Expression of neurofibromatosis 2 transcript and gene product during mouse fetal development.

Neurofibromatosis 2 (NF2) is an autosomal dominant inherited disorder that predisposes to benign tumors of the nervous system as well as a variety of ocular abnormalities. In contrast to NF1, NF2 is associated with only minor developmental abnormalities. The human NF2 gene encodes a tumor suppressor protein, termed schwannomin or merlin, which is a member of a superfamily of proteins thought to link cytoskeletal elements to cell membrane components. To determine the pattern of NF2 gene expression in mouse embryos, we sequenced the mouse NF2 gene and used in situ hybridization and antischwannom

Development of gamma delta T-cell subsets from fetal hematopoietic stem cells.

Hematopoietic stem cells (HSC) were isolated from mouse fetus, and their developmental potential was compared with adult HSC. Donor-derived V gamma 3+T cells were detected in fetal thymic lobes, repopulated in vitro with fetal liver HSC, but not in those with adult bone marrow HSC. Single clonogenic fetal HSC gave rise to thymic progeny that include V gamma 3+, other gamma delta+, and alpha beta+ T cells. No V gamma 3+ T cells were detected in adult thymus injected intrathymically with either fetal or adult HSC. These results support a hypothesis that only fetal HSC have the capacity to differ

Expression of the 43 kDa papain inhibitor during human fetal skin development.

The presence of the 43 kDa papain inhibitor protein in the skin of 40 fetuses with the gestational age varying from 9 to 40 weeks was studied. Immunohistochemistry showed the first evidence of the 43 kDa papain inhibitor in the acral parts, mostly in the nail bed, at the 14th week when the epidermis in all other sites was not stained. Staining of the follicular infundibulum as well as some parts of the uppermost epidermis and keratin layer was observed from 17 to 40 weeks. The staining was most intensive at 20-30 weeks gestational age, and later the intensity gradually decreased so that in the

A Rare Case of Fetal Cardiac Hypertrophy Developing into Acute Circulatory Insufficiency and Fetal Compromise in Type 1 Diabetic Pregnancy.

Fetal cardiac hypertrophy (CH) in pregnant women with diabetes is believed to be a benign condition. We encountered a rare case of fetal CH in a pregnant woman with type 1 diabetes, which developed into severe fetal circulatory insufficiency and acidemia. Fetal echocardiography at 37-week gestation showed cardiomegaly with a ventricular hypertrophy. Cardiac function was impaired, and pulsed Doppler findings indicated circulatory failure. The patient was diagnosed with fetal compromise due to fetal CH, and a large for gestational age boy was delivered by an urgent cesarean section. Despite myoc

Expression of carbonic anhydrase IX in human fetal joints, ligaments and tendons: a potential marker of mechanical stress in fetal development?

Carbonic anhydrase type IX (CA9) is known to express in the fetal joint cartilage to maintain pH against hypoxia. Using paraffin-embedded histology of 10 human fetuses at 10-16 weeks of gestation with an aid of immunohistochemistry of the intermediate filaments, matrix components (collagen types I and II, aggrecan, versican, fibronectin, tenascin, and hyaluronan) and CA9, we observed all joints and most of the entheses in the body. At any stages examined, CA9-poisitive cells were seen in the intervertebral disk and all joint cartilages including those of the facet joint of the vertebral column

[Assessment of fetal lung maturity using newly developed immunological measurement of fetal pulmonary surfactant apoprotein-A in amniotic fluid].

The level of immunoreactive lung surfactant apoprotein A (SP-A) was determined with a newly developed one-step ELISA kit (TDR-20) in 217 samples obtained by transabdominal or transvaginal amniocentesis from 217 pregnant women with high risk pregnancy. The lecithin/sphingomyelin ratio (L/S ratio) by two-dimensional thin layer chromatography, disaturated phosphatidylcholine (DSPC) values, shake test and the stable microbubble method were done simultaneously to compare diagnostic reliability in estimating fetal lung maturity. Amniotic fluid SP-A levels increased with advancing gestational age and

Association of proteins involved in fetal lung development with respiratory outcomes in infancy.

OBJECTIVE: We aimed to assess whether proteins which are involved in fetal lung development but also have an additional role in tissue or airway remodeling and oxidative stress response are associated with lung function and respiratory symptoms in infants. STUDY DESIGN: We performed hierarchical clustering to determine distinct profiles of nine cord blood proteins (growth factors, metalloproteinases, and autophagy markers) in 369 healthy infants born to non-asthmatic mothers from the Basel-Bern Infant Lung Development cohort (BILD). Using linear and negative binomial regression analysis, we e

Fetal Echocardiographic Features for the Prediction of the Development of Aortic Coarctation in the Perinatal Period.

