

Cognition can unfold with little regard to the events taking place in the environment, and such self-generated mental activity poses a specific set of challenges for its scientific analysis in both cognitive science and neuroscience. One problem is that the spontaneous onset of self-generated mental activity makes it hard to distinguish the events that control the occurrence of the experience from those processes that ensure the continuity of an internal train of thought once initiated. This review demonstrates that a distinction between process and occurrence (a) provides theoretical clarity

Artificial intelligence and machine learning for precision prevention of cognitive decline: integrating multimodal biomarkers, lifestyle interventions, and natural medicines from prediction to clinical practice.

Cognitive decline and neurodegenerative diseases are progressive, multifactorial conditions that may begin years before overt clinical diagnosis and reflect interactions among biological vulnerability, modifiable exposures, environmental determinants, and reduced brain resilience. This structured narrative review examines how artificial intelligence (AI) and machine learning (ML) can integrate clinical, biological, behavioral, and digital data to support earlier, personalized, and clinically actionable strategies for preserving cognitive health. Drawing on evidence from aging neuroscience, bio

Hormetic pathways in environmental enrichment in animal models and humans.

Environmental Enrichment (EE), which provides enhanced sensory, cognitive, motor, and social stimulation, has emerged as a powerful paradigm for investigating neuroplasticity and stress resilience. This chapter explores how EE functions through hormetic mechanisms-where multiple mild stimuli trigger adaptive responses that promote beneficial outcomes. Since Hebb's pioneering work on neuronal ensembles, research has demonstrated that EE enhances neurogenesis, synaptic plasticity, and neurotrophic factor expression (BDNF and NGF) while modulating inflammatory processes, epigenetic pathways, and

[Dopaminergic modulation of cerebral activity and cognitive functions].

Alterations in dopaminergic system are known to lie in the basis of such diseases as Parkinson's disease, Huntington's disease, Attention Deficit/Hyperactivity Disorder, Tourette syndrome, schizophrenia and drug abuse. This induced broad investigations of dopaminergic system in nearly all the areas of neuroscience. New insights into the pathogenesis of neuropsychiatric diseases have emerged. Research in the field of dopaminergic neurotransmission and memory was awarded Nobel prize in the year 2000. New avenues for the development of more selective drugs have been opened. Decoding neuronal criticality firing patterns for large brain based EEG models.

The critical brain hypothesis suggests that the brain operates near critical points, balancing order and disorder for optimal processing via self-organized neural firing patterns. However, important gaps remain in understanding criticality in human in vivo brain dynamics using electroencephalography (EEG), particularly in translating these systems neuroscience concepts to operational applications (e.g., cognitive state detection, human-machine teaming). We address three key aspects that yet remain to be demonstrated: (1) how to decode large-scale EEG neural recordings coordination and self-org

Exploring Psilocybin-Assisted Schema Therapy: A Conceptual Framework for Potential Therapeutic Synergies in Personality Disorders.

Personality disorders (PDs) are characterized by rigid and maladaptive patterns of self- and interpersonal functioning, leading to high clinical burden and limited treatment outcomes. Schema Therapy (ST), an integrative psychotherapy rooted in cognitive-behavioral principles, conceptualizes PDs in terms of Early Maladaptive Schemas (EMS)-pervasive cognitive-affective structures formed through unmet emotional needs-and schema modes, dynamic states organizing emotion, belief, and behavior. Evidence indicates moderate efficacy of ST, mainly for borderline personality disorder, with limited resear

Cognition and impulsivity in adults with attention deficit hyperactivity disorder with and without cocaine and/or crack dependence.

BACKGROUND: Substance use disorder (SUD) is a common comorbidity in adults with attention deficit-hyperactivity disorder (ADHD). However, there have been few studies on cognitive profiles of these patients. Impulsivity is also commonly increased in both disorders. The central aim of this study was to compare cognition and impulsivity in subjects who had ADHD and cocaine dependence (ADHD+COC group) to those with ADHD only (ADHD-noSUD group). We hypothesized that the ADHD+COC group would show more marked cognitive dysfunction and greater impulsivity than their counterparts with ADHD only. METHOD

White matter dementia then… and now.

