Social Determinants of Health, Genetic Susceptibility, and Risk of Dementia: A Prospective Cohort Study.

INTRODUCTION: The association between social determinants of health, genetic susceptibility, and dementia risk remains unclear. This study investigates their relationship and the potential mediation by chronic conditions. METHODS: Data from 225,598 UK Biobank participants (220,847 for genetic analyses), with an average age of 55.07 years and 51.10% female, were analyzed. The data were collected during 2006-2010 and analyzed in 2025. Social determinants of health scores were weighted, combined, and categorized, and a polygenic risk score for dementia was constructed. Associations were assessed

Genetic determinants of chylomicronemia in Taiwan: Predominance of LPL rare variants and APOA5 p.G185C.

BACKGROUND: Chylomicronemia is a genetically heterogeneous condition with both genetic and environmental determinants. Genetic studies have largely focused on European populations, with limited data from East Asians, particularly Han Chinese. We characterized the genetic profiles of chylomicronemia patients of mainly Han Chinese ancestry in Taiwan. OBJECTIVE: To identify and classify disease-causing variants in chylomicronemia patients. METHODS: Capture-based targeted next-generation sequencing was performed for 108 chylomicronemia patients to assess hypertriglyceridemia-related genes, inclu

Molecular and Genetic Determinants of Nephrocalcinosis: Mechanisms, Genotype-Phenotype Correlations, and Precision Medicine.

Nephrocalcinosis, defined as the deposition of calcium salts within the renal parenchyma, represents a radiologic and pathologic endpoint shared by a broad spectrum of metabolic and monogenic disorders. Advances in genomic medicine have identified more than 30 genes involved in tubular transport, mineral and acid-base homeostasis, oxalate metabolism, mitochondrial function, ciliary signaling, and nephron development, reframing nephrocalcinosis as a heterogeneous manifestation of discrete molecular defects rather than a single disease entity. Despite this diversity, these conditions converge on

Risk factors for frailty in aging: from sex and gender and genetic determinants to the exposome.

Frailty is a multidimensional geriatric syndrome of loss of physical and cognitive reserves and increased vulnerability that leads to an increased risk of adverse health outcomes. Although age is recognized as the strongest driver of frailty, dynamic nature of this syndrome enables bidirectional changes in the frailty continuum, implying that this syndrome might be prevented and even reverted when detected in early stages. The pathophysiological changes underlying frailty syndrome are initiated by prolonged exposure to a series of risk factors, in a downward spiral of events leading to manifes

Evolutionary regimes determine the accuracy of epistasis inference from temporal genetic data.

Epistasis, the non-additive effects of mutations, shapes fitness landscapes and evolutionary trajectories. Temporal genetic data reveal evolutionary dynamics and could be used to infer epistatic interactions, especially through linkage disequilibrium (LD) between interacting mutations. However, other evolutionary forces can also generate LD, challenging inference. Here, we systematically evaluated the accuracy of a variety of epistasis inference approaches across a range of selective pressures, recombination rates, and population sizes. In general, we found that inference accuracy depends on t

Genetic Determinants of Treatment-Related Bone Toxicity in Pediatric Acute Lymphoblastic Leukemia.

Osteonecrosis and fractures are serious corticosteroid-induced bone toxicities in children treated for acute lymphoblastic leukemia, yet their genetic determinants remain incompletely defined. In this study, we aimed to identify novel genetic contributors to bone toxicity and to evaluate the robustness of both newly identified and previously established risk genotypes in more recent Dana-Farber Cancer Institute treatment protocols. Whole-exome sequencing was first performed in a discovery cohort to identify genetic variants associated with osteonecrosis. A novel association with a variant in t

Epigenetic determinants of GLP-1 responsiveness: Integrating dietary modulation beyond genetics.

Glucagon-like peptide-1 receptor (GLP-1R) agonists have become major therapeutic agents for obesity and type 2 diabetes, yet clinical responses vary substantially among individuals. Although genetic variation contributes to this heterogeneity, it does not fully explain differences in glycemic control, body weight reduction, or broader metabolic outcomes. This narrative review discusses diet-driven epigenetic regulation as a potential framework linking modifiable nutritional exposures to GLP-1 physiology and variability in GLP-1R agonist responsiveness. Endogenous GLP-1 biology and the pharmaco

Genetic determinants that permit growth without the core septin Cdc12 in Cryptococcus neoformans.

Septins are conserved cytoskeletal GTPases that regulate morphogenesis, cytokinesis, and cell-surface organization in fungi. In most model and pathogenic yeasts, including Saccharomyces cerevisiae and Candida albicans, core septin subunits such as Cdc3 and Cdc12 are essential and their loss results in lethal cytokinetic defects. In Cryptococcus neoformans, by contrast, deletion of CDC12 or CDC3 yields viable cells that grow normally under permissive conditions, raising the question of how this pathogen maintains cell integrity without a functional septin complex. To address this, we compared t

Genetic Determinants of Multiple Sclerosis Susceptibility in People From Diverse Ancestral Backgrounds.

