Environmental chemicals and breast cancer: An updated review of epidemiological literature informed by biological mechanisms.

BACKGROUND: Many common environmental chemicals are mammary gland carcinogens in animal studies, activate relevant hormonal pathways, or enhance mammary gland susceptibility to carcinogenesis. Breast cancer's long latency and multifactorial etiology make evaluation of these chemicals in humans challenging. OBJECTIVE: For chemicals previously identified as mammary gland toxicants, we evaluated epidemiologic studies published since our 2007 review. We assessed whether study designs captured relevant exposures and disease features suggested by toxicological and biological evidence of genotoxicit

[Mechanism study of Zhizi Dahuang Decoction in treating atherosclerosis based on network pharmacology, molecular docking, and experimental validation].

This paper investigated the effects and mechanisms of Zhizi Dahuang Decoction(ZZDHT) in treating atherosclerosis(AS) through a comprehensive approach combining network pharmacology, molecular docking, and in vivo experiments. Active components and target sites of ZZDHT were identified and screened using TCMSP, SwissTargetPrediction, and SEA databases. AS-related target sites were obtained from GeneCards, OMIM, and TTD databases. Network pharmacology was employed to predict the core chemical constituents, core target sites, and potential mechanisms of ZZDHT in treating AS. Molecular docking was

A Tier-I leaching risk assessment of three anticoagulant compounds in the forested areas of Hawai'i.

The anticoagulant rodenticides brodifacoum, chlorophacinone, and diphacinone have been proposed for broadcast application in some forested areas in Hawai'i to protect rare and endangered native bird species from introduced mice and rats. Groundwater resources in Hawai'i are prone to contamination due to the intrinsic aquifer vulnerability to leaching from the land surface. Because of the hydrogeologic complexity, Hawai'i uses a Tier-I leaching assessment tool, CLERS, to make registration decisions for new or existing chemicals. The CLERS tool uses soil and pesticide properties as well as water

The safety and efficacy of Diphoterine for ocular and cutaneous burns in humans.

CONTEXT: Diphoterine, developed by the French company Prevor, is a polyvalent, chelating, amphoteric and slightly hypertonic solution used in the management of chemical cutaneous and ocular burns. While used extensively in Europe and Canada, it is has not been approved by the United States Occupational Safety and Health Administration (OSHA) as an alternative to the water-rinse method due to a lack of evidence of its safety and efficacy on human subjects. An unbiased and extensive systematic review was undertaken in order to better understand Diphoterine's safety and efficaciousness on humans.

ISiCLE: A Quantum Chemistry Pipeline for Establishing in Silico Collision Cross Section Libraries.

High-throughput, comprehensive, and confident identifications of metabolites and other chemicals in biological and environmental samples will revolutionize our understanding of the role these chemically diverse molecules play in biological systems. Despite recent technological advances, metabolomics studies still result in the detection of a disproportionate number of features that cannot be confidently assigned to a chemical structure. This inadequacy is driven by the single most significant limitation in metabolomics, the reliance on reference libraries constructed by analysis of authentic r

Pan-selectin inhibitors as potential therapeutics for COVID-19 treatment: in silico screening study.

Coronavirus disease 2019 (COVID-19) has spread rapidly throughout the globe. The spectrum of disease is broad but among hospitalized patients with COVID-19, respiratory failure from acute respiratory distress syndrome is the leading cause of mortality. There is an urgent need for an effective treatment. The current focus has been developing novel therapeutics, including antivirals, protease inhibitors, vaccines and targeting the overactive cytokine response with anti-cytokine therapy. The overproduction of early response proinflammatory cytokines results in what has been described as a "cytoki

Quantitative structure-activity relationship modeling of dermatomyositis activity of drug chemicals.

Dermatomyositis (DM) is an idiopathic inflammatory disorder consisting of skin and skeletal muscle involvement. Some drugs induce DM or dermatomyositis-like syndrome (DM-LS), the others provoke polymoysitis (PM) or cause elevation of serum levels of muscle enzymes (SE) or give muscle damage (M). The unexpected adverse reactions to drugs causing myositis are not a solved contemporary problem. The aim of this study was to determine the structural requirements of eliciting drug-induced DM as compared with drug induced PM. The Common Reactivity Pattern (COREPA) approach was used to describe the st

Development of quantitative structure-activity relationship and classification models for a set of carbonic anhydrase inhibitors.

Mathematical models are developed to find quantitative structure-activity relationships that correlate chemical structure and inhibition toward three carbonic anhydrase (CA) isozymes: CA I, II, and IV. Numerical descriptors are generated to encode important topological, geometric, and electronic features of molecular structure. After descriptor generation, multiple linear regression, and computational neural network (CNN) analyses are performed on various descriptor subsets to find superior models for prediction. Committees of five CNNs were utilized to average final predicted values for the 1

Embossed Colorimetric Sensor Array-Based Optoelectronic Nose for the Detection of Real Chemical Warfare Agents and Toxic Industrial Chemicals: A Practical Approach Using Off-the-Shelf-Reagents, Thermodynamic, and Kinetic Fingerprints.

In chemical emergencies, real-time monitoring of chemical warfare agents (CWAs) and toxic industrial chemicals (TICs) is essential for effective mitigation. Ideally, such detection requires the deployment of a large number of sensors capable of providing real-time information. Despite the advantages of colorimetric systems, such as ease of fabrication, low cost, high sensitivity, and low power requirement, no single platform currently exists that can detect a broad spectrum of CWAs and TICs. To address this gap, we propose an optoelectronic nose for the detection of real CWAs, including nerve

Integrating non-targeted metabolomics and toxicology networks to study the mechanism of Esculentoside A-induced hepatotoxicity in rats.

Esculentoside A (EsA) is a kind of triterpenoid saponins from the root tuber of Phytolacca acinosa Roxb. It has extensive medicinal activity, such as antibacterial, anti-inflammatory, immune regulation, and cell proliferation inhibition. However, some researches suggested that EsA can cause hepatotoxicity, whose mechanism is not precise. To ensure the safety and reliability in the clinical use of Phytolacca acinosa Roxb., it is necessary to establish a rapid and accurate method to evaluate the toxicity, analyze and verify the toxicity mechanism of EsA. Therefore, this research explored the mec

Computational screening for potential drug candidates against the SARS-CoV-2 main protease.

Background: SARS-CoV-2 is the causal agent of the current coronavirus disease 2019 (COVID-19) pandemic. They are enveloped, positive-sense, single-stranded RNA viruses of the Coronaviridae family. Proteases of SARS-CoV-2 are necessary for viral replication, structural assembly, and pathogenicity. The approximately 33.8 kDa M pro protease of SARS-CoV-2 is a non-human homologue and is highly conserved among several coronaviruses, indicating that M pro could be a potential drug target for Coronaviruses. Methods: Herein, we performed computational ligand screening of four pharmacophores (OEW, remd
