Signalling requirements for Erwinia amylovora-induced disease resistance, callose deposition and cell growth in the non-host Arabidopsis thaliana.

Erwinia amylovora is the causal agent of the fire blight disease in some plants of the Rosaceae family. The non-host plant Arabidopsis serves as a powerful system for the dissection of mechanisms of resistance to E. amylovora. Although not yet known to mount gene-for-gene resistance to E. amylovora, we found that Arabidopsis activated strong defence signalling mediated by salicylic acid (SA), with kinetics and amplitude similar to that induced by the recognition of the bacterial effector avrRpm1 by the resistance protein RPM1. Genetic analysis further revealed that SA signalling, but not signa

A L-type lectin gene is involved in the response to hormonal treatment and water deficit in Volkamer lemon.

Combination of biotic and abiotic stress is a major challenge for crop and fruit production. Thus, identification of genes involved in cross-response to abiotic and biotic stress is of great importance for breeding superior genotypes. Lectins are glycan-binding proteins with a functions in the developmental processes as well as in the response to biotic and abiotic stress. In this work, a lectin like gene, namely ClLectin1, was characterized in Volkamer lemon and its expression was studied in plants exposed to either water stress, hormonal elicitors (JA, SA, ABA) or wounding to understand whet

Imaging of jasmonic acid binding sites in tissue.

Hormones regulate the mechanism of plant growth and development, senescence, and plants' adaptation to the environment; studies of the molecular mechanisms of plant hormone action are necessary for the understanding of these complex phenomena. However, there is no measurable signal for the hormone signal transduction process. We synthesized and applied a quantum dot-based fluorescent probe for the labeling of jasmonic acid (JA) binding sites in plants. This labeling probe was obtained by coupling mercaptoethylamine-modified CdTe quantum dots with JA using N-hydroxysuccinimide (NHS) as a coupli

Methyl salicylate production and jasmonate signaling are not essential for systemic acquired resistance in Arabidopsis.

Systemic acquired resistance (SAR) develops in response to local microbial leaf inoculation and renders the whole plant more resistant to subsequent pathogen infection. Accumulation of salicylic acid (SA) in noninfected plant parts is required for SAR, and methyl salicylate (MeSA) and jasmonate (JA) are proposed to have critical roles during SAR long-distance signaling from inoculated to distant leaves. Here, we address the significance of MeSA and JA during SAR development in Arabidopsis thaliana. MeSA production increases in leaves inoculated with the SAR-inducing bacterial pathogen Pseudomo

Biochemistry and molecular biology of Arabidopsis-aphid interactions.

To ensure their survival in natural habitats, plants must recognize and respond to a wide variety of insect herbivores. Aphids and other Hemiptera pose a particular challenge, because they cause relatively little direct tissue damage when inserting their slender stylets intercellularly to feed from the phloem sieve elements. Plant responses to this unusual feeding strategy almost certainly include recognition of aphid salivary components and the induction of phloem-specific defenses. Due to the excellent genetic and genomic resources that are available for Arabidopsis thaliana (Arabidopsis), t

Induction by antipsychotics of "win-shift" in the drug discrimination paradigm.

In a two-lever, food-rewarded drug discrimination paradigm, behavior seems to be governed by a win-stay/lose-shift rule; rats continue to press the lever that yields food, and, when not rewarded, they shift responding to the alternative lever. Here, we report on the effects that antipsychotics and further neuropharmacological agents exert in those conditions. At higher doses, antipsychotics disrupt most or all behavioral parameters in this paradigm. However, at lower doses, rats may select the appropriate lever with normal latency and accuracy, obtain a first food pellet (i.e., "win"), and the

Yeast increases resistance in Arabidopsis against Pseudomonas syringae and Botrytis cinerea by salicylic acid-dependent as well as -independent mechanisms.

Cell-wall and glucopeptide components of yeast have been reported to exhibit elicitor activity. The mode of action of defense activation by yeast is not known so far. In this study, we used the model plant Arabidopsis to investigate the activation of defense responses by yeast, the effect on resistance against different pathogens, and the mode of action. Treatment of Arabidopsis plants with an autoclaved yeast suspension induced the expression of systemic acquired resistance-related genes and accumulation of the phytoalexin camalexin. Symptom development and bacterial growth after infection wi

Electrical and chemical signals involved in short-term systemic photosynthetic responses of tobacco plants to local burning.