BACKGROUND AND OBJECTIVES: The genetic basis of multiple sclerosis (MS) susceptibility has been studied extensively in European (EUR) ancestry populations. The aim of our study was to determine the genetic architecture of MS susceptibility in people of South Asian (SAS) and African (AFR) genetic ancestral backgrounds. METHODS: We recruited and genotyped a cohort of ancestrally diverse people with MS (pwMS) from across the United Kingdom. Cases were combined with controls from the UK Biobank (UKB). After genetic ancestry inference, we performed within-ancestry case-control genetic association

Elevated Blood Pressure: A Genetically Determined Risk Factor for Cerebral Artery Dissection.

BACKGROUND: We aim to investigate the potential causal link between blood pressure (BP) levels and cerebral artery dissection (CAD) risk by employing a 2-sample Mendelian randomization (TSMR) framework. METHODS: Utilizing large-scale genome-wide association studies-retrieved data, we employed various Mendelian randomization (MR) techniques, including inverse variance weighted (IVW), MR-Egger regression, weighted median, and weighted mode, to ascertain BP's causal impact on CAD. The MR-Egger intercept was calculated to assess pleiotropy presence, determining heterogeneity by Cochran's Q statis

Cardiac sympathetic neurons are additional cells affected in genetically determined arrhythmogenic cardiomyopathy.

Arrhythmogenic cardiomyopathy (AC) is a familial cardiac disease, mainly caused by mutations in desmosomal genes, which accounts for most cases of stress-related arrhythmic sudden death, in young and athletes. AC hearts display fibro-fatty lesions that generate the arrhythmic substrate and cause contractile dysfunction. A correlation between physical/emotional stresses and arrhythmias supports the involvement of sympathetic neurons (SNs) in the disease, but this has not been confirmed previously. Here, we combined molecular, in vitro and ex vivo analyses to determine the role of AC-linked DSG2

Causality of genetically determined glucosamine supplementation on cognition and sarcopenia: a Mendelian randomization study.

BACKGROUND: Evidence indicates a negative link between glucosamine and age-related cognitive decline and sarcopenia. However, the causal relationship remains uncertain. This study aims to verify whether glucosamine is causally associated with cognitive function and sarcopenia. METHOD: Forty-eight genetic variants linked to glucosamine use were extracted from the MRC-IEU consortium. Besides, we gathered cognition proxy indicators [cognitive performance and fluid intelligence score (FIS)], and sarcopenia-related indicators, namely, appendicular lean mass (ALM), whole body fat-free mass (WBFM),

Genetic background affects neutrophil activity and determines the severity of autoinflammatory osteomyelitis in mice.

The knowledge about the contribution of the innate immune system to health and disease is expanding. However, to obtain reliable results, it is critical to select appropriate mouse models for in vivo studies. Data on genetic and phenotypic changes associated with different mouse strains can assist in this task. Such data can also facilitate our understanding of how specific polymorphisms and genetic alterations affect gene function, phenotypes, and disease outcomes. Extensive information is available on genetic changes in all major mouse strains. However, comparatively little is known about th

Prevalence of Trehalase Enzymopathy Genetic Determinants in Siberian and Russian Far East Populations.

UNLABELLED: To date, it has been established that the patient's genotype plays a significant role in the formation of trehalase enzymopathy: the level of enzyme activity decreases when the G→A allele replacement occurs in the rs2276064 locus of the TREH gene. To assess the prevalence of trehalase deficiency, extensive population-based studies are needed. Clinical observations show that the reduced activity of bowel trehalase is more common in the Arctic than in European populations. The aim of this research was to analyze the frequency of the alleles and variants of trehalase gene (rs2276064 T

Genetic determinants of host- and virus-derived insertions for hepatitis E virus replication.

Hepatitis E virus (HEV) is a long-neglected RNA virus and the major causative agent of acute viral hepatitis in humans. Recent data suggest that HEV has a very heterogeneous hypervariable region (HVR), which can tolerate major genomic rearrangements. In this study, we identify insertions of previously undescribed sequence snippets in serum samples of a ribavirin treatment failure patient. These insertions increase viral replication while not affecting sensitivity towards ribavirin in a subgenomic replicon assay. All insertions contain a predicted nuclear localization sequence and alanine scann

Genetic determinants of individual variation in the superior temporal sulcus of chimpanzees (Pan troglodytes).

The superior temporal sulcus (STS) is a conserved fold that divides the middle and superior temporal gyri. In humans, there is considerable variation in the shape, folding pattern, lateralization, and depth of the STS that have been reported to be associated with social cognition and linguistic functions. We examined the role that genetic factors play on individual variation in STS morphology in chimpanzees. The surface area and depth of the STS were quantified in sample of 292 captive chimpanzees comprised of two genetically isolated population of individuals. The chimpanzees had been previou

On the determinants and the role of the payers in the uptake of genetic testing and data sharing in personalized health.

BACKGROUND: New health technologies and data offer tailored prevention and spot-on treatments, which can considerably reduce healthcare costs. In healthy individuals, insurers can participate in the creation of health capital through data and preventing the occurrence of a disease. In the onset of a disease, sequencing an individual's genome can provide information leading to the use of more efficient treatments. Both improvements are at the core of the "personalized health" paradigm. As a positive side effect, a reduction in healthcare costs is expected. However, the integration of personaliz
