A multi-centre, double-blind trial was carried out in general practice to compare the effectiveness and tolerability of clomipramine and diazepam in patients with agoraphobia or social phobia. Patients were allocated at random to receive one or other of the trial drugs over a period of 12 weeks and dosage was adjusted to need. During the last 4 weeks daily dosage ranged from 25 to 150 mg clomipramine and from 10 to 30 mg diazepam. A characterization of the benzodiazepine receptor in vivo was carried out utilizing the up-and-down method for a quantitative determination of the maximal seizure threshold to diazepam. The efficacy of diazepam in attenuation of succinylcholine-induced muscle fasciculations was studied in the dog. Dosage levels of 0.11, 0.22, 0.55, or 1.1 mg of diazepam/kg of body weight were studied in 4 groups of 5 dogs each; another group of 5 dogs served as a control. There was no difference in the occurrence or severity of succinylcholine-induced muscle fasciculations between dogs given diazepam and nontreated controls. Using serum potassium as a quantitative index of physiologic effects of muscle depolarization, differences could not be detected between treated and nontreated dogs.

Plasma levels of acetaminophen (paracetamol) and diazepam were measured in 9 children by gas chromatography after administering these drugs simultaneously in separate suppositories. The antipyretic effects of oral and rectal acetaminophen-diazepam combinations were also studied and compared with that of oral or rectal acetaminophen alone. After their first episode of febrile convulsions, 195 previously healthy children, aged 6--30 months, were given either diazepam or phenobarbitone for a year. Twenty infants aged 1-2 years who had previously had one attack of febrile convulsions were randomly given a single dose of diazepam rectally, either as a solution (0.7 mg/kg) or by suppository (5 mg). Plasma-diazepam levels were determined repeatedly during the first hour using gas chromatography. Fifteen student volunteers participated in a study to determine the duration of amnesia obtained with diazepam and with pentazocine. The group that received pentazocine experienced only slight sedation and no amnesia. There was extreme variance in the level of sedation and in the incidence and duration of amnesia in the group that received diazepam. When amnesia occurred, it lasted for an average of 24 minutes. All students in the group that received a combination of diazepam and pentazocine were well sedated; the average duration of amnesia was 25 minutes.
