Characterization of key BMP signaling molecules in the swimming crab (Portunus trituberculatus) and their expression characteristics during ovarian development.

The bone morphogenetic protein (BMP) signaling pathway is critically involved in regulating ovarian development and reproduction in animals. However, its molecular characteristics and expression patterns in Portunus trituberculatus remain poorly understood. In this study, the coding sequences (CDS) of five key components (BMP2, BMPR1B, BMPR2, Smad4, and Smad5) of the BMP signaling pathway were analyzed, and their tissue distribution across ovarian developmental stages I-VI was investigated. Bioinformatic analysis revealed that these proteins contain conserved functional domains characteristic

How the Mediterranean mussel Mytilus galloprovincialis responds to multiple environmental challenges: Vibrio splendidus and hydrocarbon mixture.

Marine organisms are exposed to a wide range of environmental stressors and must develop adaptive responses to survive. This study investigated how Mytilus galloprovincialis responds to chemical and biological stress, both separately and simultaneously. Mussels were exposed for 4 days to a hydrocarbon mixture (diesel + oil engine) and then injected with Vibrio splendidus. Six hours post-injection, responses were assessed by measuring phagocytosis, several immune-related enzymes (lysozyme, phenoloxidase, glutathione peroxidase) and chaperonins (HSP70, HSC70) activities. The contact with the pat

BmSerpin-7 negatively regulates prophenoloxidase activation and antimicrobial peptide synthesis in Bombyx mori.

The prophenoloxidase (PPO) activation pathway and the Toll-mediated antimicrobial peptide (AMP) synthesis pathway constitute critical immune responses within the insect immune system. The activation of these pathways is driven by extracellular serine protease cascades, a process counterbalanced by the negative regulation of serpins. In this study, we identified a typical serpin, BmSerpin-7, and characterized its function in the silkworm, Bombyx mori. BmSerpin-7 expression was up-regulated in the hemocytes but down-regulated in the fat body after Micrococcus luteus and Yersinia pseudotuberculos

SpBAG3 assisted WSSV infection in mud crab (Scylla paramamosain) by inhibiting apoptosis.

The function of B-cell lymphoma-2 (Bcl-2) family proteins can be divided into two categories: anti-apoptotic and pro-apoptotic. As an anti-apoptotic protein, Bcl2-associated athanogene 3 (BAG3) plays a key role in regulating apoptosis, development, cell movement, and autophagy, and mediating the adaptability of cells to stimulation. However, SpBAG3 has not been reported in mud crab (Scylla paramamosain), and the regulatory effect of SpBAG3 on apoptosis in mud crab and its function in antiviral immunity is still unknown. In this study, SpBAG3 was found, and characterized, which encoded a total

Dodecanol, metabolite of entomopathogenic fungus Conidiobolus coronatus, affects fatty acid composition and cellular immunity of Galleria mellonella and Calliphora vicina.

One group of promising pest control agents are the entomopathogenic fungi; one such example is Conidiobolus coronatus, which produces a range of metabolites. Our present findings reveal for the first time that C. coronatus also produces dodecanol, a compound widely used to make surfactants and pharmaceuticals, and enhance flavors in food. The main aim of the study was to determine the influence of dodecanol on insect defense systems, i.e. cuticular lipid composition and the condition of insect immunocompetent cells; hence, its effect was examined in detail on two species differing in susceptib

Epigallocatechin-3-gallate inhibit replication of white spot syndrome virus in Scylla paramamosain.

Epigallocatechin-3-gallate (EGCG) has exhibited potential antibacterial and anticancer activities. In this study, we tested whether EGCG can protect the mud crab Scylla paramamosain against white spot syndrome virus (WSSV). Treatment with 1.5 mg/kg EGCG killed crab hemocytes but treatment with 1 mg/kg EGCG did not. Results of the virus challenge experiment confirmed that EGCG could enhance the survival rate of WSSV-challenged crabs, and treatment with 1 mg/kg EGCG significantly increased their resistance to WSSV compared with the control. Thus, 1 mg/kg EGCG was used as the experimental dose in

The functions of serpin-3, a negative-regulator involved in prophenoloxidase activation and antimicrobial peptides expression of Chinese oak silkworm, Antheraea pernyi.

