A large proportion of nutrient cycling and ecosystem services are supported by the activities of fungal communities. Though these communities have been comparatively understudied compared to more charismatic groups, there is a growing body of evidence that the structure of these communities is also impacted by urbanization. To investigate diversity patterns, we analyzed fungal occurrences recorded in California taken between 2015 and 2024. We investigated their differences in both richness and turnover by comparing communities sampled along a gradient of percent impervious coverage as a proxy

Coronavirus disease 2019 (COVID-19) can become complicated by secondary invasive fungal infections (IFIs), stemming primarily from severe lung damage and immunologic deficits associated with the virus or immunomodulatory therapy. Other risk factors include poorly controlled diabetes, structural lung disease and/or other comorbidities, and fungal colonization. Opportunistic IFI following severe respiratory viral illness has been increasingly recognized, most notably with severe influenza. There have been many reports of fungal infections associated with COVID-19, initially predominated by pulmo

The ocular surface represents a unique microbial ecosystem continuously exposed to environmental microorganisms; however, the fungal component of the healthy equine ocular microbiome remains poorly characterized. This study aimed to characterize the ocular surface mycobiome of 12 clinically healthy Purebred Lusitano horses from three geographical regions in Portugal kept in two housing systems using Oxford Nanopore long-read sequencing and conventional fungal culture. Sequencing revealed a highly diverse fungal community with marked inter-individual variability. The most abundant genera were W

Fungal pathogens have historically been studied as autonomous agents, as self-contained units independent of their associated microbiota, with disease outcomes attributed to fungal virulence factors acting on susceptible hosts. However, many pathogenic fungi harbor bacterial associates that influence their ecology, persistence and virulence. The pathobiome concept addressed this limitation by framing disease as an outcome of multi-kingdom interactions. Yet most pathobiome studies rely on correlative analyses of microbial abundances with pathogens, lacking spatial resolution to identify which m

Hemibiotrophic fungi of the genus Colletotrichum provide valuable systems for studying plant immunity and pathogen lifestyle transitions; however, compatible and incompatible interactions have rarely been compared within the same host species. Here, we show that Arabidopsis thaliana exhibits contrasting responses to two phylogenetically distinct hemibiotrophic fungi, Colletotrichum coccodes KACC40803 and Colletotrichum scovillei KC05. Multilocus phylogenetic analyses using ITS, ACT, GAPDH, and TUB2 sequences confirmed their taxonomic identities. The two fungi also differed morphologically, par

Multidrug-resistant fungi, Candida auris, persist in the healthcare environment and colonize human skin and medical devices, causing life-threatening infections with mortality rates approaching ~60% in immunocompromised patients. Because some isolates are resistant to nearly all available antifungal drugs, alternative therapeutic approaches are urgently needed. C. auris cell wall contains β-glucan and highly conserved fungal glycan structures. Moreover, many cell wall proteins are heavily β-glycosylated, and a monoclonal antibody raised against β-glucan has been shown to recognize C. albicans

Colletotrichum species are important fungal pathogens causing anthracnose of tropical and subtropical fruit and vegetable crops. Dual culture assay indicated that Bacillus velezensis CE 100 was a strong antagonist against C. acutatum, C. coccodes, C. dematium, and C. gloeosporioides. The volatile organic compounds produced by B. velezensis CE 100 affected mycelial growth of Colletotrichum species tested in our study and caused twisted hyphal structures of all these fungal species. Chloroform crude compounds of B. velezensis CE 100 inhibited four Colletotrichum species in a concentration-depend

From 2014-2019, 8 juvenile black bears (Ursus americanus) from different geographic regions were presented to the California Department of Fish and Wildlife because of emaciation, alopecia, and exfoliative dermatitis that resulted in death or euthanasia. Autopsy and histopathology revealed that all 8 bears had generalized hyperkeratotic dermatitis, folliculitis, and furunculosis. Skin structures were heavily colonized by fungal hyphae and arthrospores; fungal cultures of skin from 7 bears yielded Trichophyton equinum, a zoophilic dermatophyte reported only rarely in non-equid species. G143A mutation-driven target-site resistance severely impairs the field efficacy of traditional Quinone outside inhibitors (QoIs). To solve this agricultural crisis, novel tetrazolinone-pyrazine derivatives were rationally designed and synthesized by integrating potentially resistance-breaking tetrazolinone toxophore with a pyrazine scaffold via oxime ether linkage. Bioassays revealed that compound 19 exhibited exceptional in vitro activity against Sclerotinia sclerotiorum with an EC50 of 0.003 mg/L, greatly exceeding the commercial QoI metyltetraprole (EC50 = 0.13 mg/L). Currently, non-albicans Candida species, including C. tropicalis, C. glabrata, and C. parapsilosis, are becoming an increasing epidemiological threat, predominantly due to the distinct collection of virulence mechanisms, as well as emerging resistance to antifungal drugs typically used in the treatment of candidiasis. They can produce biofilms that release extracellular vesicles (EVs), which are nanometric spherical structures surrounded by a lipid bilayer, transporting diversified biologically active cargo, that may be involved in intercellular communication, biofilm matrix production, and in