White matter dementia (WMD) is a concept introduced in 1988 to highlight the importance of white matter pathology in producing cognitive dysfunction and dementia. Whereas gray matter, particularly the cerebral cortex, has been primarily investigated in the dementias, subcortical pathology has long been correlated with cognitive loss, and a corticocentric perspective cannot account for the full range of neurobehavioral disorders. Within the subcortical regions, white matter is prominent, accounting for about half the volume of the adult brain, and many white matter diseases, injuries, and intox

Learning to 'talk the talk: the relationship of psychopathic traits to deficits in empathy across childhood.

BACKGROUND: Psychopathy is characterised by profound deficits in the human tendency to feel and care about what other people feel, often known as 'affective empathy'. On the other hand, the psychopath often has intact 'cognitive' empathy skills, that is, he is able to describe what and why other people feel, even if he does not share or care about those feelings. Despite a rapidly advancing neuroscience of empathy, little is known about the developmental underpinnings of this psychopathic disconnect between affective and cognitive empathy. METHODS: The parents of N = 2760, 3-13-year-olds repo

Age-related hearing loss and dementia risk across the lifespan: mechanisms, equity, and prevention.

Age-related hearing loss (ARHL) has emerged as a significant and potentially modifiable risk factor for neurodegenerative disorders, including Alzheimer's disease. A growing body of evidence links ARHL to structural and functional changes in the brain, with implications for cognitive decline and dementia onset. However, both ARHL and dementia are multifactorial conditions shaped not only by biological mechanisms but also by broader social determinants of health. Inequities in access to hearing care, underrepresentation of marginalised populations in research, and variability in intervention ou

Minimally Conscious State: Evolution of Concept, Diagnosis and Treatment.

The study of consciousness disorders is a scientific challenge, and clinical differentiation among the various sorts of alterations in consciousness is difficult. Persistent vegetative state was defined in 1972, but years later cases appeared in which diagnosed patients showed signs of cognitive activity, and therefore could not be considered vegetative. Minimally conscious state was defined in 2002. This article discusses minimally conscious state based on a literature review and the author's clinical experience. A brief historical outline is given, starting from 1886 when Horsley analyzed le

Functional connectivity analysis of brain hemodynamics during rubber hand illusion.

Embodied cognition has been eagerly studied in the recent neuroscience research field. In particular, hand ownership has been investigated through the rubber hand illusion (RHI). Most of the research measured the brain activities during the RHI by using EEG, fMRI, etc., however, near-infrared spectroscopy (NIRS) has not yet been utilized. Here we attempt to measure the brain activities during the RHI task with NIRS, and analyze the functional connectivity so as to understand the relationship between NIRS features and the state of embodied cognition. For the purpose, we developed a visuo-tactil

Anticipation of cognitive demand during decision-making.

An abundance of evidence indicates that action selection is guided, at least in certain contexts, by anticipation of action outcomes. In one particularly clear demonstration of this principle, Bechara and colleagues, studying a gambling task, observed phasic skin conductance responses just prior to actions associated with a relatively high risk of monetary loss (Bechara et al. in J Neurosci 19:5473-5481, 1999; Bechara et al. in Science 275:1293-1295, 1997; Bechara et al. in Cereb Cortex 6:215-225, 1996). In the present work, we tested for the same effect in a paradigm where choices resulted no

Proprioception and Cognitive Polarization in Autism: The Bipolar Tensile Model.

This article introduces the Bipolar Tensile Model, a neurosemiotic and integrative framework for understanding Autism Spectrum Disorder (ASD). The model reinterprets ASD not as a collection of deficits, but as the outcome of an unresolved tension between two complementary hemispheric cognitive modalities: symbolic-sequential and corporeal-perceptual. In typical development, these modalities are dynamically harmonized through an extended interhemispheric network, comprising the Default Mode Network, major cerebral commissures (corpus callosum, anterior and hippocampal), and the cerebellum.