Serpins are a superfamily of proteins engaged in various physiological processes in all kingdoms of life. To date, many striking results have demonstrated serpins are involved in the invertebrate immune system by regulating the proteolytic cascades. However, in most insect species, the immune functions of serpins in response against pathogen invasion remain obscure. In this study, we identified a full-length cDNA sequence of serpin, named serpin-3, from the Chinese oak silkworm Antheraea pernyi. Sequence alignments have indicated that Apserpin-3 might regulate the melanization reaction via inh

Targeting and retention enhancement of quantum dots decorated with amino acids in an invertebrate model organism.

The use of quantum dots (QDs) in biological imaging applications and targeted drug delivery is expected to increase. However, the efficiency of QDs in drug targeting needs to be improved. Here, we show that amino acids linked to CdTe QDs significantly increased the targeted transfer efficiency and biological safety in the invertebrate model Bombyx mori. Compared with bare QDs530, the transfer efficiency of Ala- and Gly-conjugated QDs (QDs530-Ala and QDs530-Gly) in circulatory system increased by 2.6 ± 0.3 and 1.5 ± 0.3 times, and increased by 7.8 ± 0.9 and 2.9 ± 0.2 times in target tissue silk

Deadly venom of Asobara japonica parasitoid needs ovarian antidote to regulate host physiology.

Asobara japonica (Braconidae) is an endophagous parasitoid developing in Drosophila larvae. The present study shows that A. japonica was never encapsulated in Drosophila melanogaster, and that it caused an overall inhibition of the host encapsulation reaction since injected foreign bodies were never encapsulated in parasitized hosts. Both the number of circulating hemocytes and the phenoloxidase activity decreased in parasitized larvae, and the hematopoietic organ appeared highly disrupted. We also found that A. japonica venom secretions had atypical effects on hosts compared to other braconid

Identification and molecular characterization of a mucosal lectin (MeML) from the blue mussel Mytilus edulis and its potential role in particle capture.

Molecular recognition of food particles has been suspected to play an important role in particle selection in suspension feeding bivalves. Lectins are a group of sugar-binding proteins that are widely involved in biological recognition. They have been reported in mucus covering bivalves feeding organs and were recently shown to mediate particle sorting in these animals. In this study, we report a novel putative C-type lectin from the blue mussel Mytilus edulis. The cDNA of this lectin (hereby designated MeML for M. edulis mucocyte lectin) is 459bp long encoding a 152-residue protein. MeML pres

In vitro differentiation of mouse embryonic stem cells after activation by retinoic acid.

Embryonic stem (ES) cells are pluripotent cells isolated from the inner cell mass of blastocysts. ES cells are able to differentiate into the three primitive layers (endoderm, mesoderm, and ectoderm) of the organism, including the germline. In recent reports mouse ES cells have been successfully applied in the treatment of spinal cord injury, hereditary myelin disorder of the central nervous system, and diabetes mellitus. In this study, we investigated the induction of mouse ES cell differentiation, using culture of embryoid bodies (EBs) into the diverse tissues. EBs were formed by culturing E

E3 Ubiquitin Ligase CHIP Inhibits Haemocyte Proliferation and Differentiation via the Ubiquitination of Runx in the Pacific Oyster.

Mollusca first evolve primitive immune cells (namely, haemocytes), which assemble a notable complex innate immune system, which are continuously produced through proliferation and differentiation and infused in the haemolymph. As a typical E3 ligase, CHIP is critical for immune cell turnover and homeostasis in vertebrates. In this study, a CHIP homolog (CgCHIP) with a high expression in haemocytes was identified in oysters to investigate its role in the proliferation and differentiation of ancient innate immune cells. CgCHIP exhibited a widespread distribution across all haemocyte subpopulatio

New insights into the possible multiple roles of histidine-rich glycoprotein in blue mussels.

Histidine-rich Glycoprotein (HRG) is the most abundant protein in mussel haemolymph plasma. In this study, we determined by qRT-PCR and FISH analysis the tissues involved in HRG synthesis in Mytilus galloprovincialis. The relative HRG mRNA abundance in haemocytes, digestive gland, gills, gonads, posterior adductor muscle, and mantle edge was evaluated. Immunofluorescence analysis of HRG protein distribution in the whole mussel body was performed by a specific antibody. Our data showed the highest gene expression level of HRG in the mantle edge. In particular the outer fold of the mantle edge w

DNA duplication is essential for the repair of gastrointestinal perforation in the insect midgut.