Short-term (up to 1 h) systemic responses of tobacco (Nicotiana tabacum cv. Samsun) plants to local burning of an upper leaf were studied by measuring the following variables in a distant leaf: extracellular electrical potentials (EEPs); gas exchange parameters; fast chlorophyll fluorescence induction; and endogenous concentrations of three putative chemical signaling compounds-abscisic (ABA), jasmonic (JA), and salicylic (SA) acids. The first detected response to local burning in the distant leaves was in EEP, which started to decline within 10-20 s of the beginning of the treatment, fell sha

Impaired fungicide activity in plants blocked in disease resistance signal transduction.

Fungicide action is generally assumed to be dependent on an antibiotic effect on a target pathogen, although a role for plant defense mechanisms as mediators of fungicide action has not been excluded. Here, we demonstrate that in Arabidopsis, the innate plant defense mechanism contributes to the effectiveness of fungicides. In NahG and nim1 (for noninducible immunity) Arabidopsis plants, which normally exhibit increased susceptibility to pathogens, the fungicides metalaxyl, fosetyl, and Cu(OH)2 are much less active and fail to control Peronospora parasitica. In contrast, the effectiveness of t

Neuroprotective effects of N-acetylaspartylglutamate in a neonatal rat model of hypoxia-ischemia.

Neuroprotective effects of N-acetylaspartylglutamate (NAAG), the precursor of glutamate and a selective agonist at the Group II metabotropic glutamate (mGlu) receptor, against hypoxic-ischemic brain injury were examined in a neonatal rat model of cerebral hypoxia-ischemia. The neonatal hypoxia-ischemia procedure (unilateral carotid artery ligation followed by exposure to an 8% oxygen hypoxic condition for 1.5 h) was performed in 7-day-old rat pups. Following unilateral carotid artery ligation, NAAG (0.5 to 20 mg/kg, i.p.) was administered before or after the hypoxic exposure. Brain injury was

Integrated transcriptomic and metabolomic analysis reveals differential molecular mechanisms underlying resistance of native and invasive Phytolacca americana against Spodoptera litura herbivory.

BACKGROUND: In response to herbivory from insects, plants have evolved complex defense strategies. Alien plants often escape control by specialist insects in their introduced range. This release from herbivory pressure may lead to a reallocation of the defensive strategies of alien plants, thereby promoting their invasiveness. However, the molecular mechanisms underlying this resource reallocation process remain unclear. In the present study, we used Phytolacca americana as a model species, and combined transcriptomic and metabolomic analyses to investigate the molecular basis of its invasion

Autoinhibition of plasma membrane H+-ATPase1 regulates systemic herbivore defense in Arabidopsis.

Autoinhibited plasma membrane H+-ATPases1 (AHA1) plays a crucial role in wound-induced systemic signaling, yet its role in insect resistance has not been fully explored. Prior research on fungal and plant AHAs has identified an extended carboxy-terminal tail (C-tail), which forms a regulatory apparatus involving an autoinhibitory domain. Here, we show that truncation of the C-tail of AHA1 increases pump activity and reduces the duration of repolarization after wounding. This also led to reduced expression of the jasmonate pathway marker gene JAZ10 in both wounded and systemic leaves. By specif

Salicylic acid suppresses 1O2-mediated susceptibility to Alternaria alternata in Arabidopsis.

Necrotrophic Alternaria alternata induces EXECUTER 1(EX1)/2-dependent singlet oxygen (1O2) bursts, leading to plant cell death, with jasmonic acid (JA) acting as a key signal transducer downstream of EX1/2-mediated signaling. Salicylic acid (SA), a crucial defense hormone, is known to respond to pathogen invasion and activate defense gene expression. Previous studies emphasized the importance of SA in A. alternata-induced necrosis in the light of the increased susceptibility of SA-deficient transgenic Arabidopsis NahG to A. alternata. In this study, we investigated the role of SA in A. alterna

CaMACPF1 and CaMACPF6 positively regulate hypoxia tolerance through modulating jasmonate and auxin metabolic pathways.

