3D Printed Silk Fibroin-Gelatin Construct Targeting BMP Signaling for Articular Cartilage Regeneration Within a Full-Thickness Cartilage Defect in a Surgically Induced Osteoarthritic Rat Knee Joint.

Regenerating articular cartilage within osteoarthritis (OA)-affected joints remains a long-standing challenge. This study introduces a novel chemically modified silk-based biomaterial to modulate key signaling pathways involved in OA and cartilage repair. The designed biomaterial concurrently inhibits the bone morphogenetic protein (BMP) signaling, aberrantly activated during OA, and inherently facilitates Wnt/β-catenin signaling, a crucial factor for chondrogenesis and cartilage homeostasis. The dual modulation was achieved by covalently conjugating the potent BMP inhibitor LDN-193189 to the

Topical rhubarb charcoal-crosslinked chitosan/silk fibroin sponge scaffold for the repair of diabetic ulcers improves hepatic lipid deposition in db/db mice via the AMPK signalling pathway.

BACKGROUND: Type 2 diabetes mellitus (T2DM) is closely linked to metabolic syndrome, characterised by insulin resistance, hyperglycaemia, abnormal lipid metabolism, and chronic inflammation. Diabetic ulcers (DUs) comprise consequential complications that arise as a result of T2DM. To investigate, db/db mice were used for the disease model. The findings demonstrated that a scaffold made from a combination of rhubarb charcoal-crosslinked chitosan and silk fibroin, designated as RCS/SF, was able to improve the healing process of diabetic wounds in db/db mice. However, previous studies have primar

Controlled release of stromal cell-derived factor-1α from silk fibroin-coated coils accelerates intra-aneurysmal organization and occlusion of neck remnant by recruiting endothelial progenitor cells.

This study is to test the efficacy of stromal cell-derived factor-1α (SDF-1α)-coated coils together with endothelial progenitor cells (EPCs) transplantation in occluding aneurysms. Bone marrow-derived EPC surface markers were analyzed using flow cytometry. The migratory function of EPCs in response to SDF-1α was evaluated using a modified Boyden chamber assay. Capillary-like tube formation was assessed using Matrigel gel. Coil morphologies before and after coating with SDF-1α were observed under a scanning electron microscope. The level of SDF-1α in supernatants was measured by ELISA. Sprague-

Isolation, characterization, antimicrobial and cytotoxic properties of Cydalima perspectalis silk proteins.

Natural silk produced by silkworms and some arthropod spiders stands out as a traditional protein polymer. Fibroin and sericin, which are silk proteins obtained from cocoons, have been used in the medical field for centuries. This study is the first to evaluate the physicochemical properties and biological activities of these proteins, which were isolated from the boxwood moth (Cydalima perspectalis), as a biotechnological material. This followed this separation of the fibroin and sericin proteins obtained from the cocoons using a high-temperature and high-pressure method. The morphological, t

Chemical, Thermal, Time, and Enzymatic Stability of Silk Materials with Silk I Structure.

The crystalline structure of silk fibroin Silk I is generally considered to be a metastable structure; however, there is no definite conclusion under what circumstances this crystalline structure is stable or the crystal form will change. In this study, silk fibroin solution was prepared from B. Mori silkworm cocoons, and a combined method of freeze-crystallization and freeze-drying at different temperatures was used to obtain stable Silk I crystalline material and uncrystallized silk material, respectively. Different concentrations of methanol and ethanol were used to soak the two materials w

Silk-derived sericin/fibroin mixture drink fermented with plant-derived Lactococcus lactis BM32-1 improves constipation and related microbiota: a randomized, double-blind, and placebo-controlled clinical trial.

We previously showed through clinical trials that one plant-derived lactic acid bacteria (LAB) can improve constipation. We preliminarily found that the plant-derived LAB Lactococcus lactis BM32-1 can grow in a mixture of sericin and fibroin, which are extracted from silk and have been reported to help promote health. Thus, in the present study, we evaluated the favorable effect of a sericin/fibroin mixture (S/F-M), which was extracted from silk prepared from cocoons reared in an aseptic rearing system using an artificial diet, fermented with the BM32-1 strain through a clinical trial. The tri

Drug-loaded mucoadhesive microneedle patch for the treatment of oral submucous fibrosis.

Oral submucous fibrosis is a chronic, inflammatory and potentially malignant oral disease. Local delivery of triamcinolone to lesion site is a commonly used therapy. The existing methods for local drug delivery include topical administration and submucosal injection. However, in the wet and dynamic oral microenvironment, these methods have drawbacks such as limited drug delivery efficiency and injection pain. Therefore, it is urgently needed to develop an alternative local drug delivery system with high efficiency and painlessness. Inspired by the structure of band-aid, this study proposed a n

Polydopamine-functionalized silk-based biomaterial for non-antibiotic antimicrobial therapy.

Silk fibroin is a highly promising biomaterial for wound dressings; however, its lack of inherent antibacterial activity limits broader clinical adoption. In this study, robust antibacterial properties were successfully conferred onto electrospun silk fibroin nanofiber mats using a mussel-inspired polydopamine (PDA) dip-coating strategy. Primary dip-coating deposited a uniform PDA nanolayer that served as an adhesive scaffold, enabling subsequent anchoring of dopamine nanoparticles during secondary coating. This hierarchical structure markedly increased surface roughness and reactive catechol

Silk-based biomaterials for promoting spinal cord regeneration: A review.

The management of neurological disorders is profoundly complicated by spinal cord injury (SCI), which leads to the impairment of motor and sensory functions. A major challenge in the treatment of SCI is the formation of a dysfunctional pathological microenvironment characterized by an excessive inflammatory response, deposition of inhibitory molecules, glial scarring, and vascular dysfunction. A thorough understanding of the pathological and physiological changes following SCI is essential to elucidate the mechanisms underlying functional recovery and to develop effective therapeutic intervent

Silk Acid as an Implantable Biomaterial for Tissue Regeneration.

Silk fibroin derived from the domesticated silkworm Bombyx mori is a protein-based biopolymer with low immunogenicity, intrinsic biodegradability, and tunable mechanical properties, showing great potential in biomedical applications. Using chemical modification to alter the primary structure of silk fibroin enables the expanded generation of new silk-based biomaterials. Inspired by the molecular structure of hyaluronic acid, which is enriched in carboxyl groups, an efficient method with scaling-up potential to achieve controlled carboxylation of silk fibroin to prepare silk acid (SA) is report