Invertebrate animals have the capacity of repairing wounds in the skin and gut via different mechanisms. Gastrointestinal perforation, a hole in the human gastrointestinal system, is a serious condition, and surgery is necessary to repair the perforation to prevent an abdominal abscess or sepsis. Here we report the repair of gastrointestinal perforation made by a needle-puncture wound in the silkworm larval midgut. Following insect gut perforation, only a weak immune response was observed because the growth of Escherichia coli alone was partially inhibited by plasma collected at 6 h after need

Hidden antigens from third instar Hypoderma lineatum: impact of immunization on larval survival in artificial infestations.

Soluble fractions of Hypoderma lineatum third instar fat body, haemocytes and haemolymph were formulated with Quil A and used to immunize four groups of calves while a fifth group remained untreated. Calves received two subcutaneous injections of the soluble fractions, or adjuvant only delivered two weeks apart. Two weeks after the last injection the calves were exposed to 50 newly hatched larvae of H. lineatum which were placed on the skin and allowed to penetrate. Survival of larval stages was monitored by weekly palpation and collection of emergent third instars. Antibody responses to the i

Biomphalaria glabrata transcriptome: cDNA microarray profiling identifies resistant- and susceptible-specific gene expression in haemocytes from snail strains exposed to Schistosoma mansoni.

BACKGROUND: Biomphalaria glabrata is an intermediate snail host for Schistosoma mansoni, one of the important schistosomes infecting man. B. glabrata/S. mansoni provides a useful model system for investigating the intimate interactions between host and parasite. Examining differential gene expression between S. mansoni-exposed schistosome-resistant and susceptible snail lines will identify genes and pathways that may be involved in snail defences. RESULTS: We have developed a 2053 element cDNA microarray for B. glabrata containing clones from ORESTES (Open Reading frame ESTs) libraries, suppr

Lyme borreliosis: insights into tick-/host-borrelia relations.

Lyme borreliosis (LB) is a serious infectious disease of humans and some domestic animals in temperate regions of the Northern Hemisphere. It is caused by certain spirochetes in the Borrelia burgdorferi sensu lato (s.l.) species complex. The complex consists of 11 species (genospecies). Borrelia burgdorferi sensu stricto (s.s.), Borrelia garinii and Borrelia afzelii are the major agents of human disease. Borrelia burgdorferi s.l. species are transmitted mainly by ticks belonging to the Ixodes ricinus species complex plus a few additional species not currently assigned to the complex. B. burgdo

Mechanisms of molluscan host resistance and of parasite strategies for survival.

In parallel with massive research efforts in human schistosomiasis over the past 30 years, persistent efforts have been made to understand the basis for compatibility and incompatibility in molluscan schistosomiasis. Snail plasma contains molecules that are toxic to trematodes, but these seem to kill only species that never parasitize the mollusc used as the source of plasma. A sporocyst will be killed actively by haemocytes alone if they are from a snail that is resistant to the trematode. Oxygen-dependent killing mechanisms play a major role. Enzymes such as NADPH oxidase, superoxide dismuta

Specific protein phosphorylation occurs in molluscan red blood cell ghosts in response to hypoosmotic stress.

The regulation of cellular volume upon exposure to hypoosmotic stress is accomplished by specific plasma membrane permeability changes that allow the efflux of certain intracellular solutes (osmolytes). The mechanism of this membrane permeability regulation is not understood; however, previous data implicate Ca2+ as an important component in the response. The regulation of protein phosphorylation is a pervasive aspect of cellular physiology that is often Ca2+ dependent. Therefore, we tested for osmotically induced protein phosphorylation as a possible mechanism by which Ca2+ may mediate osmoti

Drosophila heat shock factor (HSF) regulates developmental growth by maintaining the basal expression of HSP83/HSP90.

The heat shock transcription factor HSF1 is best known as a master regulator of the proteotoxic stress response, yet its functions remain incompletely understood. In Drosophila melanogaster, heat shock factor (HSF) is essential for viability, but the mechanisms by which it promotes development are unclear. Here, we show that Hsf null larvae arrest at the early second instar stage and exhibit a significant reduction in basal HSP83/HSP90 levels. Tissue-specific knockdown of Hsf revealed widespread, organ-specific requirements, including defects in endoreplication and cell growth in prothoracic a