Submergence and flooding impose severe crop losses through hypoxic stress. The Membrane Attack Complex/Perforin (MACPF) superfamily proteins function as pore-forming effectors in development and immunity across eukaryotic organisms, yet their role in plant hypoxia responses remains largely unexplored. Here, we identify two MACPF proteins in pepper (Capsicum annuum), CaMACPF1 and CaMACPF6, as positive regulators of hypoxia tolerance. CaMACPF1 and CaMACPF6 localize to endoplasmic reticulum-plasma membrane contact sites (EPCS) and the Golgi apparatus, with CaMACPF1 additionally detected in the nu

Deciphering the tomato defense network againstHelicoverpa armigera: From transcriptional reprogramming to biochemical changes.

Helicoverpa armigera (Hübner) is able to feed on more than 280 host plants; however, the mechanisms that allow it to persist on such a wide range of hosts may involve transcriptional reprogramming and biochemical alternations which are still not well understood. Therefore, we analyzed the transcriptional responses of salicylic acid (SA)- and jasmonic acid (JA)-mediated signaling pathways in tomato (Solanum lycopersicum L.), which is increasingly used as a model plant system. The SA-related genes PR1a and PAL showed increased expression both locally and systemically at 12, 24, 36, and 48  h pos

Validamycin A Induced Resistance in Rice against Rhizoctonia solani via the Salicylic Acid-Depended Signaling Pathway.

Validamycin A (VMA), the major active component of validamycin, provides effective control of rice sheath blight; however, the mechanisms underlying VMA-induced resistance to Rhizoctonia solani remain poorly understood. Here, we integrated antioxidant enzyme assays, phytohormone profiling, transcriptomics, and metabolomics to investigate VMA-induced immunity in rice. VMA improved oxidative stress tolerance by enhancing antioxidant enzyme activities and activating glutathione metabolism. Moreover, VMA established a primed multihormonal state characterized by elevated salicylic acid (SA), jasmon

Rhizoctonia solani Secretes RsCAP3 to Target Nb14-3-3b, Interfering with Hormone-Mediated Resistance in Nicotiana benthamiana.

The necrotrophic phytopathogen Rhizoctonia solani causes leaf spot and root rot in tobacco, which significantly reduce the yield and quality of the crop. Recently, much attention has been paid to R. solani effectors, but only a few of them have been characterized. Here, we identified a CAP domain-containing protein, RsCAP3, with a functional signal peptide, which triggers slight cell death on tobacco leaves. Meanwhile, RsCAP3 dramatically induces salicylic acid (SA) signaling but inhibits jasmonic acid (JA) signaling, increasing the plant's susceptibility to R. solani. Moreover, we demonstrate

H2O2-Dependent Methyl Jasmonate Regulates H2S-Induced Resistance to Fusarium oxysporum f. sp. niveum Race 2 in Citrullus lanatus.

Fusarium wilt, caused by Fusarium oxysporum (Fo), is a destructive fungal disease that reduces crop yield and quality. Hydrogen sulphide (H2S), a critical signalling molecule, modulates plant defence responses; however, its role and mechanism in combating Fo remain elusive. This study reveals that exogenous NaHS (an H2S donor) enhances watermelon resistance to Fusarium oxysporum f. sp. niveum race 2 (FON2), accompanied by elevated hydrogen peroxide (H2O2) and methyl jasmonate (MeJA) levels. Exogenous H2O2 and MeJA also enhance FON2 resistance. Conversely, silencing respiratory burst oxidase ho

Leaf abaxial and adaxial surfaces differentially affect the interaction of Botrytis cinerea across several eudicots.

Eudicot plant species have leaves with two surfaces: the lower abaxial and the upper adaxial surface. Each surface varies in a diversity of components and molecular signals, resulting in potentially different degrees of resistance to pathogens. We tested how Botrytis cinerea, a necrotroph fungal pathogen, interacts with the two different leaf surfaces across 16 crop species and 20 Arabidopsis genotypes. This showed that the abaxial surface is generally more susceptible to the pathogen than the adaxial surface. In Arabidopsis, the differential lesion area between leaf surfaces was associated wi