Dispersal is a key force in the assembly of fungal communities and the air is the dominant route of dispersal for most fungi. Understanding the dynamics of airborne fungi is important for determining their source and for helping to prevent fungal disease. This understanding is important in the San Joaquin Valley of California, which is home to 4.2 million people and where the airborne fungus Coccidioides is responsible for the most important fungal disease of otherwise healthy humans, coccidioidomycosis. The San Joaquin Valley is the most productive agricultural region in the United States, wi

Fungal class 3 L-asparaginases were identified based on sequence similarity to the bacterial enzyme ReAV from Rhizobium etli. Here, we present an integrative functional and structural characterization of six class 3 L-asparaginases derived from pathogenic fungi. Kinetic analyses revealed distinct catalytic profiles, including differences in substrate affinity and turnover rates. For three enzymes, a total of six high-resolution crystal structures were determined, capturing multiple active site states. Structural comparisons confirm the conserved fold characteristic of class 3 L-asparaginases a

Emerging evidence demonstrates the evolving role of fungi in the pathophysiology and disease progression observed in bronchiectasis. Fungal-associated traits are linked to disease severity, exacerbation frequency and airway inflammation. Structural abnormalities and impaired mucociliary clearance, characteristic of bronchiectasis, predispose to fungal colonisation, with subsequent immunopathogenic responses dependent on underlying host immunity. The diagnosis of fungal infection remains challenging in clinical settings, owing to the limitations of existing diagnostic modalities; however, the d

Candida albicans is an opportunistic, dimorphic fungus that causes candidiasis in immunocompromised people. C. albicans forms specialized structures called microcolonies that are important for surface adhesion and virulence. Microcolonies form in response to specific environmental conditions and require glycolytic substrates for optimal growth. However, fungal signaling pathways involved in sensing and transmitting these environmental cues to induce microcolony formation have not been identified. Here, we show that the C. albicans Ras1-cAMP cascade is required for microcolony formation, while

Fungal infections are often complicated by biofilm formation and concurrent inflammation, limiting the efficacy of conventional antifungal therapies. To address these challenges, we developed a novel hybrid drug delivery platform-lipidic prodrug co-crystals (LPCCs)-that combines the benefits of lipidic self-assembly and pharmaceutical co-crystallization. In this study, a lipidic prodrug is synthesized by linking catechol-containing α-aminophosphonate with phenylboronic acid-modified bifonazole (Bfz), an antifungal agent, via boronate bonding. The resulting self-assembled structures exhibit hig

Isougenol is a phytoconstituent found in several essential oils. Since many natural products are potent antimicrobials, the synthesis of hybrid molecules-combining the chemical skeleton of the phytochemical with synthetic groups-can generate substances with enhanced biological activity. Based on this, the objective of this study was to evaluate the antifungal activity of isoeugenol and hybrid acetamides against Candida albicans isolated from the oral cavity. The methodologies used were the determination of minimum inhibitory concentration (MIC), minimum fungicidal concentration (MFC), action o

How microtubules (MTs) are regulated during fungal biofilm formation is unknown. By tracking MT +end-binding proteins (+TIPS) in Aspergillus nidulans, we find that MTs are regulated to depolymerize within forming fungal biofilms. During this process, EB1, dynein, and ClipA form transient fibrous and then bar-like structures, novel configurations for +TIPS. Cells also respond in an autonomous manner, with cells separated by a septum able to maintain different MT dynamics. Surprisingly, all cells with depolymerized MTs rapidly repolymerize their MTs after air exchange above the static culture me

BACKGROUND: Climate-driven changes in moisture are expected to intensify environmental selection in soils, yet the extent to which soil texture modifies microbial responses to moisture stress remains poorly understood. Because soil texture governs water retention, nutrient diffusion, and habitat structure, it may fundamentally influence how microbial communities assemble and function under drought. Here, we investigated how moisture and soil texture interact to shape bacterial and fungal rhizosphere communities associated with Andropogon virginicus, a drought-tolerant wild relative of maize, a