PURPOSE: Aortic coarctation (CoA) is a congenital structural anomaly with one of the lowest detection rates among heart diseases. Improving diagnostic rates allows for the centralization of high-risk cases in hospitals with pediatric cardiac surgery facilities, thereby reducing neonatal mortality and morbidity. This study aims to evaluate new diagnostic protocols to enhance the clinical and outpatient management of suspected CoA cases. METHODS: A retrospective analysis was conducted, focusing on various echocardiographic parameters to identify the most predictive indicators for CoA. RESULTS:

The effects of therapeutic drugs on brain development in infants with fetal growth restriction.

Fetal growth restriction (FGR) refers to a fetus that does not reach expected full growth, secondary to placental dysfunction and reduced oxygen and nutrient availability. FGR is associated with mortality or morbidity, including suboptimal brain development. Infants with FGR have reduced brain volume and diffuse, multicellular neuropathology that mediates neurological dysfunctions. In utero, reduced fetal oxygenation initiates a haemodynamic response to preferentially direct cardiac output towards the brain via cerebral vasodilatation, termed 'brain sparing'. This brain sparing response is sus

The Impact of Low-Protein Diet on the Molecular and Cellular Development of the Fetal Kidney.

KEY POINTS: Maternal low protein diet reduced kidney size and altered branching without changing ureteric tip number, with fewer nephrons at birth and adulthood. Nephron progenitor proliferation and commitment were impaired, pretubular aggregate formation decreased, consistent with a reduced nephron number. BACKGROUND: Low nephron number has a direct effect on the development of hypertension and CKD later in life. Although intrauterine growth restriction caused by maternal low-protein diet (LPD) is believed to be a significant cause of reduced nephron endowment in impoverished communities, it

Shaping immunity: the influence of the maternal gut bacteria on fetal immune development.

The development of the fetal immune response is a highly complex process. In the present review, we describe the development of the fetal immune response and the role of the maternal gut bacteria in this process. In contrast to the previous belief that the fetal immune response is inert, it is now thought that the fetal immune response is uniquely tolerant to maternal and allo-antigens, but able to respond to infectious agents, such as bacteria. This is accomplished by the development of T cells toward regulatory T cells rather than toward effector T cells, but also by the presence of function

Assessing in-vitro models for microglial development and fetal programming: a critical review.

This review evaluates in-vitro models for studying how maternal influences during pregnancy impact the development of offspring microglia, the immune cells of the central nervous system. The models examined include primary microglia cultures, microglia cell lines, iPSC-derived microglia, PBMC-induced microglia-like cells, 3D brain organoids derived from iPSCs, and Hofbauer cells. Each model is assessed for its ability to replicate the in-vivo environment of the developing brain, with a focus on their strengths, limitations, and practical challenges. Key factors such as scalability, genetic and

Relationship of parental phthalate exposure with fetal growth and placental development at birth.

BACKGROUND: Prenatal exposure to phthalates is reported to influence fetal growth and may lead to lasting adverse effects on infants and their future development; yet, the results remain inconclusive. OBJECTIVE: This study utilized a birth cohort of 73 pregnant women-newborn pairs, including biological fathers (73 triads), to investigate the relationship between parental phthalate exposure during pregnancy and birth outcomes in newborns. METHODS: Demographic, behavioral, and clinical information, along with urine samples from both parents, were collected prior to delivery. Sixteen phthalate

Fetal development and the air pollution exposome: an integrative perspective of health pathways.

We offer an integrative perspective on how the air-pollution exposome shapes fetal development during the first 1,000 days and reverberates across mental health and behavior. Pregnant individuals and young children are disproportionately exposed to particulate matter (PM2.5), nitrogen dioxide (NO2), ozone (O3), and volatile organic compounds (VOCs) with social disadvantage amplifying risk. We bridge exposure to biology through three conduits. First, the placenta acts as a sensor and recorder, transducing signals that alter growth, immune tone, and neuroendocrine programming. Second, fetal auto…

Interactions of the maternal microbiome with diet, stress, and infection influence fetal development.

Humans and other animals contain multitudes of microorganisms including bacteria, fungi, and viruses, which make up a diverse microbiome. Across body sites including skin, gastrointestinal tract, and oral cavity there are distinct microbial niches that are made up of trillions of microorganisms that have co-evolved to inhabit and interact with the host. The microbiome also interacts with the changing environment. This tripartite interaction between the host, microbiome, and environment suggests microbial communities play a key role in the biological processes of the host, such as development a
