Maxillonasal dysplasia (Binder's syndrome). A study of craniofacial morphology, associated malformations and familial relations.

The aims of the present thesis were to study the craniofacial morphology and growth pattern in individuals with maxillonasal dysplasia, to check the presence of associated malformations and to elucidate the etiological factors. The thesis was based on five investigations. A cross-sectional cephalometric study of the craniofacial morphology of 97 Binder-subjects was performed. Comparison of 36 variables measured was made between these subjects and a control material, consisting of children with orthognathic profile and normal occlusion, matched for sex and age. Highly significant differences we

The relationship of Epstein-Barr virus to infection-related (sporadic) and familial hemophagocytic syndrome and secondary (lymphoma-related) hemophagocytosis: an in situ hybridization study.

Many cases of hemophagocytic syndrome have been associated with viral infections, particularly Epstein-Barr virus (EBV), but the pathogenesis of the syndrome remains unclear. We have examined lymph node, spleen, liver, and bone marrow sections from 12 cases, including four cases of proven or probable infection-related hemophagocytic syndrome (IHPS), three cases of familial hemophagocytic syndrome (FHPS), and five cases of secondary hemophagocytosis associated with T-cell malignant lymphoma (SHPC), for EBV RNA using a sensitive and specific in situ hybridization technique. Interindividual variability of muscle involvement on imaging in a family with FLNC c.577G>A (p.A193T)-related myopathy.

FLNC-related myopathy (FLNC-RM) with actin-binding domain (ABD) variants is known to show heterogeneous muscle involvement; however, whether such variability exists within families sharing the same variant, and which muscles contribute to this heterogeneity, remain unclear. We investigated muscle involvement patterns in four members of a single family carrying the FLNC c.577G>A (p.A193T) variant who underwent serial longitudinal muscle imaging. A five-point semi-quantitative visual assessment of fatty replacement was performed across 43 muscles using whole-body CT and MRI, covering the shoulde

Familial Risks in 317 000 Patients With Prostate Cancer in Relation to Metastases and Survival-Guiding Diagnostics.

BACKGROUND: Swedish nationwide family and cancer data offer the largest global resource for study of familial cancer. We focus here on familial risks in prostate cancer (PC) with questions on risk in individuals from families of multiple affected members and association of familial risk with metastatic disease and survival. METHODS: Familial relative risk of PC was estimated using standardized incidence ratios (SIRs) for second-generation men with a father or brother affected with PC, considering distinct groups by number and type of affected relatives. RESULTS: Familial SIRs ranged from 2.2

Outcomes of living related kidney donors of recipients with familial mediterranean fever-associated Amyloid A Amyloidosis.

BACKGROUND: Living kidney donation among carefully selected individuals is generally considered safe but post-donation health risks vary by factors including family history. Data on long-term outcomes of related living kidney donors of recipients with familial Mediterranean fever (FMFLRD) are limited. METHODS: This retrospective multicenter study reviewed the medical records of living donors who donated between 2010 and 2021 across six transplantation centers. Cardiovascular and kidney outcomes were compared between 37 FMFLRD and a propensity-matched control group of 74 donors to recipients w

Expansion of the Phenotypic Spectrum of TNRC6B-Related Neurodevelopmental Disorder in a Three-Generation Family with 22q13.1 Deletion.

Background:TNRC6B encodes a core effector of the RNA-induced silencing complex and is essential for miRNA-mediated gene silencing. Pathogenic variants in TNRC6B have recently been associated with a neurodevelopmental disorder characterised by developmental delay, intellectual disability, and behavioural difficulties. Methods: We report a three-generation family with a 22q13.1 deletion encompassing only exons 2-23 of TNRC6B. Clinical data were collected from medical records and family interviews, and the findings were compared with those of published cohorts. Results: Affected individuals prese

Partial Oculomotor Nerve Palsy Revealing PDCD10-Related Familial Cerebral Cavernous Malformation in a Child.

Familial cerebral cavernous malformation (FCCM) is a hereditary vascular disorder associated with pathogenic variants of KRIT1 (CCM1), CCM2, and PDCD10 (CCM3). Among these, PDCD10-related FCCM is considered the most severe subtype and is characterized by early onset, multiple lesions, frequent brainstem involvement, and an increased risk of intracranial hemorrhage. Brainstem symptoms may be the initial manifestations. However, isolated oculomotor nerve palsy in childhood is rare. We report the case of a 17-month-old girl with normal developmental milestones who developed mild right ptosis, str

Cross-Family Mechanistic Analysis of Plant Alkaloids Against Neglected Arboviruses and Related RNA Viruses.

Neglected arboviruses dengue (DENV), Zika (ZIKV), yellow fever (YFV), Japanese encephalitis (JEV), and chikungunya collectively affect hundreds of millions of people annually, yet no specific antiviral drug has been approved for any of them. Alkaloids, nitrogen-containing specialized metabolites produced by diverse plant families, have emerged as a promising source of broad-spectrum antiviral scaffolds. This review compiles and critically analyzes over 100 alkaloid-virus pairs across several RNA virus families, providing a comparative mechanistic analysis. Lycorine, narciclasine, emetine, and

Inhibition of the Parkinson's Disease-Related Protein DJ-1 by Endogenous Neurotoxins of the 1,2,3,4-Tetrahydroisoquinoline Family.

The protein DJ-1 appears to play a protective role in the development of Parkinson's disease (PD). Here, we show that endogenous neurotoxins of the 1,2,3,4-tetrahydroisoquinoline family (TIQs), formed upon reaction of various aldehydes such as methylglyoxal (MGO) with the neurotransmitter dopamine, act as irreversible inhibitors of the esterase activity of human DJ-1, with IC50 values between 15 and 57 μM. The presence of a catechol function appears to be essential for these inhibitory effects, which may be at the origin of the oxidation of cysteine 106, a crucial residue in the DJ-1 active si

Angiopoietin-Like Protein Family-Mediated Functions in Modulating Triglyceride Metabolism and Related Metabolic Diseases.

Hypertriglyceridemia, characterized by increased triglyceride (TG) concentrations, is considered the most important risk factor for cardiometabolic disorders, including dyslipidemia, atherosclerotic cardiovascular diseases, and non-alcoholic fatty liver disease (NAFLD). Recently, the angiopoietin-like protein (ANGPTL) family, which comprises ANGPTL1 to ANGPTL8, was confirmed to play an important role in modulating lipoprotein lipase (LPL) activity. However, understanding of the underlying mechanisms remains limited. Importantly, emerging evidence has linked several transcriptional and post-tra

Advancements in the understanding of mechanisms of the IL-6 family in relation to metabolic-associated fatty liver disease.

As lifestyle patterns change, the rates of type 2 diabetes and obesity are increasing together, leading to an increase in metabolic-associated fatty liver disease (MAFLD), now recognized as the most frequently occurring liver disease globally. MAFLD presents a significant threat to public health and imposes a substantial socioeconomic burden. This condition encompasses a spectrum of hepatic manifestations, beginning with excessive fat accumulation and hepatic steatosis, and possibly progressing to non-alcoholic steatohepatitis (NASH), liver fibrosis, cirrhosis, and liver cancer. The pathogenes
